Incubation of Swiss 3T3 or L929 cells with tumor necrosis factor (TNF) leads to the rapid stimulation of several cytosolic Ser/Thr kinases active toward myelin basic protein, the S6 peptide (RRLSSLR), the G peptide (SPQPSRRGSESSEE), and Kemptide (LRRASLG). This confirms the hypothesis that kinases other than protein kinases A and C may be involved in the TNF signal transduction. Chromatography on Mono Q resolved multiple kinase peaks with each substrate tested and moreover revealed a TNF-mediated casein kinase-2 activation in both cell lines, measurable with the specific RRREEESEEE peptide or with the G peptide. The TNF-stimulated myelin basic protein kinases-1 and -2 were identified as extracellular signal-regulated kinases-2 and -1, respectively, based on their elution pattern on Mono Q chromatography, their inactivation by protein phosphatase action, their reaction with phosphothreonine and phosphotyrosine antibodies, and by their migration on sodium dodecyl sulfate-polyacrylamide gel electrophoresis as 42- and 44-kDa proteins recognized by anti-extracellular signal-regulated kinase antibodies.  相似文献

Prevalence of coaggregation reactions among chicken lactobacilli   总被引:1,自引:0,他引:1  
L. VANDEVOORDE, H. CHRISTIAENS AND W. VERSTRAETE. 1992. Interbacterial adherence was frequently encountered among chicken lactobacilli. Fourteen of 45 combinations involving nine adhering strains were shown to be coaggregative. The coadherence mechanism was mediated by complementary heat- and sonication-sensitive cell surface structures. It was shown that intrageneric adherence enabled lactobacilli to maintain higher numbers in fed-batch reactors simulating the gastrointestinal tract. The mechanism of coaggregation can substantially increase the colonization potential of lactobacilli in environments with short residence times.  相似文献

Based on a series of physicochemical properties (differential scanning calorimetry, powder X-ray crystallographic studies and Fourier-transform infra red spectroscopic analysis) determined for N-stearoylethanolamine (NSEA) (C18:0) at different temperatures, evidence has been given that this compound can exist in (at least) three polymorphic forms. Powder X-ray crystallography clearly demonstrates the presence of three distinct molecular packings at distinct temperatures while spectral changes in the vibrational spectra reveal that the geometry of the CH(2)z.sbnd;CO functional group of the molecule is affected during the polymorphic transitions. Rationalization of the thermal physicochemical behavior of NSEA in terms of molecular packing is also proposed. It supposes rearrangement of the hydrocarbon chains upon heating of the molecule.  相似文献

Summary A solid state fermentation process was developed for the conversion of straw and cellulose under anaerobic conditions by a mixed culture of cellulolytic and methanogenic organisms. The bioconversion rate and efficiency were compared under mesophilic (35° C) and thermophilic (55° C) conditions. Cellulolytic activity was assayed in terms of sugar and overall soluble organic matter (chemical oxygen demand, COD) production. Maximum conversion rates were obtained under thermophilic conditions, i.e. 8.4 g and 14.2 g COD/kg·d, respectively, when wheat straw and cellulose were used as substrates. The cellulolytic activity of the reactor contents (23% dry matter), measured under substrate excess conditions, amounted to 50 g COD/kg·d. As a comparison, the activity of rumen contents (15% dry matter) measured by the same assay amounted to 150 g COD/kg·d. The anaerobic cellulases appeared to be substrate bound. This and the relative low activity levels attained, limit the perspectives of producing cellulase enzymes by this type of process.  相似文献

The various biological activities of tumor necrosis factor (TNF) are mediated by two receptors, one of 55 kD (TNF-R55) and one of 75 kD (TNF-R75). Although the phenotypic and molecular responses elicited by TNF in different cell types are fairly well characterized, the signaling pathways leading to them are so far only partly understood. To further unravel these processes, we focused on TNF-R55, which is responsible for mediating most of the known TNF effects. Since several studies have demonstrated the importance of receptor clustering and consequently of close association of the intracellular domains for signaling, we addressed the question of whether clustering of the intracellular domains of TNF-R55 (TNF-R55i) needs to occur in structural association with the inner side of the cell membrane, where many signaling mediators are known to reside. Therefore, we investigated whether induced intracellular clustering of only TNF-R55i would be sufficient to initiate and generate a full TNF response, without the need for a full-length receptor molecule or a transmembrane region. Our results provide clear evidence that inducible forced trimerization of either TNF-R55i or only the death domain elicits an efficient TNF response, comprising activation of the nuclear factor κB, induction of interleukin-6, and cell killing.  相似文献

Summary Growth of Rhodobacter capsulatus ATCC 23782 on silage filtrates under both axenic and non-axenic conditions was evaluated. Under both conditions, 96% assimilation of NH + 4 -N at rates of about 0.1 g/l·d was attained. Assimilation was complete when the C/N ratio was adjusted to ca. 5. Thus assimilation rates of 0.2 g/l·d were achieved for an acid-C and NH + 4 -N concentration of 2.0 and 0.4 g/l respectively. The corresponding biomass production amounted about 104 mg/g silage, dry matter.  相似文献

Little is known as to the structural requirements of the acyl side chain for interaction of acylglycerols with monoacylglycerol lipase (MAGL), the enzyme chiefly responsible for the metabolism of the endocannabinoid 2-arachidonoylglycerol (2-AG) in the brain. In the present study, a series of twelve analogues of 1-AG (the more stable regioisomer of 2-AG) were investigated with respect to their ability to inhibit the metabolism of 2-oleoylglycerol by cytosolic and membrane-bound MAGL. In addition, the ability of the compounds to inhibit the hydrolysis of anandamide by fatty acid amide hydrolase (FAAH) was investigated. For cytosolic MAGL, compounds with 20 carbon atoms in the acyl chain and 2-5 unsaturated bonds inhibited the hydrolysis of 2-oleoylglycerol with similar potencies (IC50 values in the range 5.1-8.2 microM), whereas the two compounds with a single unsaturated bond were less potent (IC50 values 19 and 21 microM). The fully saturated analogue 1-monoarachidin did not inhibit the enzyme, whereas the lower side chain analogues 1-monopalmitin and 1-monomyristin inhibited the enzyme with IC50 values of 12 and 32 microM, respectively. The 22-carbon chain analogue of 1-AG was also potent (IC50 value 4.5 microM). Introduction of an alpha-methyl group for the C20:4, C20:3, and C22:4 compounds did not affect potency in a consistent manner. For the FAAH and the membrane-bound MAGL, there was no obvious relationship between the degree of unsaturation of the acyl side chain and the ability to inhibit the enzymes. It is concluded that increasing the number of unsaturated bonds on the acyl side chain of 1-AG from 1 to 5 has little effect on the affinity of acylglycerols for cytosolic MAGL.  相似文献

Interleukin (IL)-6 gene induction was studied in response to treatment with Tumor Necrosis Factor (TNF) in the sensitive murine L929 cell line. Under conditions where TNF-mediated cytotoxicity was either increased or decreased, depending on addition of activators or inhibitors, we found that the TNF-induced IL6 gene expression was likewise enhanced or repressed. We conclude that the signal (or part of the signals) going to the nucleus and responsible for gene activation is conducted along the reaction mechanism leading to cellular toxicity.  相似文献
