Improving salinity tolerance in the most widely cultivated cereal, bread wheat (Triticum aestivum L.), is essential to increase grain yields on saline agricultural lands. A Portuguese landrace, Mocho de Espiga Branca accumulates up to sixfold greater leaf and sheath sodium (Na+) than two Australian cultivars, Gladius and Scout, under salt stress in hydroponics. Despite high leaf and sheath Na+ concentrations, Mocho de Espiga Branca maintained similar salinity tolerance compared to Gladius and Scout. A naturally occurring single nucleotide substitution was identified in the gene encoding a major Na+ transporter TaHKT1;5-D in Mocho de Espiga Branca, which resulted in a L190P amino acid residue variation. This variant prevents Mocho de Espiga Branca from retrieving Na+ from the root xylem leading to a high shoot Na+ concentration. The identification of the tissue-tolerant Mocho de Espiga Branca will accelerate the development of more elite salt-tolerant bread wheat cultivars. 相似文献

Elevated anthropogenic nitrogen (N) deposition could alter N status in temperate steppe. However, threshold observations of N status change from N limit to N saturation by far are not conclusive in these ecosystems. Research on the natural abundance of15N (δ15N) could greatly help provide integrated information about ecosystem N status. The goal of this study was to investigate the suitability of measurements of δ15N of major ecosystem N pools and several key species, plant15N fractionation, together with key vegetation and soil indicators in response to N fertilization as a tool to identify the N status in a temperate steppe in Inner Mongolia. 相似文献

Apurinic/apyrimidinic endonuclease (APE), an essential DNA repair enzyme, initiates the base excision repair pathway by creating a nick 5' to an abasic site in double-stranded DNA. Although the Chinese hamster ovary cells remain an important model for DNA repair studies, the Chinese hamster APE (chAPE1) has not been studied in vitro in respect to its kinetic characteristics. Here we report the results of a kinetic study performed on cloned and overexpressed enzyme in sf9 cells. The kinetic parameters were fully compatible with the broad range of kinetic parameters reported for the human enzyme. However, the activity measures depended on the time point of the culture. We applied inductivity coupled plasma spectrometry to measure the phosphorylation level of chAPE1. Our data showed that a higher phosphorylation of chAPE1 in the expression host was correlated to a lower endonuclease activity. The phosphorylation of a higher activity batch of chAPE1 by casein kinase II decreased the endonuclease activity, and the dephosphorylation of chAPE1 by lambda phosphatase increased the endonuclease activity. The exonuclease activity of chAPE1 was not observed in our kinetic analysis. The results suggest that noticeable divergence in reported activity levels for the human APE1 endonuclease might be caused by unaccounted phosphorylation. Our data also demonstrate that only selected kinases and phosphatases exert regulatory effects on chAPE1 endonuclease activity, suggesting further that this regulatory mechanism may function in vivo to turn on and off the function of this important enzyme in different organisms. 相似文献

The purpose of our investigation was to clarify the effects of agriculture on the process of loss of litter at the periphery of a farmland. This study revealed the generation process of an ecologically unusual phenomenon that is observed around cropland in semi-arid regions. We hypothesized that the vegetation around a farmland cannot supply plant litter to the ground surface because the ecological structure has been changed by agricultural activities. The study was conducted at Xilingol steppe, Xilingol League, Inner Mongolia Autonomous Region, China. Four study lines were established from the edge of an arable field to the surrounding meadow and parallel to the wind direction during the strong wind season. Key measurement for each line was set at the border between the farmland and steppe. Four study sites were set at intervals along each line. Plant litter, soil particle size distribution, plant species composition, plant volume, and species diversity were investigated. Despite using the same mowing method at the meadows of all study sites, the litter at the only periphery of the farmland completely disappeared. Soil particle size distribution in steppe, which was adjacent to the farmland, was similar to that of the farmland. Plant community structure at the periphery of the farmland was different from that of the far side from the farmland. This implies that soil scattered from the farmland affected the species composition of the steppe. Consequently, the change in plant community structure induced litter loss because of mowing. We concluded that plant litter was lost near the farmland because of the combined effects of farming and mowing. The results support our hypothesis that the vegetation around a farmland cannot supply plant litter because the ecological structure has been changed by agricultural activities. 相似文献

Proteoglycans are dominant glycoconjugates located on the cell surface and in extracellular spaces and consist of a core protein with one or more glycosaminoglycan side chains linked covalently. Heparan sulfate (HS) belongs to the family of glycosaminoglycans. HS has been assigned a variety of physiological and pathological functions, such as cell-cell adhesion, cell-matrix adhesion, cell proliferation, motility and differentiation, lipoprotein metabolism, blood coagulation, inflammation, tissue regeneration, tumor progression and invasion, pathogenic infection by bacteria, protozoa, and viruses, through specific interaction with a wide array of proteins, ligands, receptors, and pathogens (Bernfield, M., Gotte, M., Park, P. W., Reizes, O., Fitzgerald, M. L., Lincecum, J., and Zako, M. (1999) Annu. Rev. Biochem. 68, 729-777). We have shown here for the first time that light induces changes in pineal HS fine structure and that occurrence of the rare 3-O sulfation catalyzed by HS 3-O-sulfotransferase (3-OST2) is predominantly restricted to daytime pineal glands. 相似文献

Circadian rhythms are endogenously generated by a central pacemaker and are synchronized to the environmental LD cycle. The rhythms can be resynchronized, or reentrained, after a shift of the LD cycle, as in traveling across time zones. The authors have performed high-resolution mapping of the pacemaker to analyze the reentrainment process using rat pineal melatonin onset (MT(on)) and melatonin offset (MT(off)) rhythms as markers. Following LD (12:12) delays of 3, 6, and 12 h, MT(on) was phase locked immediately, whereas MT(off) shifted rapidly during the initial 1 through 3 cycles. In all animals, the MT(off) shifted beyond their expected phase positions in the new LD cycle, which resulted in a transient expansion of melatonin secretion duration for several cycles. It took MT(off) only 1, 2, or 3 cycles to complete most of the required phase shifts after 3, 6, or 12 h of the LD cycle delays, respectively. However, the final stabilization of phase relationships of both MT(on) and MT(off) required at least 6 cycles for rats experiencing a 3-h LD delay and much longer for the rest. These results reaffirmed the notion that both onset and offset phases of melatonin rhythms are important markers for the pacemaker and demonstrated that the reentrainment of the central pacemaker to a delay shift of the LD cycle is a 3-step process: an immediate phase lock of onset and a rapid delay shift of offset rhythms, overshoot of the offset, and, finally, a slow adjustment of both onset and offset phases. This study represents the 1st detailed analysis of the pacemaker behavior during reentrainment using melatonin and supports the notion that the eventual adaptation of the circadian pacemaker to a new time zone is a time-consuming process. 相似文献

A cDNA encoding human nucleophosmin (protein B23) was obtained by screening a human placental cDNA library in lambda gtll first with monoclonal antibody to rat nucleophosmin and then with confirmed partial cDNA of human nucleophosmin as probes. The cDNA had 1311 bp with a coding sequence encoding a protein of 294 amino acids. The identity of the cDNA was confirmed by the presence of encoded amino acid sequences identical with those determined by sequencing pure rat nucleophosmin (a total of 138 amino acids). The most striking feature of the sequence is an acidic cluster located in the middle of the molecule. The cluster consists of 26 Asp/Glu and 1 Phe and Ala. Comparison of human nucleophosmin and Xenopus nucleolar protein NO38 shows 64.3% sequence identity. The N-terminal 130 amino acids of human nucleophosmin also bear 50% identity with that of Xenopus nucleoplasmin. Northern blot analysis of rat liver total RNA with a partial nucleophosmin cDNA as probe demonstrated a homogeneous mRNA band of about 1.6 kb. Similar observations were made in hypertrophic rat liver and Novikoff hepatoma. However, the quantity of nucleophosmin mRNA is 50- and 5-fold higher in Novikoff hepatoma and hypertrophic rat liver, respectively, when compared with normal rat liver. Dot blot analysis also showed a nucleophosmin mRNA ratio of 64:5:1 in the three types of rat liver. When the protein levels were compared with Western blot immunoassays, Novikoff hepatoma showed 20 times more nucleophosmin, while only about 5 times more nucleophosmin was observed in hypertrophic rat liver than in unstimulated normal liver. 相似文献
