[目的]为了确定铜绿假单胞菌调控因子Pip对两个不同吩嗪合成基因簇(phz1和phz2)的具体调控方式与可能的调控机制.[方法]根据基因比对结果,采用同源重组技术构建Pip调控因子缺失突变株PA-PG以及克隆ip基因作互补分析;再以已构建的吩嗪基因簇缺失突变株PA-Z1G和PA-Z2K为受体菌,构建突变株PA-PD-Z1G和PA-PG-Z2K,测定并比较野生株及相关突变株的吩嗪-1-羧酸和绿脓菌素的合成量,推定Pip对两个不同吩嗪合成基因簇的调控方式.[结果]在GA培养基中,突变株PA-PG的吩嗪-1-羧酸和绿脓菌素都比野生型明显减少;互补分析显示,突变株PA-PG的吩嗪-1-羧酸和绿脓菌素都显著提高并恢复到野生株PAO1水平;突变株PA-Z1G的吩嗪-1-羧酸和绿脓菌素合成量因Pip缺失而显著减少;而突变株PA-Z2K的吩嗪-1-羧酸和绿脓菌素合成量在Pip缺失后仍保持不变.[结论]初步推定,转录调控因子Pip对铜绿假单胞菌吩嗪合成代谢的确具有促进作用;Pip通过正向调控吩嗪基因簇phz2的合成功能实现对吩嗪合成代谢的调控.  相似文献

胶质瘢痕是神经系统损伤后由反应性星形胶质细胞,小胶质细胞及其分泌的细胞外基质组成。早期的研究多集中于胶质瘢痕在抑制轴突生长,神经细胞再生等方面的作用。而最新的研究表明胶质瘢痕的形成对损伤急性期神经细胞具有重要的保护作用。本文从瘢痕组织在损伤缝合和组织重构、局部免疫调节、神经再生等方面对神经损伤的保护作用进行综述。  相似文献

SIRT2 is a tubulin deacetylase, which can play either detrimental or beneficial roles in cell survival under different conditions. While it has been suggested that reduced SIRT2 expression in human gliomas may contribute to development of gliomas, there has been no study that directly determines the effects of decreased SIRT2 activity on the survival of glioma cells. In this study we applied both pharmacological and molecular approaches to determine the roles of SIRT2 in the survival of glioma cells. Our studies, by conducting such assays as flow cytometry-based Annexin V assay and caspase-3 immunostaining, have indicated that decreased SIRT2 activity leads to apoptosis of C6 glioma cells by caspase-3-dependent pathway. Our experiments have further shown that reduced SIRT2 activity produces necrosis of C6 glioma cells. Moreover, our study applying SIRT2 siRNA has also shown that decreased SIRT2 leads to both necrosis and apoptotic changes of C6 glioma cells. Collectively, our study has provided novel evidence indicating that SIRT2 activity plays a key role in maintaining the survival of glioma cells, and that reduced SIRT2 activity can induce both necrosis and caspase-3-dependent apoptosis of C6 glioma cells. These results have also suggested that inhibition of SIRT2 might become a novel therapeutic strategy for gliomas.  相似文献

Solution (1)H NMR spectroscopy has been carried out to investigate the molecular and electronic structures of the active site in H64Q/V68F double mutant mouse neuroglobin in the cyanomet form. Two heme orientations resulting from a 180 degrees rotation about the alpha-gamma-meso axis were observed with a population ratio about 1:1, and the clearly distinguished B isomer was used to perform the study. Based on the analysis of the dipolar shifts and paramagnetic relaxation constants, the distal Gln(64)(E7) side chain is obtained to adopt an orientation that may produce hydrogen bond between the N(epsilon)H(1) and the Fe-bound cyanide. The side chain of Phe(68)(E11) is oriented out of the heme pocket just like that in triple mutant of cyanide complex of sperm whale myoglobin. A 15 degrees rotation of the imidazole ring in axial His(96) is observed, which is close to the varphi angle determined from the crystal structure of NgbCO. The quantitative determinations of the orientation and anisotropies of the paramagnetic susceptibility tensor reveal that cyanide is tilted by 8 degrees from the heme normal which allows for contact to the Gln(64)(E7) N(epsilon)H(1). The E7 and E11 residues appear to control the direction and the extent of tilt of the bound ligand. Furthermore, the tilt of the ligand has no obvious influence on the heme heterogeneity of cyanide ligation for isomer A/B of the wild type and mutant protein, indicating that factors other than steric effects, such as polarity of heme pocket, impacts on ligand binding affinity.  相似文献

The efficiency of perovskite solar cells (PSCs) is hindered by substantial defects within the grain boundaries (GBs) of polycrystalline perovskite films. Conventional post-treatment strategies struggle to precisely repair these defects at GBs. Here, a targeted grain boundary passivation strategy through solvent effects by incorporating symmetrical biphenyl molecules is proposed, 4,4′-diaminodiphenyl sulfone (DDS) and 4,4′-sulfodiphenol (SDP), aiming to mitigate defects at GBs and optimize energy level arrangements through their electric dipole effects. Compared to the pristine device, the SDP-modified device exhibits significant improvements, including a champion efficiency of 24.39% and an impressive fill factor, along with excellent operation stability. This work provides an effective and straightforward solution for improving the performance of PSCs.  相似文献

NAD+ plays important roles in various biological processes. In this study, we reported that treatment of NAD+ induces delayed autophagy in Neuro2a cells. Moreover, the effects of NAD+ on the autophagy in the cells appear to be, at least partially, mediated by oxidative stress. However, nicotinamide, a degradation product of NAD+, does not affect the autophagy. Our experiments have further indicated that the NAD+-induced autophagy contributes to the NAD+-induced decrease in the survival of these cells. In summary, our study has provided the first evidence that NAD+ treatment induces autophagy in cancer cells such as Neuro2a cells, which contributes to the NAD+-induced decrease in cancer cell survival.  相似文献

千粒重是大麦产量的重要构成因素之一。青藏高原强辐射、低温、干旱的生态环境孕育了现代农业所急需的大麦种质资源,但是迄今为止尚未见到有关青藏高原栽培大麦WTS与环境因子关系的系统性研究报道。为了揭示青藏高原栽培大麦千粒重的空间分布规律,探明不同环境因子对青藏高原栽培大麦千粒重(WTS)积累的影响程度,利用83个样点的地理、气候、土壤因子数据,研究了青藏高原栽培大麦WTS的分布特征。结果表明:(1)在地理水平方向上,青藏高原栽培大麦WTS总体呈现出斑块状交错分布的格局,形成了以西藏曲水、堆龙德庆、白朗、乃东、日喀则、扎囊、贡嘎、加查、达孜、谢通门、拉孜、定日为中心的青藏高原西南部和青海海晏、门源、刚察为中心的青藏高原东北部等2个栽培大麦WTS高值区;(2)在地理垂直方向上,栽培大麦WTS的变化呈现出"N"型分布格局,即在海拔3600.0—3900.0m和4500.0m以上形成2个WTS高值区,这2个海拔区间栽培大麦WTS分别为(49.6815±10.0764)g和(47.9500±0.1732)g;(3)影响栽培大麦WTS的环境因子从大到小的顺序是抽穗-成熟期降水量土壤速效钾含量分蘖-拔节期日照时数抽穗-成熟其平均气温抽穗-成熟期日照时数拔节-抽穗平均气温日较差地理经度。  相似文献

选择烟台海岸沙地抗沙埋强的单叶蔓荆(Vitex trifolia var.simplicifolia)为试材,在自然环境条件下根据单叶蔓荆匍匐茎长度进行了轻度(1/3茎长)、中度(2/3茎长)和重度半埋以及全埋处理。在沙埋20d后,测定了不同沙埋处理下匍匐茎各段上匍匐茎长度、枝条高度、不定根长度,以及可溶性糖、淀粉、纤维素含量,以探讨单叶蔓荆碳水化合物变化和转化在其耐沙埋中作用。结果显示,在轻度、中度半埋和全埋下单叶蔓荆匍匐茎长度均显著大于对照,被沙埋匍匐茎处有大量不定根生成;同时,可溶性糖和淀粉含量增高和纤维素含量下降,尤其是生长最快的匍匐茎顶部(如轻度半埋),茎中可溶性糖较低、淀粉增加最多,纤维素最低。但是被重度半埋和全埋的匍匐茎生长较少,茎中纤维素含量较多、淀粉含量较少。研究表明,沙埋是一种胁迫,它损伤叶片、扰乱碳水化合物代谢平衡。但它又是胁迫信号使植物产生适应性反应,它使未遭沙埋的匍匐茎顶端通过加速碳水化合物转化、分解纤维素、提高淀粉和可溶性糖含量,为顶端生长提供能量和营养,以加速匍匐茎快速生长摆脱沙埋。同时沙埋部位枝叶通过分解其纤维素,产生更多的可溶性糖和淀粉为匍匐茎不定根生长提供能量。因此,沙埋后匍匐茎内碳水化合物的转化是其快速生长和摆脱沙埋的能量来源而在其适应沙埋生长中起重要作用。单叶蔓荆对沙埋的适应性反应表现了其具有表型可塑性特性,该特性是其沙埋后维护匍匐茎顶部快速生长、不定根形成、碳水化合物转化以及具有较高抗沙埋能力的关键。  相似文献

广西乐业大石围天坑群(以下简称大石围天坑群)是最典型的塌陷型天坑群。该研究采用样线法和样方法对大石围天坑群的种子植物进行了实地调查,并结合已有资料综合分析了大石围天坑群种子植物区系特征。结果表明:大石围天坑群野生种子植物丰富,有137科445属863种;在科级和属级水平上,地理成分以热带成分为主,中国特有成分相对贫乏;与热带区系的联系主要以泛热带成分为主,与温带区系的联系主要以北温带成分为主;大石围天坑群的热带科和温带科之比以及热带属和温带属之比,均小于中国乐业-凤山世界地质公园,其种子植物区系更能反映该地区过去植物组成的"原貌",即温带成分比例过去的比现代的高,是全球气候变暖的有力证据:天坑群保育了82种珍稀濒危植物,包括30属67种野生兰科植物,是现存珍稀濒危植物的"避难所"。  相似文献

社鼠(Niviventer confucianus)属于啮齿目(Rodentia)、鼠科(Muridae)、白腹鼠属(Niviventer),关于该物种的分子系统学研究极少。为获取社鼠线粒体基因组全序列,提取其基因组总DNA,参照近缘物种线粒体基因组全序列设计34对特异性引物,利用PCR扩增全部片段后进行测序,之后对其基因组组成及结构特点进行了初步分析。结果表明,社鼠线粒体基因组全序列长16 281 bp(GenBank收录号:KJ152220),包含22个tRNA基因、13个蛋白质编码基因、2个rRNA基因和1个非编码控制区;基因组核苷酸组成为34.0%A、28.6%T、24.9%C、12.5%G。将所得序列与社鼠近缘物种(川西白腹鼠、小家鼠、褐家鼠)的线粒体全基因组进行比较,结果显示,四个物种的线粒体基因组虽然在基因组大小、部分tRNA二级结构、部分蛋白质编码基因的起始或终止密码子及控制区长度和碱基组成上有差异,但基因组结构和序列特征方面都具有较高的相似性。四个物种线粒体全基因组间的遗传距离显示,社鼠与川西白腹鼠距离最近,而与小家鼠距离最远。该研究为利用线粒体全基因组信息进行啮齿类分子系统学研究提供了有价值的资料。  相似文献
