Fragmentation of wakefulness and sleep are expected outcomes of advanced aging. We hypothesize that wake neurons develop endoplasmic reticulum dyshomeostasis with aging, in parallel with impaired wakefulness. In this series of experiments, we sought to more fully characterize age-related changes in wakefulness and then, in relevant wake neuronal populations, explore functionality and endoplasmic reticulum homeostasis. We report that old mice show greater sleep/wake transitions in the active period with markedly shortened wake periods, shortened latencies to sleep, and less wake time in the subjective day in response to a novel social encounter. Consistent with sleep/wake instability and reduced social encounter wakefulness, orexinergic and noradrenergic wake neurons in aged mice show reduced c-fos response to wakefulness and endoplasmic reticulum dyshomeostasis with increased nuclear translocation of CHOP and GADD34. We have identified an age-related unfolded protein response injury to and dysfunction of wake neurons. It is anticipated that these changes contribute to sleep/wake fragmentation and cognitive impairment in aging.  相似文献

Encholirium is a Brazilian genus of Bromeliaceae that occurs exclusively in rocky landscapes. This work aimed to generate basic information for the conservation of three Encholirium species that are endemic to the rocky mountains of Cadeia do Espinhaço, employing population genetic analyses. E. pedicellatum and E. biflorum have only one very small population each, both occurring in unprotected, private land sites, being critically endangered. E. subsecundum is more widespread, and some of its populations dwell in protected areas. Five Random Amplified Polymorphic DNA (RAPD) primers generated approximately 60 polymorphic bands for each species. This technique demonstrated the presence of a single RAPD profile for every individual sampled (except for one clone found in E. biflorum). High levels of genetic variability were not expected, due to the clonal habit of the plants and small size of the populations. Populations of E. biflorum and E. pedicellatum presented, respectively, 16.06% (Φ st  = 0.16, p<0.001) and 8.44% (Φ st  = 0.08, p<0.001) of the total genetic diversity attributable to genetic differences among groups within the populations. In E. subsecundum, 14.52% (Φ st  = 0.15, p<0.001) of the total genetic diversity was found among populations. Estimates of the Shannon’s Diversity Index provided similar results. These results are valuable for the development of conservation strategies.  相似文献

Various factors affect spatial genetic structure in plant populations, including adult density and primary and secondary seed dispersal mechanisms. We evaluated pollen and seed dispersal distances and spatial genetic structure of Carapa guianensis Aublet. (Meliaceae) in occasionally inundated and terra firme forest environments that differed in tree densities and secondary seed dispersal agents. We used parentage analysis to obtain contemporary gene flow estimates and assessed the spatial genetic structure of adults and juveniles. Despite the higher density of adults (diameter at breast height ≥ 25 cm) and spatial aggregation in occasionally inundated forest, the average pollen dispersal distance was similar in both types of forest (195 ± 106 m in terra firme and 175 ± 87 m in occasionally inundated plots). Higher seed flow rates (36.7% of juveniles were from outside the plot) and distances (155 ± 84 m) were found in terra firme compared to the occasionally inundated plot (25.4% and 114 ± 69 m). There was a weak spatial genetic structure in juveniles and in terra firme adults. These results indicate that inundation may not have had a significant role in seed dispersal in the occasionally inundated plot, probably because of the higher levels of seedling mortality.  相似文献

Rabbit antisera evoked by purified pepsin-extracted group A streptococcal M proteins were screened for the presence of joint cross-reactive antibodies by indirect immunofluorescence using thin sections of mouse knee joints. Pep M1, M5, and M18 antisera contained antibodies that cross-reacted with chondrocytes, cartilage, and synovium. Immunofluorescence inhibition assays showed that some of the joint cross-reactive epitopes were shared among the three heterologous serotypes of M protein. The pep M5 joint cross-reactive epitopes were localized to three different synthetic peptides of the C-terminal region of pep M5. Immunoblot analyses showed that the M5 joint cross-reactive antibodies recognized two proteins of human synovium and cartilage of molecular mass 56 and 58 kDa. The cross-reactive antibodies binding to the 56-kDa protein were inhibited by purified vimentin in immunoblot inhibition experiments. M protein-specific antibodies from patients with acute rheumatic fever were also shown to cross-react with joint tissue in a pattern similar to the rabbit antisera. Rabbit and human M protein-specific antibodies that were bound to articular cartilage activated significant levels of complement when compared to control serum, suggesting that M protein joint cross-reactive antibodies could potentially be involved in the pathogenesis of ARF and arthritis.  相似文献

BACKGROUND AND AIMS: A knowledge of natural populations' breeding systems is important in order to implement in situ and ex situ management and conservation practices. Using microsatellite markers, three Oryza glumaepatula populations from Brazil were studied to determine the breeding system and genetic structure parameters of this species. METHODS: Each population represented by ten families with ten individuals per family was studied using eight microsatellite primers. Families of the Rio Xingu population (XI) were obtained from the greenhouse, whereas families from Rio Solimoes (SO) and Rio Paraguay (PG) were collected from the wild. Amplified products electrophoresed on non-denaturing polyacrylamide gels were visualized with a silver staining procedure. The mating system parameters were analysed based on the mixed mating model (software MLTR) while genetic structure analyses of the three populations and their families were performed using the FSTAT software. KEY RESULTS: The mean numbers of alleles per loci were 2.5, 3.9 and 2.5, respectively for the XI, PG and SO populations. Compared with their families, higher values for the observed heterozygosity and gene diversity were estimated for the parental populations. The subdivision (based on R(ST)) and inbreeding (F(IS)) in the SO and PG populations had similar effects, while inbreeding was the main effect in the families. Multilocus outcrossing rates varied from 0.011 to 0.223 in the three populations, indicating divergence in the outcrossing rates among O. glumaepatula populations. For the species (considering SO and PG populations together) an intermediate value was observed (tm = 0.116). Biparental inbreeding varied from 0.008 to 0.123, contributing to the selfing rate in these populations. More than 50 % of the outcrossing occurred between related individuals. CONCLUSIONS: The results indicated divergence in the mating system among O. glumaepatula populations, with consequences for conservation practices. The mating system of this species was classified as mixed with a predominance of self-fertilization.  相似文献
