The chromosome-breaking activity of four 1-(phenyl)-3,3-dimethyltriazenes was tested in vitro on human peripheral blood lymphocytes using S9 mix as a metabolic activation system. 1-(4-Nitrophenyl)-3,3-dimethyltriazene was the most active compound. The difference in the frequency of chromosomal aberrations in a test with and without metabolic activation was significant at the 1% level of significance. The lowest frequency of chromosomal aberrations was induced by 1-(4-methylphenyl)-3,3-dimethyltriazene which, under the conditions of this experiment, is the least stable and probably rapidly degraded to non-active compounds. The chromosomal aberrations were also induced by 1-(4-chlorophenyl)-3,3-dimethyltriazene and 1-(4-bromophenyl)-3,3-dimethyltriazene, this activity was unrelated to metabolic activation.  相似文献

The genetic risk of workers occupationally exposed to a series of newly developed cytostatic drugs and the presumed antimutagenic potential of ascorbic acid (AA) were studied in a group of 38 chemical laboratory personnel examined for chromosome aberrations in lymphocytes, urine mutagenicity and nucleolar RNA activity before and after a 6-month prophylactic administration of AA at daily doses of 1 g for 5 days a week. Chromosome aberration tests revealed elevated aberrant cell (AB.C) rates both prior to and after AA supplementation (3.9% and 3.65% of AB.C., respectively). These values were significantly higher than those found in 18 non-exposed matching controls (1.05% of AB.C.). Tests for mutagenic activity in the urine of drug-exposed workers revealed 64% positive urine samples prior to vitaminization and 60% positive urine specimens after it; positive urine samples in the group of controls accounted for 21% of samples. In the nucleolus test, numbers of inactivated micronuclei in the exposed were initially higher than those of controls (33.4% versus 24.3%), but dropped to 20.5% after AA supplementation. These findings show that AA prophylaxis alone cannot substantially reduce the hazards associated with exposure to anti-cancer drugs.  相似文献

The condensates collected after pipe smoking of a natural tobacco and a cavendish type tobacco, either unwrapped or wrapped in a paper "saver" bag, were tested for mutagenicity in the Salmonella/mammalian microsome assay with strains TA100 and TA98. The number of revertants induced with cavendish type tobacco in the presence of metabolic activation (mouse-liver S9) was higher in both strains compared to the natural tobacco. Further increase in the number of revertants (approx. 3 times) was consistently seen when the tobacco was smoked after paper wrapping "savers".  相似文献

The population energetics of a temporary and a permanent pond population of Musculium partumeium in Southwest Ohio were studied. In the permanent pond (surface area = 396 m2, maximum depth = 0.7 m) the population was bivoltine and iteroparous whereas in the temporary pond (surface area = 1042 m2, maximum depth = 0.9 m) the population was usually univoltine and semelparous.Growth and biomass were assessed as total organic carbon and total nitrogen to provide estimates of productivity and seasonal changes in C:N for each generation. Productivity (non-respired assimilation = growth + reproduction; N-R.A. = G + R) was 6939 mgC·m–2·a–1 (3858 and 3353 mgC·m–2·a–1 for each generation) and 1661 mgC·m–2·a–1 for the permanent and temporary pond populations respectively. The average standing crop biomass (B) was 606.8 mgC·m–2 (357.5 and 249.3 mgC·m–2 for each generation) and 231.9 mgC·m–2 with overall productivity: biomass ratios of 11.4 and 7.2 for the permanent pond and temporary pond populations respectively.Respiration rates were converted to carbon equivalents (respired assimilation = R.A.) and used to evaluate the components of total assimilation (T.A. = R.A. + N-R.A.) and the efficiency of partitioning this energy to N-R.A. for G and R. When expressed as a percentage, the production efficiencies (100 × N-R.A.:T.A.) were 50.4 and 62%, and the reproductive efficiencies (100 × R:N-R.A.) were 26.4 and 18% for the permanent and temporary pond populations respectively. The reproductive efficiencies for populations of these viviparous clams are greater than those for most oviparous molluscs.The comparative information on the energetics of these populations does not completely fit any theoretical consideration of reproductive effort or life-history strategy. The effects of particle concentration and season on the filtration rates of the freshwater clamSphaerium striatinum Lamarck were assessed by measuring clearance rates of small (2.02 μm) latex beads from dilute suspensions. Filtration rates decreased as particle concentration increased over a range of 2–64 mg 1−1, with rates decreasing in similar proportion for clams of all sizes. For a 1-mg clam, rates decreased from approximately 8.4 to 0.57 ml clam−1 h−1. Seasonal filtration rates for adult clams peaked during periods of greatest reproduction. The patterns for smaller clams are similar, though proportional changes in filtration rates differ for various sizes of clams. It is estimated that clams occupying 1 m2 of stream substrate removed about 3.67 gCa−1. This is equivalent to 0.0004% of the carbon that flows past them annually. Filter-feeding provided only 24% of the calculated energy needs of the population, suggesting that another mode of feeding (e.g. deposit-feeding) may provide an important energy source for these forms. Sarcocystis-like oocysts-sporocysts were found in four species of owls (Asio otus, Bubo bubo, Strix aluco, and Tyto alba) and in five species of predatory birds (Accipiter gentilis, Accipiter nisus, Buteo buteo, Circus aeruginosus, Falco tinnunculus). In addition, the muscles of 15 of 41 (36.5%) pheasants (Phasianus colchicus) and one of two jays (Garrulus glandarius) were found to harbor three types of Sarcocystis. Three of 15 (20%) infected pheasants had type I cystozoites (6-8 X 2 microns) in muscle homogenates, but sarcocysts were not seen whereas the other 12 infected pheasants had type II cystozoites (16 X 2-3 microns) and sarcocysts (90 X 600 microns) in their muscles. The one infected jay had type III cystozoites (8-10.5 X 2.5-3 microns) and sarcocysts (35-40 X greater than 770 microns) in its muscles.  相似文献

Summary An endothelial cell line derived from a massive recurrent chyle-containing retroperitoneal lymphangioma was isolated in monolayer culture. Scanning and transmission electron microscopy and immunohistochemistry confirmed a close resemblance to blood vascular endothelium with typical cobblestone morphology, positive immunofluorescence staining for endothelial marker Factor VIII-associated antigen and fibronectin, and prominent Weibel-Palade bodies. The endothelial cells also exhibited other ultrastructural features characteristic of lymphatic endothelium, including sparse microvillous surface projections, overlapping intercellular junctions, and abundant intermediate filaments. This endothelial cell line represents a new source of proliferating lymphatic endothelium for future study, including structural and functional comparison to blood vascular endothelium. About 10% of 3887 ever-married women included in the 1984-85 Malaysian Population and Family Survey revealed that they were influenced by the new population policy to desire more children than they had originally wanted. These women were more likely to be rural Malays from the lower socioeconomic class. Ideal family size was more than four children. Children are desired for economic benefits and emotional support. The natality of the Malays has risen since 1980: their total fertility rate has increased while their contraceptive prevalence rate has dropped sharply. Coupled with a decline in the crude death rate, the present fertility preferences and behaviour of the Malays will render the target of the population policy more attainable than is reflected by the survey data.  相似文献
