We investigated the spatial expression of three genes that are expressed during seed germination and postgerminative development in Brassica napus L. using in situ hybridization procedures. Two of the mRNAs encode isocitrate lyase and a predicted polypeptide that is homologous to cysteine proteinases. We reported previously that the mRNAs are prevalent primarily in cotyledons of seedlings and accumulate with similar kinetics during postgerminative growth. Here, we show that the two mRNAs are detected in several seedling tissues, but they display different distribution patterns in both cotyledons and root-shoot axes. The third mRNA is abundant in seedling axes and accumulates specifically in the ground meristem and mature cortex of hypocotyls and roots. Distribution of the mRNA in root meristems suggests that the gene product participates in an early event in cortical cell differentiation. Our results provide insight into the physiological processes that characterize seedlings. 相似文献

We have purified and isolated cDNAs encoding the beta subunit of tomato fruit polygalacturonase isoenzyme 1 (PG1), a cell wall protein that associates with, and apparently regulates, the catalytic PG2 polypeptides. Expression of the beta subunit is fruit specific and temporally separated from the expression of PG2 during fruit development. The 37- to 39-kD beta subunit is encoded as a 69-kD precursor protein containing a signal sequence and two propeptide domains. The mature protein is composed almost entirely of the novel 14-amino acid motif FTNYGxxGNGGxxx in which many of the phenylalanine residues are post-translationally modified. The unique structural features of the motif suggest an important role in the function of the protein and hence in the activity of PG1. The beta subunit may represent a class of bifunctional plant proteins that interact both with structural components of the cell wall and catalytic proteins to localize and/or regulate metabolic activities within the cell wall. 相似文献

Baseline, post‐angling and maximum attainable blood lactate concentrations were measured for the fishery species redthroat emperor Lethrinus miniatus to gain insight into the condition of fish released following c. 30 s angling and <45 s air exposure. Mean ± s.d . baseline blood lactate was 1·5 ± 0·6 mmol l?1, which increased and plateaued around 6 mmol l?1 at 15–30 min post‐angling. These values were significantly lower than those obtained from fish maximally exhausted with a prolonged chase and air exposure protocol following capture (10·9 ± 1·8 mmol l?1), suggesting that L. miniatus is not maximally exhausted during standard angling practices. 相似文献

Many parasites with complex life cycles increase the chances of reaching a final host by adapting strategies to manipulate their intermediate host's appearance, condition or behaviour. The acanthocephalan parasite Pomphorhynchus laevis uses freshwater amphipods as intermediate hosts before reaching sexual maturity in predatory fish. We performed a series of choice experiments with infected and uninfected Gammarus pulex in order to distinguish between the effects of visual and olfactory predator cues on parasite-induced changes in host behaviour. When both visual and olfactory cues, as well as only olfactory cues were offered, infected and uninfected G. pulex showed significantly different preferences for the predator or the non-predator side. Uninfected individuals significantly avoided predator odours while infected individuals significantly preferred the side with predator odours. When only visual contact with a predator was allowed, infected and uninfected gammarids behaved similarly and had no significant preference. Thus, we believe we show for the first time that P. laevis increases its chance to reach a final host by olfactory-triggered manipulation of the anti-predator behaviour of its intermediate host. 相似文献

The use of a coastal estuary by bonnethead sharks, Sphyrna tiburo, was examined by acoustic monitoring, gillnet sampling and tag- recapture studies. Acoustic monitoring data were used to define the residency and movement patterns of sharks within Pine Island Sound, Charlotte Harbor, Florida. Sharks were monitored for periods of 1–173 days with individuals regularly moving in and out of the detection range of the acoustic system. Patterns of movement could not be correlated with tidal level or time of day. Home range sizes within the Pine Island Sound population were typically small with individuals using core areas on a daily basis. However, core areas shifted within the study site over time resulting in eventual usage of most of the available habitat. Gillnet sampling revealed that S. tiburo were abundant in shallow water near seagrass beds, but that presence of individuals at specific sites was variable. Tag-recapture data showed that most individuals remained within the Pine Island Sound region over time and did not appear to undergo long coastal migrations. The movement and residence patterns of S. tiburo suggest that individuals are resident within the estuary, but do not show site fidelity to specific areas within the estuary. 相似文献

The increase in reports of novel diseases in a wide range of ecosystems, both terrestrial and marine, has been linked to many factors including exposure to novel pathogens and changes in the global climate. Prevalence of skin cancer in particular has been found to be increasing in humans, but has not been reported in wild fish before. Here we report extensive melanosis and melanoma (skin cancer) in wild populations of an iconic, commercially-important marine fish, the coral trout Plectropomus leopardus. The syndrome reported here has strong similarities to previous studies associated with UV induced melanomas in the well-established laboratory fish model Xiphophorus. Relatively high prevalence rates of this syndrome (15%) were recorded at two offshore sites in the Great Barrier Reef Marine Park (GBRMP). In the absence of microbial pathogens and given the strong similarities to the UV-induced melanomas, we conclude that the likely cause was environmental exposure to UV radiation. Further studies are needed to establish the large scale distribution of the syndrome and confirm that the lesions reported here are the same as the melanoma in Xiphophorus, by assessing mutation of the EGFR gene, Xmrk. Furthermore, research on the potential links of this syndrome to increases in UV radiation from stratospheric ozone depletion needs to be completed. 相似文献

The mechanism of the decarboxylation of α-keto acids by isolated chloroplasts has been studied with the aid of superoxide dismutase and catalase. Using photosynthetic and enzymatic systems, which are known to catalyze peroxidic oxidations, we have been able to demonstrate that both the superoxide free radical ion and H2O2 are necessary for maximal rates of decarboxylation. In isolated chloroplasts, an auto-oxidizable electron acceptor as well as an electron donor for Photosystem I are absolute requirements for the decarboxylation. H2O2 seems to be the primary oxidant in the decarboxylation of pyruvate or glyoxylate by isolated chloroplasts. A secondary rate of decarboxylation is superimposed on the primary one, mediated by superoxide free radical ion. Mn2+ stimulates the decarboxylation probably via intermediarily-formed Mn3+ in a reaction, which is neither inhibited by catalase nor by superoxide dismutase. A decarboxylation of pyruvate or glyoxylate by isolated chloroplasts in the presence of NADP+ is initiated, as soon as the available NADP+ is fully reduced. In this case, the open-chain electron transport seems to switch from NADP+ to oxygen as the terminal electron acceptor. 相似文献

Acoustic telemetry was used to examine the size of daily activity space, small-scale movement patterns, and water quality
preferences of juvenile bull sharks in the Caloosahatchee River, Florida. Movement pattern analysis included rate of movement,
swimming depth, linearity, direction, tidal influence, diel pattern, and correlation with environmental variables. Manual
tacking occurred before and after a large freshwater influx which divided the sharks into two groups based on movement patterns.
The first group displayed increased rate of movement, distance traveled, and space utilization at night, and movements correlated
with salinity, temperature, and dissolved oxygen. The second group had an increased rate of movement, distance traveled, and
space utilization during the day, and movements correlated with temperature, dissolved oxygen, turbidity and pH. These juvenile
bull sharks displayed distinct diel movement patterns that were influenced by physical factors, which may account for the
distribution of this top-level predator in the Caloosahatchee River. 相似文献

Passive acoustic telemetry provides an important tool to study the spatial ecology and behaviour of organisms in marine and freshwater systems, but understanding the detection range of acoustic receivers is critical for interpreting acoustic data and establishing receiver spacing to maximize study efficiency. This study presents a comprehensive review of how acoustic detection range has been considered and assessed to date, summarizes important variables to monitor when determining the detection range of a receiver array, and provides recommendations to account for detection range during experimental design, analysis and data interpretation. A total of 378 passive acoustic telemetry studies (1986–2012) were scored against a set of pre-defined criteria to provide a standardized assessment of how well detection range was accounted for, from a maximum possible score of 45. Scores ranged from 0 to 39 (11.1 ± 0.4; mean ± 1 SE). Over the past decade mean scores have been consistently between 6.7 and 12.9 which indicates that detection range has not been adequately considered in most contemporary acoustic telemetry studies. Given the highly variable nature of detection range over space and time, it is necessary to create a culture of detection range testing among the scientific community. For robust telemetry studies it is recommended that consideration of detection range should be given a greater focus within study design, execution and data analysis. To aid array design in new systems, short-term detection range tests should be conducted in the most representative area of the study system prior to deployment. As well, fixed distance sentinel tags should ideally be deployed at a representative receiver site within the array to provide a continuous assessment of detection range and influential environmental parameters should be monitored to facilitate modeling of detection range variability over time. When warranted, data analysis should incorporate modeled variation in detection ranges. 相似文献
