A method is described for the histochemical detection of horseradish peroxidase in Paraplast Plus embedded brain sections. The procedure uses 150-micron-thick Vibratome-cut slices of glutaraldehyde-paraformaldehyde-fixed brain tissue. Tetramethylbenzidine stabilized by diaminobenzidine/cobalt/H2O2 is used as chromogen. The Vibratome-cut slices are dehydrated through a graded series of acetone, cleared in toluol and flat-embedded in Paraplast Plus embedding medium. Serial sections can be cut as thin as 5-7 micron. The method is universal in its application and permits optimal visualization of labeled neurons with great morphological detail at the light-microscopic level.  相似文献

In toto staining and preservation of peripheral nervous tissue   总被引:2,自引:0,他引:2  
A simple quantitative modification of the in toto staining technique for nervous networks of Sihler is described. The results are demonstrated on the innervation pattern of the hard palate of the rat. Formalin fixed hard palates of rat were macerated and bleached in an aqueous solution of 3% potassium hydroxide with a few drops of 3% hydrogen peroxide added. Thereafter, the specimens were decalcified in Sihler's solution I (1 part glacial acetic acid, 1 part pure glycerin and 6 parts 1% chloral hydrate), the process being controlled by radiography. The specimens were next stained in Sihler's solution II (1 part Ehrlich's hematoxylin, 1 part pure glycerin and 6 parts 1% chloral hydrate). After staining, the non-nervous tissues were destained in Sihler's solution I. Destaining was checked microscopically and was stopped before the finest branches of the nerves began to fade. The specimens were then washed in a weak aqueous solution of lithium carbonate and cleared in increasing concentrations of glycerin. Good visualization of nervous structures and a deep field of observation resulted; orientation of the peripheral nerves with respect to surrounding structures was readily seen and a three-dimensional image of the nervous networks was obtained.  相似文献

Efferent and afferent connections of the supratrigeminal region were studied in the rat using iontophoretically delivered horseradish peroxidase and Phaseolus vulgaris leuco-agglutinin. Projections of supratrigeminal efferents were found to the contralateral supratrigeminal region, to the ipsi- and contralateral trigeminal motor nuclei and the medullary reticular formation, and to the ipsilateral facial and hypoglossal motor nuclei. Neurons projecting to the supratrigeminal region were located in the contralateral supratrigeminal nucleus, in the ipsilateral mesencephalic trigeminal nucleus and bilaterally in the medullary reticular formation. This organization is discussed with respect to bilateral oral motor control mechanisms.  相似文献

During dehydration, numerous metabolites accumulate in vegetative desiccation-tolerant tissues. This is thought to be important in mechanically stabilizing the cells and membranes in the desiccated state. Non-aqueous fractionation of desiccated leaf tissues of the resurrection grass Eragrostis nindensis (Ficalho and Hiern) provided an insight into the subcellular localization of the metabolites (because of the assumptions necessary in the calculations the data must be treated with some caution). During dehydration of the desiccant-tolerant leaves, abundant small vacuoles are formed in the bundle sheath cells, while cell wall folding occurs in the thin-walled mesophyll and epidermal cells, leading to a considerable reduction in the cross-sectional area of these cells. During dehydration, proline, protein, and sucrose accumulate in similar proportions in the small vacuoles in the bundle sheath cells. In the mesophyll cells high amounts of sucrose accumulate in the cytoplasm, with proline and proteins being present in both the cytoplasm and the large central vacuole. In addition to the replacement of water by compatible solutes, high permeability of membranes to water may be critical to reduce the mechanical strain associated with the influx of water on rehydration. The immunolocalization of a possible TIP 3;1 to the small vacuoles in the bundle sheath cells may be important in both increased water permeability as well as in the mobilization of solutes from the small vacuoles on rehydration. This is the first report of a possible TIP 3;1 in vegetative tissues (previously only reported in orthodox seeds).  相似文献

For functioning of a root system, the temporal development of distribution of roots in the soil is important. For example, for computing uptake of water and nutrients the root length density distribution might be required. A way to describe root proliferation is to consider it as a diffusion process with a first-order sink term accounting for decay. In this paper, analytical solutions are derived for two-dimensional diffusion of roots both in a rectangular area, and in a cylindrical volume. The source of root dry matter is located at the surface. Root dry matter enters the soil domain through a part of the soil surface. It is shown that different distribution patterns are obtained, with different ratios of the diffusion coefficients in horizontal and vertical direction. From the solutions obtained it can be shown that for the situation where the dry matter enters through the complete surface eventually a steady-state occurs where root length density decreases exponentially with depth, as often is found in experiments.  相似文献

Aboveground dry mass, total root dry mass and root length density of the fine roots of Thuja occidentalis `Brabant' were determined under non- and drip-irrigated field conditions. Two-dimensional diffusion parameters for dynamic root growth were estimated based on dry mass production of the fine roots and the concept of the convective-diffusion model of cylindrical root growth and proliferation. Drip irrigation increased above-ground dry mass and the shoot:root ratio compared with no irrigation. Dry mass of the coarse roots increased as well due to drip irrigation. No effect on total or fine root dry mass was found. Drip irrigation increased root length densities in the top 0.1 m but not significantly. However, drip irrigation decreased root proliferation in depth by 27%, whereas proliferation in the horizontal direction was not altered. Measured root length densities were overestimated by 6–21% by the model (0.68 < R 2 < 0.92).  相似文献

How the brain maintains an accurate and stable representation of visual target locations despite the occurrence of saccadic gaze shifts is a classical problem in oculomotor research. Here we test and dissociate the predictions of different conceptual models for head-unrestrained gaze-localization behavior of macaque monkeys. We adopted the double-step paradigm with rapid eye-head gaze shifts to measure localization accuracy in response to flashed visual stimuli in darkness. We presented the second target flash either before (static), or during (dynamic) the first gaze displacement. In the dynamic case the brief visual flash induced a small retinal streak of up to about 20 deg at an unpredictable moment and retinal location during the eye-head gaze shift, which provides serious challenges for the gaze-control system. However, for both stimulus conditions, monkeys localized the flashed targets with accurate gaze shifts, which rules out several models of visuomotor control. First, these findings exclude the possibility that gaze-shift programming relies on retinal inputs only. Instead, they support the notion that accurate eye-head motor feedback updates the gaze-saccade coordinates. Second, in dynamic trials the visuomotor system cannot rely on the coordinates of the planned first eye-head saccade either, which rules out remapping on the basis of a predictive corollary gaze-displacement signal. Finally, because gaze-related head movements were also goal-directed, requiring continuous access to eye-in-head position, we propose that our results best support a dynamic feedback scheme for spatial updating in which visuomotor control incorporates accurate signals about instantaneous eye- and head positions rather than relative eye- and head displacements.  相似文献
