Abstract— Pretreatment with sulfhydryl-reactive agents, such as N-ethylmaleimide and p-chloromercuriphenylsul-fonic acid, invariably resulted in marked inhibition of the binding of dl -(E)-2-amino-4-[3H]propyl-5-phosphono-3-pentenoic acid ([3H]CGP 39653), a competitive antagonist at an N-methyl-d -aspartate (NMDA)-sensitive subclass of central excitatory amino acid receptors, in brain synaptic membranes extensively washed and treated with Triton X-100, but did not significantly affect the binding of L-[3H]-glutamic acid ([3H]Glu), an endogenous agonist. The pre-treatment was effective in reducing the binding of [3H]-CGP 39653 at equilibrium, without altering the initial association rate, and decreased the affinity for the ligand. Pretreatment with sulfhydryl-reactive agents also enhanced the potencies of NMDA agonists to displace [3H]-CGP 39653 binding and attenuated those of NMDA antagonists, but had little effect on the potencies of the agonists and antagonists to displace [3H]Glu binding. The binding of both [3H]CGP 39653 and [3H]Glu was similarly sensitive to pretreatment with four different proteases in Tritontreated membranes, whereas pretreatment with phospho-lipase A2 or C markedly inhibited [3H]CGP 39653 binding without altering [3H]Glu binding. Moreover, both phospho-lipases not only induced enhancement of the abilities of NMDA agonists to displace the binding of [3H]CGP 39653 and [3H]Glu, but also caused diminution of those of NMDA antagonists. These results suggest that both sulfhydryl-reactive agents and phospholipases may predominantly interfere with radiolabeling of the NMDA recognition domain in a state favorable to an antagonist by [3H]CGP 39653, with concomitant facilitation of that in an agonist-preferring form by [3H]Glu. The possible presence of multiple forms of the NMDA recognition domain is further supported by these data.  相似文献

为保护广西北部湾海岸带、海岛的植物多样性和生态系统多样性,通过样方、样带法野外实地调查并结合文献资料,对其外来入侵植物的物种组成、原产地、生活型、入侵途径和危害状况等进行了分析。结果表明,广西北部湾海岸带、海岛共有入侵植物64种,隶属28科55属,其中菊科(Asteraceae)最多(15种)。草本植物最多,有48种(75.00%)。原产地来自美洲的植物最多,有49种。入侵风险等级可划分为5个等级,其中Ⅰ级严重危害的有8种(12.50%)。与广西、广东、海南及华南地区的外来入侵植物在物种组成、生活型和原产地等方面呈现出较强的相似性;北部湾与广西中越边境内陆地区来自美洲的入侵植物都超过60%。因此推测广西外来入侵植物有两条可能的入侵线路:一是从海南登录,二是从中越边境跨入。广西北部湾海岸带、海岛的外来入侵植物总数(相对整个广西)虽较少,但其8种Ⅰ级严重危害植物的防治,仍需引起重视。  相似文献

In the process of bioethanol production, more stable and active cellulase in high temperature condition is required. In this study, syringic acid was applied in cellulase hydrolysis system. At 70°C, TvEG3 activity increased 201.36%, CtBglA activity decreased 72.79% by syringic acid. With syringic acid assisting, TvEG3 thermostability was improved, CtBglA thermostability was reduced. Syringic acid scarcely affected CtCBH. In hydrolysis system with the cellulases containing TvEG3, CtCBH, and CtBglA, the reducing sugar yield improved by 28.37% with syringic acid assisting. With the molecular dynamic simulation in syringic acid system, the backbone root-mean-square deviation (RMSD) and the residue root-mean-square fluctuation (RMSF) of TvEG3, CtCBH reduced, while the RMSD and RMSF of CtBglA increased. The reduction in the number of secondary structures, especially α-helix, caused the structure of CtBglA in the presence of syringic acid to collapse at high temperature. More secondary structures in TvEG3 and more α-helix in CtCBH in the presence of syringic acid make them more stable at high temperatures. These means syringic acid can stabilize TvEG3 and CtCBH structure, destabilize CtBglA structure at high temperature. In summary, this study not only provides insight into cellulase hydrolysis at high temperature with syringic acid assisting but also demonstrates the promoting mechanism of syringic acid.  相似文献

本文总结了应用相似系数即关联系数进行植物区系相似性分析的现状,指出了存在的问题.然后,从集合论角度讨论了区系相似性、相似关系及其相似系数的实质.作者以为在区系相似性分析中应用R.R.Sokal和C.D.Michener(1958)提出的简单匹配系数比较适宜,同时亦能避免以往区系相似性分析中缺乏可比性及某些“表相”相似等问题.最后,还提出了总体相似系数和类型相似系数二个新概念,以便按照吴征镒教授关于中国植物区系研究的学术思想统一研究各个不同地区植物区系的相似性.对此,作者用了6个区系实例进行了演算说明.  相似文献

利用EB病毒转化可产生较高水平人IgG和特异性抗2型登革病毒人抗体的Hu-TLC-SCID小鼠脾细胞,通过免疫组化、免疫荧光和PCR法检测转化细胞的人B细胞表面标志、EB病毒抗原和EB病毒基因。结果表明,被转化的Hu-TLC-SCID小鼠脾细胞能继续产生抗2型登革病毒的特异性人抗体,并具有人B细胞的、SmIgG标志及EB病毒潜伏膜蛋白-1(LMP-1)基因,可表达LMP-1和EB病毒核抗原(EBNA)。  相似文献

对原生质体电融合技术获得的11株酵母多倍体融合株,进行了一系列生理特性的分析比较.结果发现,在电融合过程中,融合株的细胞体积及DNA含量并不随着核倍性呈线性递增.而是有其特殊的变化规律。当糖化酵母sta1、sta2和sta3在细胞核中各自纯合时,明显表现出表达的剂量效应。其中sta1、sta2纯合的融合株.在YEPS培养中,GA分泌可被淀粉大量诱导,表现出一定的二次生长特性。从分析结果推测,在亲株5301-14D及HU-TY—1A中可能有对GA分泌不利的因素。  相似文献

Exposure of pyrimidines of DNA to ionizing radiation under aerobic conditions or oxidizing agents results in attack on the 5,6 double bond of the pyrimidine ring or on the exocyclic 5-methyl group. The primary product of oxidation of the 5,6 double bond of thymine is thymine glycol, while oxidation of the 5-methyl group yields 5-hydroxymethyluracil. Oxidation of the 5,6 double bond of cytosine yields cytosine glycol, which decomposes to 5-hydroxycytosine, 5-hydroxyuracil and uracil glycol, all of which are repaired in DNA by Escherichia coli endonuclease III. We now describe the products of oxidation of 5-methylcytosine in DNA. Poly(dG-[3H]dmC) was gamma-irradiated or oxidized with hydrogen peroxide in the presence of Fe3+ and ascorbic acid. The oxidized co-polymer was incubated with endonuclease III or 5-hydroxymethyluracil-DNA glycosylase, to determine whether repairable products were formed, or digested to 2'-deoxyribonucleosides, to determine the total complement of oxidative products. Oxidative attack on 5-methylcytosine resulted primarily in formation of thymine glycol. The radiogenic yield of thymine glycol in poly(dG-dmC) was the same as that in poly(dA-dT), demonstrating that 5-methylcytosine residues in DNA were equally susceptible to radiation-induced oxidation as were thymine residues.  相似文献
