Selected commercial and technical grade pesticides were tested against the egg, preparasite and adult stages of Agamermis unka , a nematode parasite of the brown planthopper, Nilaparvata lugens . The commercial insecticide, diazinon (LC = 0.37 ppm), was most toxic to the 50 preparasites, followed by phenthoate (LC = 0.43 ppm), BPMC (LC = 0.44 ppm), IBP 50 50 (LC = 0.46 ppm), cartap hydrochloride (LC = 0.82 ppm) and buprofezin + isoprocarb 50 50 (LC = 1.11 ppm). The least toxic commercial pesticide tested was the fungicide, pencycuron 50 (LC = 2.19 ppm). Out of 12 technical grade insecticides tested, phenthoate, monocrotophos, 50 diazinon and carbofuran (LC = 0.37-0.46 ppm) were highly toxic to the preparasites, followed by 50 buprofezin, BPMC and fenitrothion (LC = 0.74-0.86 ppm). Fenthion, etofenprox, chlorpyrifos, 50 imidacloprid and MIPC (LC = 1.11-2.19 ppm) were the technical grade insecticides least toxic 50 to the preparasites. Most preparasites survived for up to 24 h at the low insecticide concentrations (0.63 and 0.31 ppm). Preparasites that were exposed to BPMC for 24 h at concentrations as high as 5.0 ppm and survived the treatments infected brown planthopper nymphs. Four selected insecticides-chlorpyrifos, BPMC, imidacloprid and carbofuran-had significant adverse effects on A. unka egg hatching. Eggs that were in the insecticide solution for 168 h fared poorly with imidacloprid having the best survival ( > 2% of the eggs hatching at 0.04 ppm). No eggs hatched from the other insecticide treatments. Three selected insecticides, BPMC, imidacloprid and chlorpyrifos, tested against adult A. unka showed that most adults survived the exposure to the insecticides between 0.31 and 2.5 ppm. At 5.0 ppm of BPMC or chlorpyrifos none of the adults survived, whereas with imidacloprid 70% of the adults survived. Egg deposition by the surviving adults was greatly reduced in those treated with the insecticides compared with those in the controls. Imidacloprid had some negative impact on the preparasites' ability to infect BPH nymphs, but it had the least detrimental effect of the insecticides tested on preparasite survival and on the eggs and adults of A. unka .  相似文献

High mountain ranges within and around Anatolia have played an important role in the biogeography of mountainous organisms, which are threatened by climate change. Using the Psorodonotus caucasicus species group (PCG), whose range is the Caucasus, Anatolia and the Balkans, as a model group, this study aims to determine the importance of Anatolian mountain ranges and to estimate possible future effects of global warming on such lineages by establishing an analogy between interglacial dynamics and global warming. Twenty populations, representing all extant species of the PCG, were studied. Sequences of four mitochondrial loci and three nuclear loci were obtained and used in phylogenetic, time estimation and phylogeographic analyses of PCG members. Additionally, ecological modelling analyses were conducted to estimate the range of PCG members at present and during the last glacial maximum and the last interglacial, with the aim of determining the roles of altitudes and glacial cycles in shaping the distribution of the PCG. Phylogenetic analyses supported the monophyly of the PCG as a whole and each individual species with the exception of P. suphani. The radiation of the PCG started approximately 3 Ma, according to beast . Species distribution models suggested a wider range during the last glacial maximum than during the last interglacial and at present, but these models also suggested that present conditions represent a substantial range loss. The PCG dispersed to the Balkans through two independent corridors from the Caucasus. The following conclusions were reached based on the results: (i) the uplift of the Pontic and East Anatolian Blocks during the mid‐Pliocene and the closing of the Mediterranean and the Paratethys connection in the Plio–Pleistocene and the mid‐Pleistocene Transition are the main events that triggered the radiation of the PCG; (ii) the PCG radiated from a Caucasian origin and invaded the Balkans twice, first through Northern Anatolia and second via the ‘Taurus Way’ dispersal corridor; (iii) lowlands are a barrier to the dispersal of PCG members; (iv) the last four intense glacial periods had little effect on the radiation of the PCG; (v) the range of the PCG has receded substantially from its original southern margin, and P. salmani and P. anatolicus have become extinct since the 1950s, possibly because of global warming; and (vi) automatic species delimitation tests supported species status of the morphospecies P. caucasicus, P. macedonicus, P. fieberi, P. illyricus and P. ebneri, but not that of P. suphani, thus, P. suphani Taylan & Sirin, 2014 syn.n. synonymized with P. caucasicus (Fischer von Waldheim, 1846).  相似文献

Three types of invertase (invertase I, II and III) are separatedfrom the soluble and insoluble fractions (4,500xg, 10 min supernatantand pellets of the homogenate, respectively) of baker's yeastby a DEAE cellulose column chromatography. The invertases Iand II are eluted with 0.1 M sodium acetate buffer (pH 3.9)and with 0.1 M sodium acetate buffer (pH 6.2) containing 0.1M NaCl from DEAE cellulose respectively, whereas the invertase-IIIremains adsorbed on the cellulose under these conditions. Theyare present in proportions of 2.5: 1 : 0.06 in the soluble fractionand 1.4: 1 : 0.12 in the insoluble fraction of the fresh baker'syeast cells. While in-vertase-II remains at a constant level,invertases I and III in the soluble fraction increase upon incubationof cells for the formation of invertase under the continuoussupply of sucrose. Abstract.
  • 1 Monarch butterflies, Danaus plexippus, from two overwintering populations, were found to have Ophryocystis elektroscirrha spores on their scales at rates between 53% and 68%. The frequency of butterflies with O.elektroscirrha spores remained about the same between sites and throughout the winter.
  • 2 The spores, recovered from all parts of the body of the butterfly, were most numerous on the abdomen, particularly near the posterior third.
  • 3 Butterflies with spores survived as long as those without detectable spores at 10.1°C ±0.4 SE and 78.3% r.h. ±0.6SE. Insects with spores held at 19.4°C ±0.4SE and 44.9% r.h. ±1.5SE showed a significantly higher rate of moisture loss and survived a shorter period than monarch butterflies without detectable spores.

Plants traditionally used to dye carpets and woven matting in the eastern Mediterranean region were investigated. The survey was carried out in Kadirli, Feke, Aladağ, the surrounding villages of Adana, and in Osmaniye (Turkey). According to the survey results, 37 species of plants belonging to 29 families were used in natural dye production. The total dye contents of collected plants ranged from 1.5% to 10.0%. Mordants and dyes obtained from some of these plants were used to treat wool yarn and the yarn's dyed properties were tested. Some of the plants were also examined in terms of cultivation and fifteen were transferred to a Dye Plants Collection Garden. They were Alnus glutinosa (L.) Gaertn. ssp. antitaurica Yalt., Arctium minus ssp. pubens (Bab.) Arènes, Chrysanthemum segetum L., Cistus creticus L., Cotinus coggygria Scop., Datisca cannabina L., Galium verum ssp. glabrescens Ehrend., Isatis tinctoria ssp. tomentella (Boiss. & Balansa) P.H.Davis, Melissa officinalis ssp. inodora Bornm., Mentha longifolia (L.) Huds. ssp. longifolia , Reseda lutea var. lutea L., Rhamnus oleoides ssp. graecus (Boiss. & Reut.) 目的:对His/GST-HDAC1在大肠杆菌BL-21中的表达进行研究。方法:HDAC1的完整基因片断被克隆到pColdⅠ载体和pGEX载体上,并在其N末端分别联有His和GST;采用大肠杆菌BL21对HDAC1进行表达;采用亲和色谱对HDAC1进行纯化;用SDS-PAGE和蛋白质印迹来验证表达和纯化效果;对HDAC1活性进行测定。结果:多数HDAC1存在于大肠杆菌BL-21细胞裂解液的沉淀组分和纯化过程中的未结合组分中,小部分HDAC1可从细胞裂解液的上清液中得以纯化,但未显现出酶活性。用FPLC对HDAC1进行进一步分离,结果表明,HDAC1发生了分子聚集,使得它们的分子量大于正常分子量。结论:活性His/GST-HDAC1不能用大肠杆菌BL21成功表达。  相似文献
