Here we examine 3157 foxes from 6 districts of the Slovak Republic in order to determine for the first time the distribution, prevalence and identity of Mesocestodes spp. endemic to this part of central Europe. During the period 2001-2006, an average of 41.9% of foxes were found to harbour Mesocestoides infections. Among the samples we confirmed the widespread and common occurrence of M. litteratus (Batsch, 1786), and report the presence, for the first time, of M. lineatus (Goeze, 1782) in the Slovak Republic, where it has a more restricted geographical range and low prevalence (7%). Using a combination of 12S rDNA, CO1 and ND1 mitochondrial gene sequences together with analysis of 13 morphometric characters, we show that the two species are genetically distinct and can be differentiated by discrete breaks in the ranges of the male and female reproductive characters, but not by the more commonly examined characters of the scolex and strobila. Estimates of interspecific divergence within Mesocestoides ranged from 9 to 18%, whereas intraspecific variation was less than 2%, and phylogenetic analyses of the data showed that despite overlapping geographical ranges, the two commonly reported European species are not closely related, with M. litteratus more closely allied to North American isolates of Mesocestoides than to M. lineatus. We confirm that morphological analysis of reproductive organs can be used to reliably discriminate between these often sympatric species obtained from red foxes.  相似文献

Our knowledge of temperate broadleaf forest ecology is based mostly on the study of production forests, which lack the terminal stage of forest development and have a simpler stand structure than old-growth and primeval forests. How primeval and production forests differ in net primary production (NPP) is not well known. In three primeval and three nearby production forests of European beech (Fagus sylvatica) in the Slovakian Carpathians, we measured aboveground biomass stocks (live and dead), aboveground NPP (ANPP) and parameters characterizing canopy structural diversity (leaf area index and its spatial variation). Our study aims were (1) to explore the role of canopy structural diversity for ANPP and (2) to assess evidence of a productivity decline in the terminal stage. While aboveground live biomass stocks were on average 20% greater in the primeval forests (386 vs. 320 Mg ha?1; insignificant difference at two sites), deadwood mass stocks were on average four times larger than in the production forests (86 vs. 19 Mg ha?1). ANPP was similarly high in the primeval and production forests (10.0 vs. 9.9 Mg ha?1 y?1) and did not decrease towards the terminal stage. Production models indicate that, in the primeval forests, about 10% of ANPP (ca. 1 Mg ha?1 y?1) was generated by effects related to leaf area heterogeneity, evidencing a positive effect of structural diversity on forest productivity, even though species diversity was low. This study helps to better understand the impact of forest management on the productivity and carbon storage in temperate woodlands.  相似文献

Tissue engineering (TE) of long tracheal segments is conceptually appealing for patients with inoperable tracheal pathology. In tracheal TE, stem cells isolated from bone marrow or adipose tissue have been employed, but the ideal cell source has yet to be determined. When considering the origin of stem cells, cells isolated from a source embryonically related to the trachea may be more similar. In this study, we investigated the feasibility of isolating progenitor cells from pleura and pericard as an alternative cells source for tracheal tissue engineering. Porcine progenitor cells were isolated from pleura, pericard, trachea and adipose tissue and expanded in culture. Isolated cells were characterized by PCR, RNA sequencing, differentiation assays and cell survival assays and were compared to trachea and adipose-derived progenitor cells. Progenitor-like cells were successfully isolated and expanded from pericard and pleura as indicated by gene expression and functional analyses. Gene expression analysis and RNA sequencing showed a stem cell signature indicating multipotency, albeit that subtle differences between different cell sources were visible. Functional analysis revealed that these cells were able to differentiate towards chondrogenic, osteogenic and adipogenic lineages. Isolation of progenitor cells from pericard and pleura with stem cell features is feasible. Although functional differences with adipose-derived stem cells were limited, based on their gene expression, pericard- and pleura-derived stem cells may represent a superior autologous cell source for cell seeding in tracheal tissue engineering.  相似文献

Background

The taxonomic distinctiveness of Ascaris lumbricoides and A. suum, two of the world''s most significant nematodes, still represents a much-debated scientific issue. Previous studies have described two different scenarios in transmission patterns, explained by two hypotheses: (1) separated host-specific transmission cycles in highly endemic regions, (2) a single pool of infection shared by humans and pigs in non-endemic regions. Recently, A. suum has been suggested as an important cause of human ascariasis in endemic areas such as China, where cross-infections and hybridization have also been reported. The main aims of the present study were to investigate the molecular epidemiology of human and pig Ascaris from non-endemic regions and, with reference to existing data, to infer the phylogenetic and phylogeographic relationships among the samples.

Methodology

151 Ascaris worms from pigs and humans were characterized using PCR-RFLP on nuclear ITS rDNA. Representative geographical sub-samples were also analysed by sequencing a portion of the mitochondrial cox1 gene, to infer the extent of variability at population level. Sequence data were compared to GenBank sequences from endemic and non-endemic regions.

Principal Findings

No fixed differences between human and pig Ascaris were evident, with the exception of the Slovak population, which displays significant genetic differentiation. The RFLP analysis confirmed pig as a source of human infection in non-endemic regions and as a corridor for the promulgation of hybrid genotypes. Epidemiology and host-affiliation seem not to be relevant in shaping molecular variance. Phylogenetic and phylogeographical analyses described a complex scenario, involving multiple hosts, sporadic contact between forms and an ancestral taxon referable to A. suum.

Conclusions/Significance

These results suggest the existence of homogenizing gene flow between the two taxa, which appear to be variants of a single polytypic species. This conclusion has implications on the systematics, transmission and control programs relating to ascariasis.  相似文献

A small, low-copy-number cryptic 5.5-kb plasmid, designated pEF47, was isolated from a rumen bacterium Enterococcus faecalis 47 and partially characterized. Comparisons by restriction and partial sequence analysis of pEF47 demonstrated high homology to pS86 plasmid, recently isolated from a human clinical strain E. faecalis S-86. PCR followed by sequence analysis and Southern hybridization showed that pS86/pEF47-related replicons are regularly encountered in plasmids from Gram-positive cocci.  相似文献

The impact of heating on the peristence of water repellency, saturated hydraulic conductivity, and water retention characteristics was examined on soils from both forest and meadow sites in southwest Slovakia shortly after a wet spell. The top 5 cm of meadow soils had an initial water drop penetration time WDPT at 20°C of 457 s, whereas WDPT in the pine forest was 315 s for the top 5 cm and 982 s if only the top 1 cm was measured. Heating soils at selected temperatures of 50, 100, 150, 200, 250 and 300°C caused a marked drop in water drop penetration time WDPT from the initial value at 20°C. However, samples collected in different years and following an imposed cycle of wetting and drying showed much different trends, with WDPT sometimes initially increasing with temperature, followed by a drop after 200–300°C. The impact of heating temperature on the saturated hydraulic conductivity of soil was small. It was found for both the drying and wetting branches of soil water retention curves that an increase in soil water repellency resulted in a drop in soil water content at the same matric potential. The persistence of soil water repellency was strongly influenced by both the sampling site and time of sampling, as it was characterized by the results of WDPT tests.  相似文献

The effect of the amendment with alginite, an organic rock originating from the biomass of fossilized unicellular algae, on microbial activity of forest soils was tested using a pot experiment. Five variants of soil-alginite mixtures were tested in three replicates with two forest soils: a loose sandy soil and a sandy loam. Gravimetric moisture closely correlated with the dose of alginite in both soils. Basal respiration and catalase activity increased with the dose of alginite in the sandy soil, but not in the sandy loam, where the highest response was observed at intermediate doses of alginite. The correlations of microbial activity parameters with moisture in the sandy soil were also much closer than in the sandy loam. The amendment with alginite was thus effective in improving some of the selected microbial activity indicators, but the optimum dose of alginite strongly depends on soil texture.  相似文献

? Premise of the study: Knowledge of functional leaf traits can provide important insights into the processes structuring plant communities. In the genus Sorbus, the generation of taxonomic novelty through reticulate evolution that gives rise to new microspecies is believed to be driven primarily by a series of interspecific hybridizations among closely related taxa. We tested hypotheses for dispersion of intermediacy across the leaf traits in Sorbus hybrids and for trait linkages with leaf area and specific leaf area. ? Methods: Here, we measured and compared the whole complex of growth, vascular, and ecophysiological leaf traits among parental (Sorbus aria, Sorbus aucuparia, Sorbus chamaemespilus) and natural hybrid (Sorbus montisalpae, Sorbus zuzanae) species growing under field conditions. A recently developed atomic force microscopy technique, PeakForce quantitative nanomechanical mapping, was used to characterize the topography of cell wall surfaces of tracheary elements and to map the reduced Young's modulus of elasticity. ? Key results: Intermediacy was associated predominantly with leaf growth traits, whereas vascular and ecophysiological traits were mainly parental-like and transgressive phenotypes. Larger-leaf species tended to have lower modulus of elasticity values for midrib tracheary element cell walls. Leaves with a biomass investment related to a higher specific leaf area had a lower density. Leaf area- and length-normalized theoretical hydraulic conductivity was related to leaf thickness. ? Conclusions: For the whole complex of examined leaf traits, hybrid microspecies were mosaics of parental-like, intermediate, and transgressive phenotypes. The high proportion of transgressive character expressions found in Sorbus hybrids implies that generation of extreme traits through transgressive segregation played a key role in the speciation process.  相似文献
