The molecular mechanisms of cell cycle control are now known in enough detail to warrant mathematical modeling by kinetic equations. Despite the repetitive nature of the cell division cycle, the most appropriate models emphasize steady-state solutions rather than limit cycle oscillations, because cells progress toward division by passing a series of checkpoints (steady states).  相似文献

The accumulation of several hundred proteins during the nuclear division cycle of Physarum polycephalum was measured by digital image processing of silver-stained two-dimensional (2D) polyacrylamide gels. In contrast to previous studies, we have used an organism with a naturally synchronous cell cycle, so there are no uncertainties concerning synchronization artifacts or cell-sorting artifacts, and we have measured the specific amounts of each protein rather than the rate of synthesis. Since one-dimensional SDS-PAGE shows no significant fluctuations in the most abundant plasmodial proteins, we have loaded 2D gels so that proteins of low-to-moderate abundance appear in the linear range of the silver stain standard curve. Only five proteins showed reproducible, measureable fluctuations during the cell cycle. One of these proteins was tubulin. Full quantitative information was obtained by analysing the digital images of silver-stained gels by a general image processing system.  相似文献

Ultrastructural data are presented on time-course changes in isolated rat hepatocyte suspensions exposed either to 1.2 or 1.8 mM CCl4 for up to 1 h. The subcellular changes at the lower concentration, but not the higher, are shown to closely parallel those reported to occur in rat hepatocytes following ingestion of CCl4.  相似文献

Despite intense study by physicists and biologists, we do not fully understand the unsteady aerodynamics that relate insect wing morphology and kinematics to lift generation. Here, we formulate a force partitioning method (FPM) and implement it within a computational fluid dynamic model to provide an unambiguous and physically insightful division of aerodynamic force into components associated with wing kinematics, vorticity, and viscosity. Application of the FPM to hawkmoth and fruit fly flight shows that the leading-edge vortex is the dominant mechanism for lift generation for both these insects and contributes between 72–85% of the net lift. However, there is another, previously unidentified mechanism, the centripetal acceleration reaction, which generates up to 17% of the net lift. The centripetal acceleration reaction is similar to the classical inviscid added-mass in that it depends only on the kinematics (i.e. accelerations) of the body, but is different in that it requires the satisfaction of the no-slip condition, and a combination of tangential motion and rotation of the wing surface. Furthermore, the classical added-mass force is identically zero for cyclic motion but this is not true of the centripetal acceleration reaction. Furthermore, unlike the lift due to vorticity, centripetal acceleration reaction lift is insensitive to Reynolds number and to environmental flow perturbations, making it an important contributor to insect flight stability and miniaturization. This force mechanism also has broad implications for flow-induced deformation and vibration, underwater locomotion and flows involving bubbles and droplets.  相似文献

A deterministic/probabilistic model of the cell division cycle is analysed mathematically and compared to experimental data and to other models of the cell cycle. The model posits a random-exiting phase of the cell cycle and a minimum-size requirement for entry into the random-exiting phase. By design, the model predicts exponential "beta-curves", which are characteristic of sister cell generation times. We show that the model predicts "alpha-curves" with exponential tails and hyperbolic-sine-like shoulders, and that these curves fit observed generation-time data excellently. We also calculate correlation coefficients for sister cells and for mother-daughter pairs. These correlation coefficients are more negative than is generally observed, which is characteristic of all size-control models and is generally attributed to some unknown positive correlation in growth rates of related cells. Next we compare theoretical size distributions with observed distributions, and we calculate the dependence of average cell mass on specific growth rate and show that this dependence agrees with a well-known relation in bacteria. In the discussion we argue that unequal division is probably not the source of stochastic fluctuations in deterministic size-control models, transition-probability models with no feedback from cell size cannot account for the rapidity with which the new, stable size distribution is established after perturbation, and Kubitschek's rate-normal model is not consistent with exponential beta-curves.  相似文献

Probabilistic models of the cell cycle maintain that cell generation time is a random variable given by some distribution function, and that the probability of cell division per unit time is a function only of cell age (and not, for instance, of cell size). Given the probability density, f(t), for time spent in the random compartment of the cell cycle, we derive a recursion relation for n(x), the probability density for cell size at birth in a sample of cells in generation n. For the case of exponential growth of cells, the recursion relation has no steady-state solution. For the case of linear cell growth, we show that there exists a unique, globally asymptotically stable, steady-state birth size distribution, *(x). Several reports indicate that lactate can serve as an energy substrate for the brain. The rate of oxidation of this substrate by cultured rat brain astrocytes was 3-fold higher than the rate with glucose, suggesting that lactate can serve as an energy source for these cells. Since transport into the astrocytes may play an important role in regulating nutrient use by individuals types of brain cells, we investigated the uptake ofl-[U-14C]lactate by primary cultures of rat brain astrocytes. Measurement of the net uptake suggested two carrier-mediated mechanisms and an Eadie-Hofstee type plot of the data supported this conclusion revealing 2 Km values of 0.49 and 11.38 mM and Vmax values of 16.55 and 173.84 nmol/min/mg protein, respectively. The rate of uptake was temperature dependent and was 3-fold higher at pH 6.2 than at 7.4, but was 50% less at pH 8.2. Although the lactate uptake carrier systems in astrocytes appeared to be labile when incubated in phosphate buffered saline for 20 minutes, the uptake process exhibited an accelerative exchange mechanism. In addition, lactate uptake was altered by several metabolic inhibitors and effectors. Potassium cyanide and -cyano-4-hydroxycinnamate inhibited lactate uptake, but mersalyl had little or no effect. Phenylpyruvate, -ketoisocaproate, and 3-hydroxybutyrate at 5 and 10 mM greatly attenuated the rate of lactate uptake. These results suggest that the availability of lactate as an energy source is regulated in part by a biphasic transport system in primary astrocytes.This data was presented in part at the meeting of the Federation of American Societies for Experimental Biology in May 1989.  相似文献

I investigated competition for light between canopy plants and juvenile valley oaks (Quercus lobata Nee) in a mixed-broadleaf woodland of California's northern Coast Ranges. Canopy effects on understory light supply were separated among the overlying adult valley oak, the adult's woody understory, and neighboring trees and shrubs through a series of light sampling surveys and measurements of the number, size, and spatial distribution of neighboring plants. Light supply in the understory was primarily influenced by neighboring plants, with no detectable effect of the overlying adult valley oak. Light supply in the understory averaged 25% full sun due to a high frequency of canopy gaps and a typically open understory. Seedling response to understory light supply was investigated in an experimental sunfleck gradient (10%, 19%, and 100% full sun). Between 10% and 100% full sun, seedling growth increased by 90% and the shoot:∗∗∗root ratio changed from 1.561 to 0.607. Shade seedlings were also taller and produced fewer, larger, and thinner leaves than seedlings grown in full sun. A field survey of the spatial distribution and crown morphology of saplings and young adults found 1) the distance between young valley oaks and neighboring overstory trees to increase with neighbor size, and 2) crowns of the young oaks to be skewed away from neighbors. Although shading by the canopy was only moderate, canopy effects on understory light supply may restrict juvenile recruitment of valley oak in this woodland.  相似文献
