To characterize the functional cell surface, the zeta potentials and elemental surface composition of Streptococcus salivarius HB and a range of mutants with known molecular surface structures were determined. Zeta potentials of fully hydrated cells were measured as a function of pH in dilute potassium phosphate solutions, yielding isoelectric points of the strains. Elemental composition (O, C, N, and P) of the outer 2 to 5 nm of the freeze-dried cell surfaces were measured by X-ray photoelectron spectroscopy. An increasing loss of proteinaceous fibrillar surface antigens of the mutants was found to be accompanied by a progressive decrease in the N/C ratio from 0.104 in the parent strain HB to 0.053 in mutant HBC12. Simultaneously, the value of the isoelectric point shifted from 3.0 to 1.3. In a previous study (A.H. Weerkamp, H.C. van der Mei, and J. W. Slot, Infect. Immun. 55:438-455, 1987) on the cell surfaces of the same strains, it was shown that removal of fibrils led to increased exposure of (lipo)teichoic acid at the surface, which explains the low isoelectric point caused by the low pKa of the phosphate groups.  相似文献

The adhesion of Streptococcus mitis to solid substrata from phosphate suspensions with various ionic strengths was studied and compared with the adhesion of polystyrene particles. At all ionic strengths, the interfacial free energy of adhesion governed the relative number of bacteria or polystyrene particles adhering at equilibrium, except that in a low-ionic-strength buffer, adhesion occurred less frequently because of increased electrostatic repulsion. Large differences between bacterial and polystyrene particle adhesion were observed, as indicated by the ratio of bacteria to polystyrene particles adhering, which decreased from 30 to 4 with a change from low to high ionic strength.  相似文献

A human feeding study was performed with Lactococcus lactis TC165.5, which is genetically marked by insertion of the sucrose-nisin conjugative transposon Tn5276 and chromosomal resistance to rifampin and streptomycin. The fate of strain TC165.5 and its nucleic acids was monitored by conventional plating methods and by molecular detection techniques based on specific PCR amplification of the nisin (nisA) gene from DNA extracted from human feces. A method was developed for the efficient extraction of microbial DNA from human feces. The results show that a fraction of viable cells of L. lactis TC165.5 survived passage through the human gastrointestinal tract. Only cells that passed within 3 days of ingestion could be recovered from the feces of the volunteers, and they accounted for approximately 1% of the total number of cells consumed. The presence of nisA in DNA extracted from feces could be detected up to 4 days, when viable cells were no longer present.  相似文献

1. Starved cells of a glucose-grown strain of Staphylococcus aureus are resistant to the action of staphylococcin 1580. Reinitiation of sensitivity is readily obtained upon the addition of glucose, but only weakly with L-lactate, although the latter induces higher ATP levels and supports L-glutamic acid uptake better than glucose does. The NADH/NAD+ ratio correlates with the staphylococcin sensitivity. 2. Starved pyruvate-grown cells remain partially susceptible and full sensitivity is restored both in the presence of glucose and L-lactate. 3. Arsenate but not dicyclohexylcarbodiimide (DCCD) blocks the reinitiation of sensitivity in the presence of glucose. Both arsenate and DCCD block sensitivity in the presence of L-lactate. 4. Aerobically grown cells are sensitive to staphylococcin 1580 under anaerobic conditions. Anaerobically grown cells are less susceptible, but sensitivity can be restored by glucose and also by L-lactate plus nitrate when cells are previously induced for nitrate reductase. 5. Starved cells of a mutant strain defective in the maintenance of a high-energy state of the membrane are normally sensitive in the presence of glucose, but resistant in the presence of L-lactate. A strain lacking a functional respiratory chain (men-) is also sensitive with glucose but resistant in the presence of L-lactate. 6. It is concluded that the initiation of the staphylococcin 1580 action is under control of a mechanism regulating the energy flow in the cell, and involving the presence of a high-energy phosphorylated compound.  相似文献

The presence of lactose-utilizing Lactococcus species in nondairy environments was studied by using identification methods based on PCR amplification and (sub)species-specific probes derived from 16S rRNA sequences. Environmental isolates from samples taken on cattle farms and in the waste flow of a cheese production plant were first identified to the genus level, using a Lactococcus genus-specific probe. Isolates which showed a positive signal with this probe were further identified to the (sub)species level. Lactococcus lactis isolates were also characterized at the phenotypic level for the ability to hydrolyze arginine, to ferment citrate, and to produce proteases and bacteriocins. With specific PCR amplifications, the presence of sequences related to citP, coding for citrate permease; prtP, coding for protease; and nisA or nisZ, the structural genes for production of nisin A or nisin Z, respectively, was verified. By these methods, it was possible to isolate lactococci from various environmental sources, such as soil, effluent water, and the skin of cattle. The strains of L. lactis isolated differed in a number of properties, such as the ability to hydrolyze arginine or the absence of citP-related sequences, from those found in industrial starter cultures. The results indicate that the majority of the industrially produced lactococci do not survive outside the dairy environment, although natural niches are available. However, from those niches strains with the potential to be developed into novel starter cultures may be isolated.  相似文献

The kinetics of adhesion of Streptococcus sanguis CH3 from suspension to polymers with different surface free energies were studied by using three bacterial concentrations (2.5 X 10(7), 2.5 X 10(8), and 2.5 X 10(9) cells per ml-1). Substratum surface free energies (gamma s) ranged from 18 to 120 erg cm-2. The kinetics of bacterial adhesion to these surfaces showed a typical two-step adhesion process, indicating an equilibrium in both steps. In the initial adhesion step (step 1), low equilibrium numbers of adhering bacteria were counted on substrata with surface free energies lower than 55 erg cm-2. A maximal number adhered on substrata with higher surface free energies. At the lowest bacterial concentration tested, the highest number of bacteria were found on substrata with a surface free energy around 55 erg cm-2. For each substratum, step 2 started after a characteristic time interval tau, being short (30 min) for gamma s less than 50 and long (120 min) for gamma s greater than 50 erg cm-2. The relationship between the substratum surface free energy and the number of bacteria adhering at equilibrium after step 2 was similar to, although less distinct than, that during step 1 with a slight indication of a bioadhesive minimum around gamma s = 35 erg cm-2. The results are indicative of a two-step adhesion model, in which step 1 is controlled by macroscopic substratum properties.  相似文献

Abstract The surface free energies ( γ b) of a variety of oral streptococci were determined from contact angle measurements on bacterial deposits, using the concept of dispersion and polar components. At least four strains of each species were tested. Strains of Streptococcus mutans, S. sanguis and S. salivarius possessed relatively high surface free energies (103 ± 12 mJ · m−2) and at the species level no significant difference was found. In contrast, the strains of S. mitis had remarkably low surface free energies (45 ± 14 mJ · m−2). S. milleri appeared to be a heterogeneous species, showing surface free energies over a range of 32–119 mJ · m−2. No significant differences were observed between laboratory strains and strains freshly isolated from the oral cavity.  相似文献

A flow cell system was developed which allowed the study of bacterial adhesion to solid substrata at well-defined shear rates. In addition, the system enabled the solid surfaces to be coated with a proteinaceous film under exactly the same shear conditions. In this flow cell system, adhesion of three strains of oral streptococci from a phosphate-buffered solution onto three different substrata was studied as a function of time in the absence and presence of a bovine serum albumin (BSA) coating at a shear rate of 21 s-1. To obtain a wide range in surface free energies (gamma) representative strains (gamma b 38-117 mJ m-2) and solid substrata (gamma s 20-109 mJ m-2) were selected. The number of bacteria adhering was counted microscopically. In the absence of a BSA coating a linear relation was found between the number of bacteria adhering at saturation (nb,s) and the calculated interfacial free energy of adhesion (delta Fadh) for each of the three strains. In the presence of a BSA coating the number of bacteria adhering was greatly decreased in all cases. However, despite the presence of the BSA coating there was still a linear relation between the number of bacteria adhering at saturation and the interfacial free energy of adhesion, calculated on the basis of the surface free energy of the uncoated substrata. It can be concluded that the bare, uncoated substratum still influenced bacterial adhesion in spite of the marked influence of a BSA coating.  相似文献
