Abstract. Although the oesophageal appendages in the four enchytraeids Enchytrueus crypticus, Fredericia strinta, Buchholzia appendiculata , and Achaeta sp. are quite different from one another in shape and position, their histology and ultrastructure are basically the same. These are intestinal appendages, the lumina of which distally end blind and proximally open into the oesophagus. Almost all of the few cells in their single-layered epithelium have a microvillous, cilia-free border at the apex, facing towards the lumen, and basally comprise an extremely extensive labyrinth. The presence of the latter, composed of very thin cell processes, and of numerous mitochondria identifies the organs as energy-producing and -consuming, transport-active structures. Their possible function as a food-moistening organ or osmoregulatory organ is discussed, and they are compared with other intestinal appendages in enchytraeids and other oligochaetes.  相似文献

Plasma protein extravasation was studied in the rat abdominal skin. Substance P (SP), neurokinin A (NKA) and B (NKB) were found to induce extravasation with a threshold dose of about 1 pmol. Calcitonin gene-related peptide (CGRP) caused no or little extravasation alone but it potentiated the action of SP, NKA, NKB, and physalaemin. The potentiation of the SP-induced extravasation was unaffected by pretreatment with capsaicin, indomethacin or compound 48/80, it was reduced by neuropeptide Y or pretreatment with mepyramine plus cimetidine, and was abolished in streptozotocin diabetic rats. CGRP augmented extravasation induced by histamine, reduced the effect of ATP or adenosine and did not alter extravasation by serotonin, bradykinin or neurotensin. These results indicate that in addition to SP the novel mammalian tachykinins NKA and NKB may be considered as mediator candidates for neurogenic plasma extravasation. CGRP is a possible mediator of antidromic vasodilation. Furthermore, CGRP potentiates the extravasation caused by coexisting tachykinins and could thereby augment neurogenic inflammation. The diverse interactions of CGRP with other inflammatory mediators suggest multiple sites of action.  相似文献

Summary Fractionation of rabbit skeletal muscle cytosol on Aminohexyl-Sepharose has resulted in the identification of a latent ATP, Mg-dependent protein phosphatase whose catalytic subunit is in the active conformation, but is inhibited by the presence of more than one modulator unit. The partially purified enzyme is converted to an inactive, kinase FA-dependent form upon incubation at 30°C unless modulator-specific polyclonal antibodies are added to the preparation. The immunoglobulins also relieve the inhibition which is responsible for the low basal phosphatase activity of the enzyme, and they counteract all of the heat-stable inhibitor activity present in the preparation. Addition of free catalytic subunit abolishes the inhibition of the latent enzyme in a dose-dependent way, but cannot prevent the inactivation process. Cytosolic alcohol dehydrogenase in the deermouse is coded by a single genetic locus and a strain of the deermouse which is alcohol dehydrogenase negative exists. These two strains of the deermouse were used to extend insight into the role of cytosolic alcohol dehydrogenases in the conversion of retinol into retinoic acid. Retinoic acid synthesis from physiological concentrations of retinol (7.5 microM) with cytosol from the alcohol dehydrogenase negative deermouse was 13% (liver), 14% (kidney), 60% (testes), 78% (lung), and 100% (small intestinal mucosa) of that observed with cytosol from the positive deermouse. The rates in the negative strain ranged from 0.3 to 0.7 nmol/h/mg protein: sufficient to fulfill cellular needs for retinoic acid. Ten millimolar 4-methylpyrazole inhibited retinoic acid synthesis 92, 94, 26, and 30% in kidney, liver, lung, and testes of the positive deermouse, respectively, but only 50, 30, 0, and 0% in the same tissues from the negative deermouse. Ethanol (300 mM) did not inhibit retinoic acid synthesis in kidney cytosol from the negative strain. Therefore multiple cytosolic dehydrogenases, including alcohol dehydrogenases, contribute to retinol metabolism in vitro. The only enzyme(s) likely to be physiologically significant to retinoic acid synthesis in vivo, however, is the class of dehydrogenase, distinct from ethanol dehydrogenase, that is common to both the positive and the negative deermouse. This conclusion is supported by the data described above, the kinetics of retinoic acid synthesis and retinal reduction in kidney cytosol from the negative deermouse, and the very existence of the alcohol dehydrogenase negative deermouse. This work also shows that microsomes inhibit the cytosolic conversion of retinol into retinoic acid and that the synthesis of retinal, a retinoid that has no known function outside of the eye, does not reflect the ability or capacity of a sample to synthesize retinoic acid.  相似文献

Plasmalemma-rich microsomal vesicles were prepared from whole leaf and acid-washed epidermal tissue of Vicia faba L. cv. Osnabrücker Markt by aqueous two-phase partitioning in dextran T-500 and polyethylenglycol 1350 aqueous phases. These vesicles were tightly sealed and predominantly right-side out, and contained a K+ -stimulated, mg2+-dependent and vanadate-sensitive ATPase. The enzyme from both tissues exhibited nearly identical properties: pH optimum 6.4, Km for ATP 0.60 mM(whole leaf) and 0.67 mM (epidermis). Vmax -480 nmol (mg protein)1 min1 (whole leaf) and 510 nmol (mg protein)1 min1 (epidermis), I50 (Na3,VO4) 7.5 μM (whole leaf) and 15 μM (epidermis). The enzyme was not inhibited by NO3(50 mM)or sodium azide (I mM). DCCD (20 μM) reduced enzyme activity to 50% (whole leaf) and 58% (epidermis), gramicidin S (20 μM) to 36% (whole leaf) and 41%(epidermis). Ca2+ inhibited the ATPase [I50, C2+: 0.5 mM(whole leaf) and 0.8 mM(epidermis)]. Ca2+ inhibited the ATPase [I50, C2+ 0.5 mM(whole leaf) und 0.8 (epidermis)]. The vanadate-sensitive ATPase from whole leaf and epidermal tissue was slightly but significantly stimulated by fusicoccin (FC) at a concentration (0.13 μM) promoting stomatal opening. The stimulation was not seen in the solubilized ATPase. Stomata of the cultivar used here were insensitive lo (±)ABA up to 2 μM level which is effective in most other cultivars and species. Likewise, at this concentration no effect of ABA on the activity of the epidermal ATPase was observed. The data are discussed with respect to the interaction of FC and ABA with the ATPase.  相似文献

Populations ofTococa occidentalis (Melastomataceae) and the inhabiting ants (Myrmelachista sp.) were observed for more than eight months in the Peruvian Amazon (Sira mountains). They represent a complex coevolutionary system: the plants offer shelter (leaf domatia, hollow stems) and food (leaf glands), whereas the ants kill all surrounding plants, including large trees up to 10 m, by chemical weapons. Experiments with exposed plants revealed a highly specialized way to attack meristematic tissue and leaf nervature, which leads to a quick decay of the plant individuals. The clearing of the vegetation by the ants allows theTococa population to expand mostly by vegetative shoots to large monocultures (up to 30 m in diameter) free from any other plant species. Artificially introduced plant individuals, from differentT. occidentalis populations, are regarded as a foreign species by the ants.The succession of such aTococa-Myrmelachista system begins with one or a few founder plants on a light place in the midst of the vegetation.Myrmelachista soon inhabits their host plants which otherwise would not survive and begin to clear the place from all foreign plant species.Tococa expands quickly, forming circle shaped populations. Distantly situated canopy trees shade theTococa population after a number of years and cause their decay. The whole place appears contaminated for years and no other plant can establish itself. Some of the consequences of these open places are erosion and a severe influence on the regeneration of the forest.  相似文献
