The effect of ionophore antibiotics, valinomycin and nigericin, on the generation of the membrane potential, the pH gradient and the efficacy of phage infection in tetracycline-resistant staphylococci has been studied. Valinomycin at a concentration of 0.5 microM induces the dissipation of the membrane potential, and nigericin at a concentration of 12.0 microM decreases the value of the pH gradient on the membrane of staphylococci. The separate use of antibiotics has no essential influence on the efficacy of phage infection. The combined use of valinomycin and nigerimycin produces the maximum inhibition of phage infection (64.5%) at the stage of the introduction of DNA into the bacterial cell, which is indicative of a definite role played by the membrane potential and the pH gradient in the transport of phage DNA into staphylococcal cells.  相似文献

The generation of transmembrane difference of electrochemical potentials was registered on the intact cells and ultrasonication-obtained membrane vesicles of Staphylococcus aureus with the application of transmembrane electrophoresis of permeant anions, potassium transport in the presence of valinomycin and 8-anilinonaphthalene-1-sulphonate fluorescence. The membrane potential is formed when the chain of electron transfer or H+-ATPase functions or when the pH gradient varies (the nonenzymic pathway). M-chlorinecarbonylcyanidephenylhydrazonium, a protonophore uncoupler potassium cyanide, an inhibitor of the respiratory chain, N',N-dicyclohexylcarbodiimide, an inhibitor of ATPase, cause the membrane potential dissipation. The orientation of the transmembrane electric field is as follows: "minus" inside cells and "plus" inside membrane vesicles.  相似文献

Rhesus monkeys have been kept in horizontal position under klinostatic or antiorthostatic hypokinetic conditions for 7 and 19 days. Using scanning electron microscope, studies were made of the otolithic membrane of the utricle, the receptor surface of the utricle, crista ampullaris of the lateral semicircular canals, the organ of Corti, the stria vascularis and spiral ligament. Data are presented for a subset of lymphocytes characterized by FISH as missing painted chromosomal material. These lymphocytes occur in both control and irradiated subjects. These cells have a much greater frequency of one-way translocations than cells in which all of the painted chromosomal material is present. Their presence contributes to interindividual variability in control translocation yields. These cells do not appear to be more prevalent in persons exposed to high radiation doses. It is suggested that their exclusion when selecting cells for analysis may improve the sensitivity of FISH as a biological dosimeter at low doses. Mechanisms for the production of these one-way translocations in vivo are also discussed, with a proposal that their variable frequency in individuals may be consistent with exposure to chemical clastogens.  相似文献

A specimen of the blackfin flounder Glyptocephalus stelleri showing a reversal of the body sides was found for the first time. Records of flounders of the genus Pseudopleuronectes with abnormal body coloration are given. The occurrence of pigmentation on both sides of the body and the reversal of sides are indicative of the manifestation of atavism in flatfishes. The high variability of phenotypic characters, which is common to some species of flatfish with clinal variations, is illustrated using the example of the marbled flounder.  相似文献
