Production ofpotato spindle tuber viroid (PSTV) by large scale fermentationof PSTV–infected potato cell suspension cultures.—J.exp. Bot 36: 1985–1995. Cell suspension cultures of Solatiumdemissum, infected with the potato spindle tuber viroid (PSTV),were scaled up to volumes of up to 800 dm3 to provide sufficientand uniform plant material for subsequent studies on viroidbiosynthesis. Here we describe the technological aspects ofproducing the required amounts of biomass and viroid. The cells,which had been maintained on a medium containing expensive coconutmilk, were first adapted to rapid growth on the less expensiveB5–medium. The physiological state of the cells was monitoredby in vivo 31P–NMR spectroscopy Under the chosen conditionsthe scale–up from 10 dm3 inoculum from shake flasks tothe harvest of the 800 dm3 stirred fermenter lasted 38 d andprovided 112 kg biomass. Growth characteristics and viroid productionin shake flasks and large bioreactors were rather similar. Gelelectrophoretic analysis of isolated nucleic acids using silverstaining and Northern blot hybridization revealed a PSTV–contentof approximately 700 µg PSTV per kg fresh mass of culturedcells. The colour patterns of Heliconius butterflies are built up from an array of serially homologous pattern elements known as the nymphalid groundplan. An analysis of the phenotypic effects of ten genetic loci from H. melpomene and H. cydno reveals that each alters the expression either of a single element of the groundplan or of an entire row of serially homologous elements. Five of the ten loci affect the size (or presence/absence) of specific pattern elements, two affect the colour in which a pattern element is expressed, two affect pattern-inducing activity of the wing veins, and one appears to affect an overall threshold for pattern determination. Three of the ten loci have identical effects on homologues of the fore- and hindwing. We show that most of the apparently large and qualitative phenotypic effects of these genes can be readily explained by relatively small and quantitative changes in the dimensions or positions of specific pattern elements.  相似文献

Abstract. The application of molecular approaches such as mutant analysis and recombinant DNA technology, in conjunction with immunology, are set to revolutionize our understanding of the nitrate assimilation pathway. Mutant analysis has already led to the identification of genetic loci encoding a functional nitrate reduction step and is expected to lead ultimately to the identification of genes encoding nitrate uptake and nitrite reduction. Of particular significance would be identification of genes whose products contribute to regulatory networks controlling nitrogen metabolism. Recombinant DNA techniques are particularly powerful and have already allowed the molecular cloning of the genes encoding the apoprotein of nitrate reductase and nitrite reductase. These successes allow for the first lime the possibility to study directly the role of environmental factors such as type of nitrogen source (NO3 or NH4+) available to the plant, light, temperature water potential and CO2 and O2 tensions on nitrate assimilation gene expression and its regulation at the molecular level. This is an important advance since our current understanding of the regulation of nitrate assimilation is based largely on changes of activity of the component steps. The availability of mutants, cloned genes, and gene transfer systems will permit attempts to manipulate the nitrate assimilation pathway.  相似文献

The colour patterns of Heliconius butterflies are composed from a relatively simple set of pattern elements whose homologues are recognizable throughout the genus. Although Heliconius colour patterns look quite different from those of most nymphalids, these pattern elements are seen to derive from the generalized nymphalid groundplan. The differences arise primarily from the loss or positional shift of certain pattern elements, a high degree of fusion between individual pattern elements, and, in the forewing, asymmetries of the pattern elements relative to the wing-cell midline. The scheme of homologies we present is consistent with what is currently known about the comparative morphology and developmental physiology of colour pattern formation in Lepidoptera, and provides a framework for the interpretation of developmental, evolutionary and genetic studies in Heliconius.  相似文献

The tempo and mode of morphological evolution are influencedby several factors, among which evolutionary transformationsin developmental processes are likely to be important. Comparingthe embryos of extant species in an explicit phylogenetic framwork allows the estimation of minimum average rates of evolutionin quantitative developmental parameters. It also allows delineationof the maximum time that complex qualitative transformationsin developmental mechanism take to evolve. This paper analyzesrates of quantitative and qualitative developmental evolutionusing examples drawn primarily from echinoderms. The resultsdemonstrate that rates of developmental evolution can be comparableto rates of morphological evolution. There is no indicationthat rates of evolution in development are lower for earlierstages, contrary to the prediction of "tree" models of epigeneticinteractions. In particular, rates of evolution in oogenesiscan exceed rates of evolution in adult body size. Rates of developmentalevolution can vary by up to two orders of magnitude within aclade. Whether such large scale variation in evolutionary ratesof developmental processes is a general phenomenon can onlybe answered by further study.  相似文献

Sewage sludge was monitored for the presence of salmonellas after it had been dumped on a pasture so that it formed a number of extensive, thick pats. Although salmonellas were isolated from the sewage sludge, and also from grass and soil beneath the sludge for up to 94 days, they were not isolated from the faeces of grazing horses or surrounding drainage ditches. The results are discussed with regard to the possible hazards resulting from the use of sewage sludge on pasture.  相似文献

Ninety-three papers on home-range analysis using radio-tracking data were reviewed; these papers were found in a literature search of 18 of the major journals likely to include such papers, published in the 5-year period to the end of 1988. The review showed that even 25 years after the first radio-tracking studies, in the majority of papers there was still insufficient attention given to accurate and sufficient data collection, and to using appropriate and sophisticated analytical techniques to assess home-range size and configuration. This paper is designed to help people undertaking a radio-tracking study to avoid some of the most common pitfalls. It is based on some of the problems we have experienced studying several species of larger mammals. We use our collective experience to produce a guide on how to plan a radio-tracking study, to highlight some of the potential problems in designing the study and collecting the data, and to identify some of the difficulties that may be encountered during the analytical stages. The advantages and disadvantages of the most frequently used methods of home-range analysis are discussed and methods for determining the minimum number of radio-fixes and techniques for adjusting inadequate sample sizes are described, as are the problems that may be caused by autocorrelated data.  相似文献
