Relation between endurance time and maximal oxygen consumption during supramaximal running

The relationship between speed and the maximal length of time supramaximal runs can be sustained (temps-limite, tlim) has been studied in seven male subjects (physical education students). Within the range of intensity studied, tlim strictly depends on maximal oxygen consumption (VO2max). The relationship between tlim and the relative energy cost of the exercises per unit of time (E), calculated by subtracting the maximal power of aerobic metabolism (Emaxox) from E, removes the interindividual differences of tlim. The function tlim = f(E-Emaxox) is described by an empirical equation of the form: tlim = a.exp[-b(E-Emaxox)] (r = 0.979; P less than 0.001), where the parameters a and b are respectively equal to 330.8 and 0.14 and where tlim, E and Emaxox are respectively expressed in seconds and in watts per kg of body weight.     

Age and sex composition of small ruminants at Nouakchott markets, Mauritania

A survey of species, breeds, sex and age composition of sheep and goats offered for sale was made at Nouakchott (Mauritania) livestock markets. Some 82.8% of animals presented at the markets were sheep and 95% of these were of the Short-haired Maure breed, the remainder being Long-haired Maure. Goats of the Sahel breed comprised 81% of all animals of this species, the others being Sahara and Eastern Dwarf types, but goats represented only 17.2% of all animals brought to market. Castrated Short-haired Maure sheep were 63.6% of this breed whereas castrates of Long-haired Maure sheep were 40.6% of the total. There were significantly more castrates than entire males presented for sale for sheep but the reverse was the case for goats. Entire males of both species were sold at younger ages than castrates and males and castrates combined were sold at younger ages than females. This study has provided some indications of the types of stock offered for sale but further studies at peak sale times would allow more detailed analysis. Such an analysis should be coupled with a survey of owners/herders and of traders in order to determine the factors operating on market forces.     

Cardiovascular changes induced by isometric exercise in aged subjects

Blood pressure and heart rate during static contraction of forearm muscles holding a tension of 30% maximal voluntary strength for 5 mn have been measured. Eleven young men and 11 elderly male subjects were used. Maximal voluntary strength is reduced by 19% in elderly subjects. Relative mean cardio-vascular responses were unaffected by age.     

Impact of spray-drying on the pili of Lactobacillus rhamnosus GG

The preservation of the viability of microorganisms in probiotic formulations is the most important parameter ensuring the adequate concentration of live microorganisms at the time of administration. The formulation and processing techniques used to produce these probiotic formulations can influence the preservation of the microbial viability. However, it is also required that the bacteria maintain their key probiotic capacities during processing, formulation and shelf life. In this study, we investigated the impact of spray-drying on different cell wall properties of the model probiotic strain Lactobacillus rhamnosus GG, including its adherence to intestinal epithelial cells. The dltD gene knock-out mutant, L. rhamnosus GG CMPG5540, displaying modified cell wall lipoteichoic acids, showed significantly increased colony-forming units after spray-drying and subsequent storage under standard conditions compared to wild-type L. rhamnosus GG. In contrast, disruption of the biosynthesis of exopolysaccharides or pili expression did not impact survival. However, spray-drying did significantly affect the adherence capacity of L. rhamnosus GG. Scanning electron microscopy confirmed that the pili, key surface factors for adherence to intestinal cells and mucus, were sheared off during the spray-drying process. These data thus highlight that both the functionality and viability of probiotics should be assessed during the spray-drying process and subsequent storage.     

Electric activity of the rectus femoris in supramaximal running

The electrical activity of the rectus femoris has been analysed in nine subjects during the course of a supramaximal run performed until exhaustion. The integrated electromyogram and the median frequency do not change significantly as a function of time.     

Proteomics analysis of cytokine-induced dysfunction and death in insulin-producing INS-1E cells: new insights into the pathways involved

Cytokines released by islet-infiltrating immune cells play a crucial role in beta-cell dysfunction and apoptotic cell death in the pathogenesis of type 1 diabetes and after islet transplantation. RNA studies revealed complex pathways of genes being activated or suppressed during this beta-cell attack. The aim of the present study was to analyze protein changes in insulin-producing INS-1E cells exposed to inflammatory cytokines in vitro using two-dimensional DIGE. Within two different pH ranges we observed 2214 +/- 164 (pH 4-7) and 1641 +/- 73 (pH 6-9) spots. Analysis at three different time points (1, 4, and 24 h of cytokine exposure) revealed that the major changes were taking place only after 24 h. At this time point 158 proteins were altered in expression (4.1%, n = 4, p < or = 0.01) by a combination of interleukin-1beta and interferon-gamma, whereas only 42 and 23 proteins were altered by either of the cytokines alone, giving rise to 199 distinct differentially expressed spots. Identification of 141 of these by MALDI-TOF/TOF revealed proteins playing a role in insulin secretion, cytoskeleton organization, and protein and RNA metabolism as well as proteins associated with endoplasmic reticulum and oxidative stress/defense. We investigated the interactions of these proteins and discovered a significant interaction network (p < 1.27e-05) containing 42 of the identified proteins. This network analysis suggests that proteins of different pathways act coordinately in a beta-cell dysfunction/apoptotic beta-cell death interactome. In addition the data suggest a central role for chaperones and proteins playing a role in RNA metabolism. As many of these identified proteins are regulated at the protein level or undergo post-translational modifications, a proteomics approach, as performed in this study, is required to provide adequate insight into the mechanisms leading to beta-cell dysfunction and apoptosis. The present findings may open new avenues for the understanding and prevention of beta-cell loss in type 1 diabetes.     

Low dose insemination in cattle with the Ghent device

A new artificial insemination device for semen deposition near the uterotubal junction (UTJ) in cattle (Ghent device) was developed at Ghent University (Belgium). In this study, UTJ insemination of dairy cows with the Ghent device was compared with the conventional insemination technique to evaluate the effect on pregnancy rates after insemination with different doses of semen. In each of three field trials, the cows (n=795, 659, 360) and heifers (n=253, 182, 231) were randomly assigned to receive 12 million sperm deposited in the uterine body using conventional techniques (control) or a reduced sperm dose (RSD) deposited in the same manner as the control or bilateral deposition near the uterotubal junction using the Ghent device (Ghent). Sperm dosages for RSD and Ghent inseminations were 8, 4, and 2 million sperm for field trials 1-3, respectively. Colonization and transmission of methicillin-resistant Staphylococcus aureus ST398 in nursery piglets

A transmission experiment was performed to evaluate the spread of methicillin-resistant Staphylococcus aureus (MRSA) ST398 in nursery piglets. Reproduction ratios (R(0)) in three experimental groups were found to vary between 3.92 and 52.54, indicating that after introduction, MRSA ST398 will spread easily among weaned piglets, with a tendency to become established.
