During the last two centuries the average lifespan has increased at a rate of approximately 3 months/year in both sexes, hence oldest old people are becoming the population with the fastest growth in Western World. Although the average life expectancy is increasing dramatically, the healthy lifespan is not going at the same pace. This underscores the importance of studies on the prevention of age-related diseases, in order to satisfactorily decrease the medical, economic and social problems associated to advancing age, related to an increased number of individuals not autonomous and affected by invalidating pathologies. In particular, data from experimental studies in model organisms have consistently shown that nutrient signalling pathways are involved in longevity, affecting the prevalence of age-related loss of function, including age-related diseases. Accordingly, nutrigerontology is defined as the scientific discipline that studies the impact of nutrients, foods, macronutrient ratios, and diets on lifespan, ageing process, and age-related diseases. To discuss the potential relevance of this new science in the attainment of successful ageing and longevity, three original studies performed in Sicily with local foods and two reviews have been assembled in this series. Data clearly demonstrate the positive effects of nutraceuticals, functional foods and Mediterranean Diet on several biological parameters. In fact, they could represent a prevention for many age-related diseases, and, although not a solution for this social plague, at least a remedy to alleviate it. Thus, the possibility to create a dietary pattern, based on the combined strategy of the use of both nutraceuticals and functional foods should permit to create a new therapeutic strategy, based not only on a specific bioactive molecule or on a specific food but on a integrated approach that, starting from the local dietary habits, can be led to a “nutrafunctional diet” applicable worldwide. 相似文献

In recent years, there has been a growing interest, supported by a large number of experimental and epidemiological studies,
for the beneficial effects of some phenolic substances, contained in commonly used spices and herbs, in preventing various
age-related pathologic conditions, ranging from cancer to neurodegenerative diseases. Although the exact mechanisms by which
polyphenols promote these effects remain to be elucidated, several reports have shown their ability to stimulate a general
xenobiotic response in the target cells, activating multiple defense genes. Data from our and other laboratories have previously
demonstrated that curcumin, the yellow pigment of curry, strongly induces heme-oxygenase-1 (HO-1) expression and activity
in different brain cells via the activation of heterodimers of NF-E2-related factors 2 (Nrf2)/antioxidant responsive element (ARE) pathway. Many studies clearly demonstrate that activation ofNrf2 target genes,
and particularly HO-1, in astrocytes and neurons is strongly protective against inflammation, oxidative damage, and cell death.
In the central nervous system, the HO system has been reported to be very active, and its modulation seems to play a crucial
role in the pathogenesis of neurodegenerative disorders. Recent and unpublished data from our group revealed that low concentrations
of epigallocatechin-3-gallate, the major green tea catechin, induces HO-1 by ARE/Nrf2 pathway in hippocampal neurons, and
by this induction, it is able to protect neurons against different models of oxidative damages. Furthermore, we have demonstrated
that other phenolics, such as caffeic acid phenethyl ester and ethyl ferulate, are also able to protect neurons via HO-1 induction.
These studies identify a novel class of compounds that could be used for therapeutic purposes as preventive agents against

Background
Ageing affects many components of the immune system, including innate immune cells like monocytes. They are important in the early response to pathogens and for their role to differentiate into macrophages and dendritic cells. Recent studies have revealed significant age-related changes in genomic DNA methylation in peripheral blood mononuclear cells, however information on epigenetic changes in specific leukocyte subsets is still lacking. Here, we aimed to analyse DNA methylation in purified monocyte populations from young and elderly individuals.Findings
We analysed the methylation changes in monocytes purified from young and elderly individuals using the HumanMethylation450 BeadChip array. Interestingly, we found that among 26 differentially methylated CpG sites, the majority of sites were hypomethylated in elderly individuals. The most hypomethylated CpG sites were located in neuropilin 1 (NRP1; cg24892069) and neurexin 2 (NRXN2; cg27209729) genes, and upstream of miR-29b-2 gene (cg10501210). The age-related hypomethylation of these three sites was confirmed in a separate group of young and elderly individuals.Conclusions
We identified significant age-related hypomethylation in human purified monocytes at CpG sites within the regions of NRP1, NRXN2 and miR-29b-2 genes. 相似文献6.
ABSTRACT: This paper pays attention to the modifiable lifestyle factors such as diet and nutrition that might influence life extension and successful ageing. Previous data reported that in Sicily, the biggest Mediterranean island, there are some places where there is a high frequency of male centenarians with respect to the Italian average. The present data show that in Sicani Mountain zone there are more centenarians with respect to the Italian average. In fact, in five villages of Sicani Mountains, there were 19 people with an age range of 100-107?years old from a total population of 18,328 inhabitants. So, the centenarian number was 4.32-fold higher than the national average (10.37 vs. 2.4/10,000); the female/male ratio was 1.1:1 in the study area, while the national ratio is 4.54:1. Unequivocally, their nutritional assessment showed a high adherence to the Mediterranean nutritional profile with low glycemic index food consumed. To reach successful ageing it is advisable to follow a diet with low quantity of saturated fat and high amount of fruits and vegetables rich in phytochemicals. 相似文献

On March 19, 2008 a Symposium on Pathophysiology of Ageing and Age-Related diseases was held in Palermo, Italy. Here, the
lectures of M. Racchi on History and future perspectives of Alzheimer Biomarkers and of G. Scapagnini on Cellular Stress Response
and Brain Ageing are summarized. Alzheimer's disease (AD) is a heterogeneous and progressive neurodegenerative disease, which
in Western society mainly accounts for clinica dementia. AD prevention is an important goal of ongoing research. Two objectives
must be accomplished to make prevention feasible: i) individuals at high risk of AD need to be identified before the earliest
symptoms become evident, by which time extensive neurodegeneration has already occurred and intervention to prevent the disease
is likely to be less successful and ii) safe and effective interventions need to be developed that lead to a decrease in expression
of this pathology. On the whole, data here reviewed strongly suggest that the measurement of conformationally altered p53
in blood cells has a high ability to discriminate AD cases from normal ageing, Parkinson's disease and other dementias. On
the other hand, available data on the involvement of curcumin in restoring cellular homeostasis and rebalancing redox equilibrium,
suggest that curcumin might be a useful adjunct in the treatment of neurodegenerative illnesses characterized by inflammation,

During the past century, humans have gained more years of average life expectancy than in the last 10,000 years; we are now
living in a rapidly ageing world. The sharp rise in life expectancy, coupled to a steady decline in birth rates in all developed
countries, has led to an unprecedented demographic revolution characterized by an explosive growth in the number and proportion
of older people. Ageing is a complex process that negatively impacts the development of the immune system and its ability
to function. Progressive changes in the T and B cell systems over the life span have a major impact on the capacity to respond
to immune challenge. These cumulative age-associated changes in immune competence are termed Immunosenescence: some immunological
parameters are commonly notably reduced in the elderly and, reciprocally good function is tightly correlated to health status.
Hence, a better understanding of Immunosenescence and the development of new strategies to counteract it are essential for

Longitudinal studies are defining progressive alterations to the immune system associated with increased mortality in the
very elderly. Many of these changes are exacerbated by or even caused by chronic T cell stimulation by persistent antigen,
particularly from Cytomegalovirus. The composition of T cell subsets, their functional integrity and representation in the
repertoire are all markedly influenced by age and by CMV. How these findings relate to epidemiological, functional, genetic,
genomic and proteomic studies of human T cell immunosenescence was the subject of intense debate at an international conference
