Association of PKA with the AMPA receptor GluR1 subunit via the A kinase anchor protein AKAP150 is crucial for GluR1 phosphorylation. Mutating the AKAP150 gene to specifically prevent PKA binding reduced PKA within postsynaptic densities (>70%). It abolished hippocampal LTP in 7-12 but not 4-week-old mice. Inhibitors of PKA and of GluR2-lacking AMPA receptors blocked single tetanus LTP in hippocampal slices of 8 but not 4-week-old WT mice. Inhibitors of GluR2-lacking AMPA receptors also prevented LTP in 2 but not 3-week-old mice. Other studies demonstrate that GluR1 homomeric AMPA receptors are the main GluR2-lacking AMPA receptors in adult hippocampus and require PKA for their functional postsynaptic expression during potentiation. AKAP150-anchored PKA might thus critically contribute to LTP in adult hippocampus in part by phosphorylating GluR1 to foster postsynaptic accumulation of homomeric GluR1 AMPA receptors during initial LTP in 8-week-old mice. 相似文献

The neurodegenerative disorder spinocerebellar ataxia 12 (SCA12) is caused by CAG repeat expansion in the non-coding region of the PPP2R2B gene. PPP2R2B encodes Bbeta1 and Bbeta2, alternatively spliced and neuron-specific regulatory subunits of the protein phosphatase 2A (PP2A) holoenzyme. We show here that in PC12 cells and hippocampal neurons, cell stressors induced a rapid translocation of PP2A/Bbeta2 to mitochondria to promote apoptosis. Conversely, silencing of PP2A/Bbeta2 protected hippocampal neurons against free radical-mediated, excitotoxic, and ischemic insults. Evidence is accumulating that the mitochondrial fission/fusion equilibrium is an important determinant of cell survival. Accordingly, we found that Bbeta2 expression induces mitochondrial fragmentation, whereas Bbeta2 silencing or inhibition resulted in mitochondrial elongation. Based on epistasis experiments involving Bcl2 and core components of the mitochondrial fission machinery (Fis1 and dynamin-related protein 1), mitochondrial fragmentation occurs upstream of apoptosis and is both necessary and sufficient for hippocampal neuron death. Our data provide the first example of a proapoptotic phosphatase that predisposes to neuronal death by promoting mitochondrial division and point to a possible imbalance of the mitochondrial morphogenetic equilibrium in the pathogenesis of SCA12. 相似文献

It is recommended to use the capacity of pathogenic staphylococci to be lysed by polyvalent therapeutic staphylococcal bacteriophage in the capacity of an additional simple and accessible criterion of staphylococcus pathogenicity. Of 147 strains of the pathogenic plasmacoagulating staphylococci 101 were lysed by the phage and of 166 nonplasmocoagulating nonpathogenic strains--only 6. This test correlated with the other signs of staphylococcus (lecithinase and hemolytic activity). The simplicity and sufficient specificity of this test permits to use it in any practical laboratory. Polyvalent diagnostic phage can be used on the basis of therapeutic bacteriophage by its additional adaptation to the pathogenic strains of staphylococcus. 相似文献

The diagnostic arsenal for extracranial stenosis includes both ultrasound techniques, such as transcranial Doppler and duplex sonography, which are not only screening methods, but also are used at surgery, and high-tech neuroradiological techniques, such as spiral computed tomographic and magnetic resonance angiography, which can visualize structural changes in the vessel walls and qualitatively and functionally evaluate blood flow in the vascular bed. The purpose of this investigation was to determine the efficacy of non-invasive diagnostic methods, such as magnetic resonance angiography, including phase-contrast magnetic resonance angiography and spiral computed tomographic angiography, in the detection and comprehensive evaluation of stenoses and to compare the feasibilities of noninvasive procedures with the gold standard--selective angiography. 相似文献

Results are presented from the experimental studies and numerical simulations of the behavior of dust grains in the plasma
of an inductive RF discharge. The experiments were carried out with neon at a pressure of 25–500 Pa and with 1.87-μm melamine
formaldehyde grains. The discharge was excited by a ring inductor supplied from a generator operating at a 100-MHz frequency.
The effective dust-grain interaction potential used in numerical simulations involved the spatial dependence of the grain
charge on the plasma floating potential, grain-interaction anisotropy resulting from the focusing of the drift ion current
by the negatively charged grains, and specific features of the shielding of the dust grains by the plasma electrons and ions
recombining both in the plasma bulk and on the grain surface. The results of Monte Carlo simulations show that the dust grains
form specific filament structures observed experimentally in the plasma of an inductive electrodeless discharge.

Changes in the intracellular concentration of Ca2+ ([Ca2+]in) that occur during prolonged depolarization of the plasma membrane were studied in isolated neurons of the edible snailHelix pomatia, using the calcium-sensitive probe Fura-2. The dependence of the amplitude of the calcium response on the value of the depolarization in the presence of 5 mM caffeine, in contrast to that observed in a normal solution, practically disappeared. This fact indicates that caffeine promotes calcium-dependent release of Ca2+ from the intracellular depots, which is the determining factor in the increase in [Ca2+]in during depolarization. Processes of reduction of [Ca2+]in to the steady-state levels were described by one exponential function, and in the presence of caffeine they occurred twice as rapidly as in the normal solution. Such an acceleration of the kinetics of the relaxation of [Ca2+]in is evidently associated with an increase in the efficiency of the work of the calcium pump of the intracellular calcium depots, which might lead to a decrease in the steady-state of level of [Ca2+]in even below the level observed for the normal extracellular solution.A. The genome of the NDV strain Sterna/Astrakhan/2755/2001 isolated from a wild bird of the Volga River delta in 2001 was completely sequenced. The phylogenetic analysis of the strain investigated and other NDV strains clearly demonstrated that Sterna/Astrakhan/2755/ 2001 belonged to the lineage 5b. Comparative analysis of molecular genetic markers of pathogenicity of strain Sterna/Astrakhan/2755/ 2001 allows its velogenic character to be suggested. 相似文献
