虎杖象甲培植共生真菌形成的共生体系是植菌昆虫菌业中的典型代表。共生真菌Penicillium herquei如何向虎杖象甲Euops chinensis提供营养尚未明确。本研究发现共生真菌P. herquei的菌丝表面存在大量瘤状凸起物及由凸起物衍生的附属丝等特化结构,该结构可能为虎杖象甲提供营养;对共生真菌的营养研究表明,共生真菌能高效利用山梨醇、蔗糖、海藻糖、葡萄糖等单糖或双糖,以及酪氨酸、甘氨酸、谷氨酰胺等昆虫非必须氨基酸,同时在高碳和最适碳源条件下有利于菌丝特化附属物的产生。研究结果不仅提供了植菌卷叶象甲菌业中共生真菌在营养方面的适应性进化证据,而且为进一步揭示共生真菌适应卷叶象甲的营养机制奠定了基础。  相似文献

Strobilomyces echinocephalus Gelardi & Vizzini sp. nov. is described as a new species, based on collections from Kunming (Yunnan Province, China). Macro-morphological and micro-morphological features, ecological data and analyses of RPB1 and ITS2 sequences support the establishment of this new taxon. Colour pictures of fresh basidiomata in habitat, scanning electron microscope (SEM) micro-photographs of the spores, and a line-drawing displaying the most relevant anatomical characters are provided in this paper. Strobilomyces zangii Gelardi nom. nov. is proposed to replace the name Heimiella nigricans M. Zang. A dichotomous key to the Strobilomyces species at global level is also provided at the end of this work.  相似文献

昆虫菌业(fungiculture)是一种类似于人类种植业的昆虫种植体系,包括种植、耕作、收获和营养依赖4个过程,可分为高级的社会性昆虫如切叶蚂蚁、白蚁等和低级的非社会性昆虫如食菌小蠹虫、卷叶象甲、蜥蜴甲虫、树蜂等,它们均能种植并取食真菌。近年来随着组学及微生物组技术的发展,植菌昆虫与其共生真菌协同进化的分子机制研究方面取得了重要进展。系统发育分析阐明了植菌昆虫的起源与进化历程,并显示出与共生真菌系统发育的一致性;共生真菌细胞核数量也从双核增加到最多17个核,而染色体倍型也从单倍体增加为二倍体甚至多倍体;组学分析则揭示了植菌昆虫与其共生真菌在精氨酸、碳水化合物、木质素及几丁质合成或降解等方面显示出了高度的协同进化。本文系统综述了植菌昆虫及其共生真菌的系统进化、核进化及基因组进化进展,并探讨这种协同进化机制的生物学意义。  相似文献

Filamentous fungi are excellent sources for the production of a group of bioactive small molecules which are often called secondary metabolites(SMs). The advanced genome sequencing technology combined with bioinformatics analysis reveals a large number of unexplored biosynthetic gene clusters(BGCs) in the fungal genomes. To unlock this fungal SM treasure, many approaches including heterologous expression are being developed and efficient cloning of the BGCs is a crucial step to do this.Here, we present an efficient strategy for the direct cloning of fungal BGCs. This strategy consisted of Splicing by Overlapping Extension(SOE)-PCR and yeast assembly in vivo. By testing 14 BGCs DNA fragments ranging from 7 kb to 52 kb, the average positive rate was over 80%. The maximal insertion size for fungal BGC assembly was 52 kb. Those constructs could be used conveniently for the heterologous expression leading to the discovery of novel natural products. Thus, our results provide an efficient and quick method for the low cost direct cloning of fungal BGCs.  相似文献

Lignin degradation by white-rot fungi has received considerable attention as a means for reducing accumulation of lignocellulosic wastes in the environment. The stimulatory effect of surfactants on fungal lignocellulose bioconversion also has attracted wide interest. In this study the influence of dirhamnolipid biosurfactant on biodegradation of rice straw by Phanerochaete chrysosporium was investigated. It was shown that the biodelignification process of rice straw can be significantly enhanced by the presence of dirhamnolipid biosurfactant. In particular, the dirhamnolipid at the concentration of 0.007% increased the peak activity of lignin peroxidase (LiP) by 86% without affecting the manganese peroxidase (MnP) activity. The water-soluble organic carbon (WSOC) contents in the straw substrates as well as the microbial growth and activity were effectively improved by dirhamnolipid, while the degradation rate of lignin increased by 54% with dirhamnolipid of 0.007%. Observed chemical structural and morphological changes showed that the straw substrates were delignified in the presence of dirhamnolipid with the formation of terrace-like fragments separated from the inner cellular fibers and the release of simple compounds. Variation partitioning analysis revealed that the dirhamnolipid addition induced a significant straw biodelignification which explained 22.1% (P = 0.013) of the variance.  相似文献

To supply essential information for improving mass production and biocontrol efficacy, two-stage cultivation on agar plates was used to evaluate the environmental conditions affecting mycelial growth and sporulation of seven biocontrol fungi. Maximum growth and sporulation occurred on acid media for Paecilomyces (Pa.) lilacinus IPC-P, Pochonia (Po.) chlamydosporia HSY-12-14, and Lecanicillium lecanii CA-1-G, and on alkaline media for Metarhizium anisopliae isolates. All fungi preferred a certain water potential and temperature for sporulation. Light greatly inhibited the growth of P. lilacinus IPC-P, M. anisopliae SQZ-1-21, and L. lecanii CA-1-G but enhanced the sporulation of P. lilacinus M-14, P. chlamydosporia HSY-12-14, and L. lecanii CA-1-G.  相似文献

对来自不同地方的7个弯孢节丛孢(ArthrobotrysmusiformisDrechsler)和9个厚皮单顶孢(Monacrosporiumeudermatum(Drechsler)Subram)标准菌株用4个随机引物(OPW-18、OPW-16、OPM-1、OPK-19)进行了随机扩增多态性DNA(RAPD)分析。扩增谱带聚类分析结果不能将弯孢节丛孢和厚皮单顶孢的菌株分开,具有附属丝的菌株没有被聚在一起,菌株间地理起源上的亲缘关系也没有体现,证明RAPD分析不能作为节丛孢属和单顶孢属的属间鉴别分析,附属丝地捕食线虫真菌不具分类学意义,不能以之作为成立小掘氏霉属的证据,系统聚类关系与地理来源基质也没有相关性。  相似文献

真菌是生物多样性最丰富和生存环境最多样的生物类群之一,除了我们熟知的土壤、植物残体、水体等一般环境外,还存在极地、高温、高盐等各种特殊环境.对真菌而言,特殊环境是指绝大多数真菌不能生存的环境,而特殊环境真菌是指特殊环境中特有的真菌或适应特殊环境的真菌.特殊环境人为地划分为以下3种类型:第一类,具有某种特定的理化限制因素的环境;第二类,某些特殊基质;第三类,受多因素限制的特殊复杂环境.特殊环境真菌有着独特的适应性,并进化出各种机制来占据生态位.它们在细胞构造、代谢方式、进化机制等方面的特殊性引起了科学家的广泛关注.本文介绍了各种特殊生态环境下生活的真菌类群以及它们的生存机制,并对今后的研究方向进行了展望.  相似文献

The effects of foliar spraying with spermidine (Spd), ranging in concentration from 0.25 to 0.50mmol/L, on the antioxidant system under Cd2 stress (range 0.1- 0.2 mmol/L Cd2 ) in Typha latifolia L.grown hydroponically were investigated in order to offer a referenced evidence for an understanding of the mechanism by which polyamines (PAs) relieve the damage to plants by heavy metal and improve the phytoremediation efficiency of heavy metal-contaminated water. The results showed that Cd2 stress inhydrogen peroxide (H2O2) and malondialdehyde (MDA) contents in both leaves and caudices. With theexception of superoxide dismutase (SOD) activity in the leaves, an increase in the activities of catalase (CAT), guaiacol peroxidase (GPX), and glutathione reductase (GR) was observed in both leaves and caudices,SOD activity was increased in caudices, and ascorbate peroxidase (APX) activity was increased in leaves following Cd2 treatment. The reduced glutathione (GSH) content in both leaves and caudices and the reductive ascorbate content in leaves was obviously increased, which were prompted by the application of exogenous Spd. Spraying with Spd increased the activity of GR and APX in both leaves and caudices,whereas the activity of SOD, CAT, and GPX was increased only in caudices following spraying with Spd.with Spd. The decrease in MDA was more obvious following the application of 0.25 than 0.50 mmol/L Spd.It is supposed that exogenous Spd elevated the tolerance of T. latifolia under Cd2 stress primarily by increasing GR activity and the GSH level.  相似文献
