Using two-dimensional polyacrylamide gel electrophoresis, the GroEL homologue ofBacillus subtilis was shown to be induced upon infection with Ø105clz, a clear plaque mutant of the temperate bacteriophage Ø105. Bacillus subtilis anaerobic respiration and fermentative growth capabilities were compared to two other facultative anaerobes, Bacillus licheniformis and Escherichia coli. In glycerol defined medium, B. subtilis grew with nitrate, but not nitrite or fumarate, while B. licheniformis grew with nitrate or fumarate, but not nitrite. Growth of E. coli occurred in glycerol defined medium with either nitrate, nitrite, or fumarate. In order to grow by fermentation, B. subtilis required both glucose and pyruvate, while B. licheniformis and E. coli were capable of using either glucose or pyruvate. 相似文献

The hypothesis was tested that upper limits to height growth in trees are the result of the increasing bending moment of trees as they grow in height. The increasing bending moment of tall trees demands increased radial growth at the expense of height growth to maintain mechanical stability. In this study, the bending moment of large lodgepole pine (Pinus contorta Dougl. Ex Loud. var. latifolia Engelm.) was reduced by tethering trees at 10 m height to counter the wind load. Average bending moment of tethered trees was reduced to 38% of control trees. Six years of tethering resulted in a 40% increase in height growth relative to the period before tethering. By contrast, control trees showed decreased height growth in the period after tethering treatment. Average radial growth along the bole, relative to height growth, was reduced in tethered trees. This strongly suggests that mechanical constraints play a crucial role in limiting the height growth of tall trees. Analysis of bending moment and basal area increment at both 10 m and 1.3 m showed that the amount of wood added to the stem was closely related to the bending moment produced at these heights, in both control and tethered trees. The tethering treatment also resulted in an increase in the proportion of latewood at the tethering height, relative to 1.3 m height. For untethered control trees, the ratio of bending stresses at 10 m versus 1.3 m height was close to 1 in both 1998 and 2003, suggesting a uniform stress distribution along the outer surface of the bole. 相似文献

Heat shock inBacillus subtilis may induce as many as 66 proteins after temperature upshift from 37° to 48°C. Four induced proteins were analyzed by microsequencing techniques. These were identified as the homologues for GroEL, DnaK, enolase, and glyceraldehyde-3-phosphate dehydrogenase (GAPDH), which are heat shock proteins in other systems. The identities of GroEL and DnaK were confirmed additionally by Western blot analysis. As a control, a protein whose synthesis was repressed approximately threefold by heat shock was identified by microsequencing as flagellin. 相似文献

We address the optical properties of dense disordered yet well-separated metal nanoparticle arrays produced by physical vapor deposition through anodized aluminum oxide membrane masks. Using variations in synthesis parameters, the particle diameters vary from 14 to 50 nm and average center separation from 45 to 112 nm. Ag nanoparticle arrays with no long-range periodicity exhibit apparently random formation of high-intensity depolarized regions relative to orientation of incident electric field. We analyze this behavior numerically using coupled dipole model and explain the contrast formation in recorded scattering images. 相似文献

An incompetent mutant (4S Com(-)) does not release competence-inducing factor (CF) into the culture medium and is not infected with TP-1C phage deoxyribonucleic acid (DNA) unless CF is added to the transfection assay. A TP-1C phage-resistant mutant (4S Ton-r) releases relatively large amounts of CF into the culture medium but is not infected with TP-1C phage DNA, even in the presence of CF. The production of CF by the wild type or Ton-r mutant and the ability of the wild type or Com(-) mutant to react with CF does not occur after these cultures have grown at 67 C for 1 hr or longer. A preliminary characterization of the CF is described. The autolytic enzyme or the temperate phage of the wild type and the Ton-r and Com(-) mutants do not have competence-inducing activity. 相似文献

The variable cyanide-sensitivity of the iron-containing alcohol dehydrogenase isoenzyme (ADH II) of the ethanol-producing
bacterium Zymomonas mobilis was studied. In aerobically grown permeabilized cells, cyanide caused gradual inhibition of ADH II, which was largely prevented
by externally added NADH. Cyanide-sensitivity of ADH II was highest in cells grown under conditions of vigorous aeration,
in which intracellular NADH concentration was low. Anaerobically grown bacteria, as well as those cultivated aerobically in
the presence of cyanide, maintained higher intracellular NADH levels along with a more cyanide-resistant ADH II. It was demonstrated
that cyanide acted as a competitive inhibitor of ADH II, competing with nicotinamide nucleotides. NADH increased both cyanide-resistance
and oxygen-resistance of ADH II. 相似文献
