An anaerobic continuous culture study was made with Campylobacter spec. to determine growth yields under various growth conditions. The growth media contained 0.1% (w/v) yeast extract as carbon source. When grown in an aspartate-limited culture Y asp max was 4.6. Inclusion of formate in the culture medium hardly affected the true growth yield. The number of ATP equivalents generated in the fumaratereductase system was 0.66 and the Y ATP max was 7.0. In the nitrate reduction with formate 1.7 ATP equivalents were generated, and a YNO 3- max of 12.2 was observed. The true growth yield obtained with a mixture of lactate and aspartate was lower than that found with aspartate alone.  相似文献

Introduction

Ultrasonography (US) might have an added value to clinical examination in diagnosing early rheumatoid arthritis (RA) and assessing remission of RA. We aimed to clarify the added value of US in RA in these situations performing a systematic review.

Methods

A systematic literature search was performed for RA, US, diagnosis and remission. Methodological quality was assessed; the wide variability in the design of studies prohibited pooling of results.

Results

Six papers on the added value of US diagnosing early RA were found, in which at least bilateral metacarpophalangeal (MCP), wrists and metatarsophalangeal (MTP) joints were scanned. Compared to clinical examination, US was superior with regard to detecting synovitis and predicting progression to persistent arthritis or RA. Eleven papers on assessing remission were identified, in which at least the wrist and the MCP joints of the dominant hand were scanned. Often US detected inflammation in patients clinically in remission, irrespective of the remission criteria used. Power Doppler signs of synovitis predicted X-ray progression and future flare in patients clinically in remission.

Conclusions

US appears to have added value to clinical examination for diagnosing of RA when scanning at least MCP, wrist and MTP joints, and, when evaluating remission of RA, scanning at least wrist and MCP joints of the dominant hand. For both purposes primarily power Doppler US might be used since its results are less equivocal than those of greyscale US.  相似文献

Forest-to-rubber plantation conversion is an important land-use change in the tropical region, for which the impacts on soil carbon stocks have hardly been studied. In montane mainland southeast Asia, monoculture rubber plantations cover 1.5 million ha and the conversion from secondary forests to rubber plantations is predicted to cause a fourfold expansion by 2050. Our study, conducted in southern Yunnan province, China, aimed to quantify the changes in soil carbon stocks following the conversion from secondary forests to rubber plantations. We sampled 11 rubber plantations ranging in age from 5 to 46 years and seven secondary forest plots using a space-for-time substitution approach. We found that forest-to-rubber plantation conversion resulted in losses of soil carbon stocks by an average of 37.4±4.7 (SE) Mg C ha−1 in the entire 1.2-m depth over a time period of 46 years, which was equal to 19.3±2.7% of the initial soil carbon stocks in the secondary forests. This decline in soil carbon stocks was much larger than differences between published aboveground carbon stocks of rubber plantations and secondary forests, which range from a loss of 18 Mg C ha−1 to an increase of 8 Mg C ha−1. In the topsoil, carbon stocks declined exponentially with years since deforestation and reached a steady state at around 20 years. Although the IPCC tier 1 method assumes that soil carbon changes from forest-to-rubber plantation conversions are zero, our findings show that they need to be included to avoid errors in estimating overall ecosystem carbon fluxes.  相似文献

Aims

Our goal was to assess how management and sward functional diversity affect nitrogen response efficiency (NRE), the ratio of plant biomass production to supply of available nitrogen (N) in temperate grassland.

Methods

A three-factorial design was employed: three sward compositions, two mowing frequencies, and two fertilization treatments.

Results

NRE was largely influenced by fertilization followed by mowing frequency and sward composition. NRE was larger in unfertilized than fertilized plots, in plots cut thrice than plots cut once per year, and in control swards than in monocot- or dicot-enhanced swards. Fertilization decreased NRE through decreases in both N uptake efficiency (plant N uptake per supply of available N) and N use efficiency (NUE, biomass produced per plant N uptake) whereas mowing frequency and sward composition affected NRE through N uptake efficiency rather than NUE. The largest NRE in the control sward with 70 % monocots and 30 % dicots attests that these proportions of functional groups were best adapted in this grassland ecosystem.

Conclusions

Optimum NRE may not be a target of most farmers, but it is an appropriate tool to evaluate the consequences of grassland management practices, which farmers may employ to maximize profit, on environmental quality.  相似文献

During 2000, four cases of fatal Lassa fever were imported from Africa to Europe. In two patients, consecutive serum samples were available for monitoring of virus load and cytokine levels in addition to standard laboratory data. Both patients had non-specific early clinical symptoms including high fever. Patient 1 developed multi-organ failure and died of hemorrhagic shock on day 15 of illness, while patient 2 died of respiratory failure due to aspiration without hemorrhage on day 16. Ribavirin was administered to both patients beginning only on day 11. High serum aspartate aminotransferase and lactate dehydrogenase (LDH) levels were remarkable in both patients. Patient 1 had an initial virus load of 10(6) S RNA copies/ml as measured by real-time RT-PCR. Viremia increased steadily and reached a plateau of approximately 10(8)-10(9) copies/ml 4 days before death, while IFN-gamma and TNF-alpha rose to extremely high levels only shortly before death. In contrast, in patient 2 the virus load decreased from 10(7) to 10(6) copies/ml during the late stage of illness which was paralleled by a decrease in the IFN-gamma and TNF-alpha levels. The IL-10 level increased when specific IgM and IgG appeared. These data suggest that a high virus load and high levels of pro-inflammatory cytokines in the late stage of Lassa fever play an important role in the pathogenesis of hemorrhage, multi-organ failure, and shock in Lassa fever.  相似文献

New developments and expanding indications have resulted in a significant increase in the number of patients with pacemakers and internal cardioverterdefibrillators (ICDs). Because of its unique capabilities, magnetic resonance imaging (MRI) has become one of the most important imaging modalities for evaluation of the central nervous system, tumours, musculoskeletal disorders and some cardiovascular diseases. As a consequence of these developments, an increasing number of patients with implanted devices meet the standard indications for MRI examination. Due to the presence of potential life-threatening risks and interactions, however, pacemakers and ICDs are currently not approved by the Food and Drug Administration (FDA) for use in an MRI scanner. Despite these limitations and restrictions, a limited but still growing number of studies reporting on the effects and safety issues of MRI and implanted devices have been published. Because physicians will be increasingly confronted with the issue of MRI in patients with implanted devices, this overview is given. The effects of MRI on an implanted pacemaker and/or ICDs and vice versa are described and, based on the current literature, a strategy for safe performance of MRI in these patients is proposed. Recent studies suggest that the direction and magnitude of changes in soil organic carbon (soil C) pools following forest-to-pasture conversion in the tropics are dependent upon initial soil conditions and local factors (e.g. pre-conversion soil C content, soil texture, vegetation productivity, and management practices). The goal of this study was to understand how landscape-scale variation in soil-forming factors influenced the response of soil C pools to forest clearing and pasture establishment in northeastern Costa Rica. We measured soil C and its stable isotopic composition in 24 paired pasture and reference forest sites distributed over large gradients of edaphic characteristics and slope throughout a 1400 km2 region. We used the large difference in stable C isotopic signatures of C3 vegetation (rain forest) versus C4 vegetation (pasture grasses) as a tracer of soil C dynamics. Soil C pools to 30 cm depth ranged from 26% lower to 23% higher in pastures compared to paired forests. The presence of non-crystalline clays and percent slope explained between 27 and 37% of the variation in the direction and magnitude of the changes in soil C storage following pasture establishment. Stable carbon isotopes (13C) in the top soil (0–10 cm) showed a rapid incorporation of pasture-derived C following pasture establishment, but the vegetation in these pastures never became pure C4 communities. The amount of forest-derived soil C in pasture topsoils (0–10 cm) was negatively correlated to both pasture age and the concentrations of non-crystalline iron oxides. Together these results imply that site factors such as soil mineralogy are an important control over soil C storage and turnover in this region.  相似文献

Chemokines, like stromal cell-derived factor-1 (SDF1/CXCL12), are small secreted proteins that signal cells to migrate. Because SDF1 and its receptor CXCR4 play important roles in embryonic development, cancer metastasis, and HIV/AIDS, this chemokine signaling system is the subject of intense study. However, it is not known whether the monomeric or dimeric structure of SDF1 is responsible for signaling in vivo. Previous structural studies portrayed the SDF1 structure as either strictly monomeric in solution or dimeric when crystallized. Here, we report two-dimensional NMR, pulsed-field gradient diffusion and fluorescence polarization measurements at various SDF1 concentrations, solution conditions, and pH. These results demonstrate that SDF1 can form a dimeric structure in solution, but only at nonacidic pH when stabilizing counterions are present. Thus, while the previous NMR structural studies were performed under acidic conditions that strongly promote the monomeric state, crystallographic studies used nonacidic buffer conditions that included divalent anions shown here to promote dimerization. This pH-sensitive aggregation behavior is explained by a dense cluster of positively charged residues at the SDF1 dimer interface that includes a histidine side chain at its center. A heparin disaccharide shifts the SDF1 monomer-dimer equilibrium in the same manner as other stabilizing anions, suggesting that glycosaminoglycan binding may be coupled to SDF1 dimerization in vivo.  相似文献

Atmospheric nitrogen (N) deposition is rapidly increasing in tropical regions. We investigated how a decade of experimental N addition (125 kg N ha?1 year?1) to a seasonal lowland forest affected depth distribution and contents of soil nitrous oxide (N2O), carbon dioxide (CO2) and methane (CH4), as well as natural abundance isotopic signatures of N2O, nitrate (NO3 ?) and ammonium (NH4 +). In the control plots during dry season, we deduced limited N2O production by denitrification in the topsoil (0.05–0.40 m) as indicated by: ambient N2O concentrations and ambient 15N-N2O signatures, low water-filled pore space (35–60%), and similar 15N signatures of N2O and NO3 ?. In the subsoil (0.40–2.00 m), we detected evidence of N2O reduction to N2 during upward diffusion, indicating denitrification activity. During wet season, we found that N2O at 0.05–2.00 m was mainly produced by denitrification with substantial further reduction to N2, as indicated by: lighter 15N-N2O than 15N-NO3 ? throughout the profile, and increasing N2O concentrations with simultaneously decreasing 15N-N2O enrichment with depth. These interpretations were supported by an isotopomer map and by a positive correlation between 18O-N2O and 15N-N2O site preferences. Long-term N addition did not affect dry-season soil N2O-N contents, doubled wet-season soil N2O-N contents, did not affect 15N signatures of NO3 ?, and reduced wet-season 15N signatures of N2O compared to the control plots. These suggest that the increased NO3 ? concentrations have stimulated N2O production and decreased N2O-to-N2 reduction. Soil CO2-C contents did not differ between treatments, implying that N addition essentially did not influence soil C cycling. The pronounced seasonality in soil respiration was largely attributable to enhanced topsoil respiration as indicated by a wet-season increase in the topsoil CO2-C contents. The N-addition plots showed reduced dry-season soil CH4-C contents and threshold CH4 concentrations were reached at a shallower depth compared to the control plots, revealing an N-induced stimulation of methanotrophic activity. However, the net soil CH4 uptake rates remained similar between treatments possibly because diffusive CH4 supply from the atmosphere largely limited CH4 oxidation.  相似文献
