Biomass production and carbohydrate reduction were determined for a two-stage continuous fermentation process with a simulated potato processing waste feed. The amylolytic yeast Saccharomycopsis fibuligera was grown in the first stage and a mixed culture of S. fibuligera and Candida utilis was maintained in the second stage. All conditions for the first and second stages were fixed except the flow of medium to the second stage was varied. Maximum biomass production occurred at a second stage dilution rate, D(2), of 0.27 h (-1). Carbohydrate reduction was inversely proportional to D(2), between 0.10 and 0.35 h (-1).  相似文献

Bilayer tableting technology has gained popularity in recent times, as bilayer tablets offer several advantages over conventional tablets. There is a dearth of knowledge on the impact of material properties and process conditions on the performance of bilayer tablets. This paper takes a statistical approach to develop a model that will determine the effect of the material properties and bilayer compression process parameters on the bonding strength and mode of breakage of bilayer tablets. Experiments were carried out at pilot scale to simulate the commercial manufacturing conditions. As part of this endeavor, a seven-factor half-fraction factorial (27−1) design was executed to study the effect of bilayer tablet compression process factors on the bonding strength of bilayer tablets. Factors studied in this work include: material properties (plastic and brittle), layer ratio, dwell time, layer sequence, first- and second-layer forces, and lubricant concentration. Bilayer tablets manufactured in this study were tested using the axial tester, as it considers both the interfacial and individual layer bonding strengths. Responses of the experiments were analyzed using PROC GLM of SAS (SAS Institute Inc, Cary, North Carolina). A model was fit using all the responses to determine the significant interactions (p < 0.05). The results of this study indicated that nature of materials played a critical role on the strength of bilayer compacts and also on mode of fracture. Bilayer tablets made with brittle materials in both the layers are strongest, and fracture occurred in the first layer indicating that interface is stronger than layers. Significant interactions were observed between the selected factors and these results will provide an insight into the interplay of material properties, process parameters, and lubricant concentration on the bonding strength and mode of breakage of bilayer tablets.

Puccinia graminis f. sp. tritici, the causative agent of stem rust in wheat, is known for its high virulence variability and ability to evolve new virulence to resistance genes. Thus, pyramiding of several resistance genes in a single line is the best strategy for a sustainable control of wheat stem rust. Sr13 is one of the few resistance genes that are effective against wide ranging P. graminis f. sp. tritici races, including the pestilent race Ug99. Its effectiveness to Ug99 makes it a valuable source for resistance to stem rust. Molecular markers play a pivotal role in the genetic characterization of the new sources of resistance as well as in stacking two or more resistance genes in a single line. Therefore, the aim of this study was to develop molecular markers for Sr13 facilitating efficient pyramiding of Sr genes. Based on the 158 F2 individuals derived from a cross of Khapstein/9*LMPG × Morocco and SSR analyses, the Sr13 locus was mapped on chromosome 6A of wheat, and a genetic map comprising about 90 cM was constructed with the closest marker barc37 being located 4.0 cM distally of Sr13. Of the nine mapped markers, barc37 amplified an allele specific for the presence of Sr13 as shown by testing different cultivars and breeding lines. These newly developed markers will increase the efficiency of incorporating Sr13 into cultivars that are widely adopted, but susceptible to hazardous Ug99 and/or assist for the development of new elite lines that are resistant to Ug99.  相似文献

Age of mature Oreochromis niloticus in Lake Awassa, Ethiopia, was estimated by analysing optical macrozones (translucent and opaque) in sagittal otoliths from fish sampled over a 12-month period. Seasonal record on the type of macrozone at the edge of otoliths suggested that two translucent macrozones associated with biannuli were formed each year; one during January and February and another during June and July. Formation of translucent macrozones coincided with minimum water temperature, spawning associated loss in condition and presumably with reduction in the quantity and quality of the food consumed by the fish. Relative marginal increment analysis showed that biannulus formation may be completed in March and in August each year. A concurrent study has confirmed a biannual recruitment in O. niloticus in Lake Awassa, from which mid-February and mid-August were taken as median hatch-dates. A procedure to assign otolith age is described which considers median hatch-dates and uses the number of biannuli in otoliths without discriminating between fish from the two recruitment cohorts. We also show a procedure to discriminate between the two cohorts by using the number of biannuli, conditions on the edge of the otolith and time of capture to assign age for the two recruitment cohorts separately.  相似文献

Twenty three polymorphic microsatellite markers were developed for citrus plant pathogenic fungus, Colletotrichum gloeosporioides, and were used to analyze genetic diversity and population structure of 163 isolates from four different geographical regions of Ethiopia. These loci produced a total of 118 alleles with an average of 5.13 alleles per microsatellite marker. The polymorphic information content values ranged from 0.104 to 0.597 with an average of 0.371. The average observed heterozygosity across all loci varied from 0.046 to 0.058. The gene diversity among the loci ranged from 0.106 to 0.664. Unweighted Neighbor-joining and population structure analysis grouped these 163 isolates into three major groups. The clusters were not according to the geographic origin of the isolates. Analysis of molecular variance showed 85% of the total variation within populations and only 5% among populations. There was low genetic differentiation in the total populations (FST = 0.049) as evidenced by high level of gene flow estimate (Nm = 4.8 per generation) among populations. The results show that Ethiopian C. gloeosporioides populations are generally characterized by a low level of genetic diversity. The newly developed microsatellite markers were useful in analyzing the genetic diversity and population structure of the C. gloeosporioides populations. Information obtained from this study could be useful as a base to design strategies for better management of leaf and fruit spot disease of citrus in Ethiopia.  相似文献

A strain of Clostridium bifermentans isolated from a munitions-supplemented anaerobic digester is known to degrade 2,4,6-trinitrotoluene (TNT) in rich media such as Brain Heart Infusion (BHI) broth. In order to make this biodegradation process commercially feasible, a new growth medium was developed. Corn steep liquor and molasses were selected as possible nitrogen and carbon sources. A medium containing 2.4% corn steep liquor and 0.4% molasses was chosen based on the value for maximum specific growth rate. The values of µm and Ks from continuous runs were of 0.029 min-1 and 0.488 g/L, respectively. To reduce the overhead cost for maintaining an anoxic environment for this obligatory anaerobic bacterium, the threshold oxygen level under which the bacterium can survive was determined. The degradation of TNT was then carried out in a batch bioreactor, and in a continuously stirred tank bioreactor. The solubility of TNT was enhanced by using the surfactant, Tween 80, and the optimal concentration of Tween 80 was determined to be 2.5%. This concentration of Tween 80 increased TNT solubility by 100% while the effect on growth rate was minimal. Degradation of TNT was then carried out in a continuously stirred tank bioreactor and was found to follow Michaelis-Menten-type kinetics. Degradation of TNT was found to be possible under limited oxygen presence, but at a reduced degradation rate.  相似文献

The emergence and rapid spread of virulent races of wheat stem rust has driven a search for sources of resistance for durable resistance breeding. This study was carried out to identify possible sources of stem rust resistance between Ethiopian wheat lines. Two hundred and fifty‐two wheat accessions and a universal suscept, cultivar Morocco were evaluated for their resistance at the seedling stage to the stem rust isolate Ug99 in a controlled environment. Ninety‐one lines that exhibited intermediate and susceptible seedling reactions were further field tested in 2012 main season for their slow rusting characteristics. Among the 91, 38 genotypes that had high to moderate level of slow rusting were advanced to a 2013 off season field evaluation. Slow rusting resistance at the adult‐plant stage was assessed through the determination of final disease severity (FRS), coefficient of infection (CI), and relative area under disease progressive curve (rAUDPC). The results revealed that wheat lines H04‐2, 204408‐3, 214551‐1, 231545‐1, 7041‐1, 7514‐1, 226385‐1, 226815‐1, 7579‐1 and 222495‐1 had low values of FRS, CI and rAUDPC and were regarded as good slow rusting lines. Of these 231545‐1, 7041‐1, 226815‐1 and 7579‐1 exhibited complete susceptibility at the seedling stage, with infection types ranging from 3? to 3+, which suggests that they possess true slow rusting resistance. Lines 237886‐1, 227059‐1, 203763‐1, 226275‐1, 227068‐2, 226278‐1 and 7994‐1 had moderate values for the stem rust resistance parameters and were identified as possessing a moderate level of slow rusting. High correlations were observed between different parameters of slow rusting. Among the slow rusting lines 231545‐1, H04‐2 and 222495‐1 had high yields and kernel weight in both seasons. The slow rusting lines identified from this study can be used to breed for stem rust resistance in wheat.  相似文献
