We previously reported on a new boiling stable protein isolated from aspen plants (Populus tremula), which we named SP1. SP1 is a stress-related protein with no significant sequence homology to other stress-related proteins. It is a 108-amino-acid hydrophilic polypeptide with a molecular mass of 12.4 kDa (Wang, W. X., Pelah, D., Alergand, T., Shoseyov, O., and Altman, A. (2002) Plant Physiol. 130, 865-875) and is found in an oligomeric form. Preliminary electron microscopy studies and matrix-assisted laser desorption ionization time-of-flight mass spectrometry experiments showed that SP1 is a dodecamer composed of two stacking hexamers. We performed a SDS-PAGE analysis, a differential scanning calorimetric study, and crystal structure determination to further characterize SP1. SDS-PAGE indicated a spontaneous assembly of SP1 to one stable oligomeric form, a dodecamer. Differential scanning calorimetric showed that SP1 has high thermostability i.e. Tm of 107 degrees C (at pH 7.8). The crystal structure of SP1 was initially determined to 2.4 A resolution by multi-wavelength anomalous dispersion method from a crystal belonging to the space group I422. The phases were extended to 1.8 A resolution using data from a different crystal form (P21). The final refined molecule includes 106 of the 108 residues and 132 water molecules (on average for each chain). The R-free is 20.1%. The crystal structure indicated that the SP1 molecule has a ferredoxin-like fold. Strong interactions between each two molecules create a stable dimer. Six dimers associate to form a ring-like-shaped dodecamer strongly resembling the particle visualized in the electron microscopy studies. No structural similarity was found between the crystal structure of SP1 and the crystal structure of other stress-related proteins such as small heat shock proteins, whose structure has been already determined. This structural study further supports our previous report that SP1 may represent a new family of stress-related proteins with high thermostability and oligomerization.  相似文献

The association between HLA alleles (A, B, DRB1), haplotypes and AIDS progression in HIV-1 infected patients was investigated by analyzing and comparing the differences gene frequencies of HLA alleles (A, B, DRB1) and haplotypes in HIV-1 infected and AIDS individuals in Hubei province of China. Four hundred and twenty- four HIV-1 seropositive individuals were divided into two groups: HIV-1 infected group and AIDS patient group, according to diagnostic criteria. HLA-A, B, and DRB1 allele typing was performed using polymerase chain reaction-sequence-specific oligonucleotide (PCR-SSOP) and polymerase chain reaction-sequencing based typing (PCR-SBT) techniques. Our study revealed that B*57:01 seemed resistant to AIDS progression, and the presence of DRB1*04:05 was associated with a poor disease outcome in HIV-1 infection. These associations were independent of age, sex, and transmission route of the host. No association was observed between HLA-A, B, DRB1 homozygotes, HLA-Bw4, Bw6 serological types and AIDS progression. We concluded that HLA gene polymorphism has a significant role in HIV-1 infection/AIDS progression. This observational study may open up avenues for precision medicine in the personalized prevention and treatment of AIDS.  相似文献

In this study, we investigated hepatitis C virus (HCV) molecular epidemiology and evolutionary dynamics. Both E1 and NS5B sequences were characterized in 379 of 433 patients in southern China and classified into five major subtypes: 1b in 256 patients, 6a in 67 patients, 2a in 29 patients, 3a in 14 patients, and 3b in 13 patients. Using the E1 sequences obtained, along with those from other studies using samples from China, we inferred the HCV epidemic history by means of coalescence strategies. Five Bayesian skyline plots (BSPs) were estimated for the five subtypes. They concurrently highlighted the rapid growth in the HCV-infected population size from 1993 to 2000, followed by an abrupt slowing. Although flanked on both sides by variable population sizes, the plots showed distinct patterns of rapid HCV growth. Coincidently, 1993 to 2000 was a period when contaminated blood transfusions were common in China due to a procedural error in an officially encouraged plasma campaign. The abrupt slowing in 1998 to 2000 corresponded to the central government outlawing paid blood donations in 1998. Using a parametric model, the HCV population growth rates were estimated during 1993 to 2000. It was revealed that the 6a rate was the highest, followed by those of 1b, 2a, 3b, and 3a. Because these rates differed significantly (P < 1e−9) from each other, they may help explain why 6a is increasingly prevalent in southern China and 1b is predominant nationwide. These rates are approximately 10-fold higher than those reported elsewhere. These findings suggested that during the plasma campaign, certain barriers to efficient viral transmission were removed, allowing wide HCV dissemination.  相似文献

Background

Recently in China, HCV 6a infection has shown a fast increase among patients and blood donors, possibly due to IDU linked transmission.

Methodology/Findings

We recruited 210 drug users in Shanwei city, Guangdong province. Among them, HCV RNA was detected in 150 (71.4%), both E1 and NS5B genes were sequenced in 136, and 6a genotyped in 70. Of the 6a sequences, most were grouped into three clusters while 23% represent emerging strains. For coalescent analysis, additional 6a sequences were determined among 21 blood donors from Vietnam, 22 donors from 12 provinces of China, and 36 IDUs from Liuzhou City in Guangxi Province. Phylogeographic analyses indicated that Vietnam could be the origin of 6a in China. The Guangxi Province, which borders Vietnam, could be the first region to accept 6a for circulation. Migration from Yunnan, which also borders Vietnam, might be equally important, but it was only detected among IDUs in limited regions. From Guangxi, 6a could have further spread to Guangdong, Yunnan, Hainan, and Hubei provinces. However, evidence showed that only in Guangdong has 6a become a local epidemic, making Guangdong the second source region to disseminate 6a to the other 12 provinces. With a rate of 2.737×10−3 (95% CI: 1.792×10−3 to 3.745×10−3), a Bayesian Skyline Plot was portrayed. It revealed an exponential 6a growth during 1994–1998, while before and after 1994–1998 slow 6a growths were maintained. Concurrently, 1994–1998 corresponded to a period when contaminated blood transfusion was common, which caused many people being infected with HIV and HCV, until the Chinese government outlawed the use of paid blood donations in 1998.

Conclusions/Significance

With an origin from Vietnam, 6a has become a local epidemic in Guangdong Province, where an increasing prevalence has subsequently led to 6a spread to many other regions of China.  相似文献

Proteins inducible by dehydration and abscisic acid (ABA), have been identified in a number of species and have been suggested to play a role in desiccation tolerance. Recently, we identified a novel boiling-stable protein (BspA) which accumulated in shoots of aspen ( Populus tremula L.) cultured in vitro, in response to gradual water stress and ABA application (Pelah et al. 1995. Tree Physiol. 15: 673–678.). Accumulation of BspA, and of the water stress-related protein dehydrin dsp- 16 and sucrose synthase from the resurrection plant. Craterostigma plantagineum , was examined in two greenhouse-grown Populus species to investigate the relationship between the presence of the proteins and water stress tolerance. Detached leaves of Populus tomentosa lost more water than Populus popularis , resulting in a significant decrease in leaf water potential. Using electrolyte leakage analysis, it was found that detached leaves of Populus popularis are more tolerant to water stress than those of Populus tomentosa . Using western blots with the corresponding antibodies, we have found in Populus popularis accumulation of BspA and sucrose synthase due to water stress, and the constitutive presence of a dehydrin-like protein. In contrast, a low expression of BspA was found in Populus tomentosa , but not of sucrose synthase and dehydrin-like proteins. Desiccation tolerance in many tissues can be partly attributed to soluble sugars. Analysis of the amount of soluble sugars did not reveal clear-cut differences between the two species, except for significant sucrose accumulation and glucose reduction in water-stressed Populus tomentosa and increase in glucose in water-stressed Populus popularis . The data obtained points to a positive correlation between increased water stress tolerance of one poplar species as compared with another and accumulation of water stress-related proteins and sucrose synthase.  相似文献

In recent years, Populus species have acquired an important place in basic and applied research of woody plants. The practical role of Populus species in world forestry and their importance to research as a woody-plant model have led to increasing interest in tissue-culture and molecular techniques, as well as the development of transformation procedures for this genus. A simple technical procedure is described here step-by-step, for the first time, as a routine method for transforming Populus tremula using a disarmed Agrobacterium tumefaciens hypervirulent strain. The procedure begins with the inoculation of stem explants with bacterial suspension, followed by a short period of co-cultivation on a highly regenerative medium. Transformed shoots are selected on regeneration medium containing antibiotics and the presence of the inserted target genes is checked using a rapid and efficient PCR test. Selected shoots are transferred to a rooting medium, under the same selection pressure, and propagated via stem cuttings. Selected plants can be hardened and transferred to the green-house within 4 months of inoculation. The method has proven efficient for several gene constructs, selection on Kan or Hyg, and three different Agrobacterium strains.  相似文献
