The rate and period of DNA synthesis during meiotic prophasewere examined using lily microsporocytes. Meiocytes at the earlyleptotene stage were cultured for discrete periods in the presenceof inhibitors of DNA synthesis, deoxyadenosine and nalidixicacid. Deoxyadenosine, which arrests meiotic development at theearly zygotene stage, markedly suppressed DNA synthesis to 35%of control at 2 mM. Nalidixic acid simply reduced the rate ofDNA synthesis, resulting in prolongation of the synthetic period.The relevance of DNA synthesis to meiotic development is discussed. Japanese encephalitis viruses (JEV) were well propagated in human glioma cells, 118MGC until the first 24 hrs after virus infection. However, after 24 hrs, virus growth rate was quickly reduced. This unusual pattern of virus growth was different from the cases in others cells, e.g. IMR-32, Vero and C6/36 cells. The fact that actinomycin-D retained the high yields of JEV in 118MGC cells suggests that some suppressing factors against JEV replication are produced in MGC cells. Interestingly, culture fluids of 118MGC cells indicated inhibitory effect to JEV reproduction, but other culture fluids from several cell lines had no effect. This inhibitory effect of the MGC-culture fluids was lost by heat-treatment at 60 C. In addition, the infectivity of JEV was rapidly decreased by the incubation with MGC-culture fluids. These findings suggest that 118MGC cells produce and secret some inhibitory factors against JEV replication.  相似文献

The antibiotic cordycepin (3'-deoxyadenosine), a known specificinhibitor of nucleic acid synthesis and polyadenylation of RNA,remarkably retarded decrease in the chlorophyll and proteincontents of senescing tobacco leaf disks. The effectivenessof cordycepin, at its optimum concentration (ca. 4 x 10–5M), was 53% as effective as a cytokinin, benzyladenine at 10–6M.  Relationships between leaf or shoot size, number, and arrangement in response to light were investigated to test the hypothesis that these characteristics are linked. In order to test this hypothesis, the divergence in allometry and shoot dynamics in saplings of Japanese beech (Fagus crenata) obtained from four populations and having different leaf sizes were examined in a nursery under both full sun and shade conditions. Trees with different leaf sizes also showed large differences in canopy structure, particularly when shade-grown saplings were compared. The final leaf mass distributions of the large-leaf populations were conical or “bottom - heavy”, while those of the small-leaf populations were planar or “top - heavy”. The slope of the allometric relations between leaf mass and shoot and branch mass in small-leaved populations were steeper than those in large-leaf populations. The four populations were classified into two growth types: populations producing a few large leaf and shoot modules corresponded to “stem growth type”, and those producing many small leaf and shoot modules corresponded to “leaf growth type”. These kinds of intra-specific variation in architecture and growth of F. crenata trees may influence the structure and dynamics of forests in accordance with differences in competitive ability or sensitivity to disturbances such as windstorm. To assess the impact of Bombus terrestris invasion on the foraging efficiency of native Japanese bumblebees, consumption and acquisition of floral resources during foraging on flowers of native Japanese plant species were investigated using enclosures with three treatments: one with only B. terrestris (exotic), one with both B. terrestris and native Japanese bumblebee species (mixed), and one with only Japanese species (native), but with the bumblebee density held constant. Changes in the body mass of queens and the nest mass of colonies for two days did not significantly differ among four combinations of the species and treatment, B. terrestris in the exotic and mixed treatments and Japanese species in the mixed and native treatments. Thus, it is not clear that B. terrestris has higher foraging efficiency than native species and that B. terrestris individuals more negatively affect the foraging efficiency of native species than individuals of the native species themselves. The nectar standing crop of Cirsium kamtschaticum was smaller in the exotic treatment than in the mixed and native treatments. Quantification of annual carbon sequestration is very important in order to assess the function of forest ecosystems in combatting global climate change and the ecosystem responses to those changes. Annual cycling and budget of carbon in a forested basin was investigated to quantify the carbon sequestration of a cool-temperate deciduous forest ecosystem in the Horonai stream basin, Tomakomai Experimental Forest, northern Japan. Net ecosystem exchange, soil respiration, biomass increment, litterfall, soil-solution chemistry, and stream export were observed in the basin from 1999–2001 as a part of IGBP-TEMA project. We found that 258 g C m–2 year–1 was sequestered annually as net ecosystem exchange (NEE) in the forested basin. Discharge of carbon to the stream was 4 g C m–2 year–1 (about 2% of NEE) and consisted mainly of dissolved inorganic carbon (DIC). About 43% of net ecosystem productivity (NEP) was retained in the vegetation, while about 57% of NEP was sequestered in soil, suggesting that the movement of sequestered carbon from aboveground to belowground vegetation was an important process for net carbon accumulation in soil. The derived organic carbon from aboveground vegetation that moved to the soil mainly accumulated in the solid phase of the soil, with the result that the export of dissolved organic carbon to the stream was smaller than that of dissolved inorganic carbon. Our results indicated that the aboveground and belowground interaction of carbon fluxes was an important process for determining the rate and retention time of the carbon sequestration in a cool-temperate deciduous forest ecosystem in the southwestern part of Hokkaido, northern Japan.  相似文献

This data paper reports litter fall data collected in a network of 21 forest sites in Japan. This is the largest litter fall data set freely available in Japan to date. The network is a part of the Monitoring Sites 1000 Project launched by the Ministry of the Environment, Japan. It covers subarctic to subtropical climate zones and the four major forest types in Japan. Twenty-three permanent plots in which usually 25 litter traps were installed were established in old-growth or secondary natural forests. Litter falls were collected monthly from 2004, and sorted into leaves, branches, reproductive structures and miscellaneous. The data provide seasonal patterns and inter-annual dynamics of litter falls, and their geographical patterns, and offer good opportunities for meta-analyses and comparative studies among forests.  相似文献

To determine whether latitudinal variation in herbivore impact exists, we examined three major herbivorous insect feeding types (chewers, gallers, and miners) on/in leaves of Japanese beech. Herbivores were collected with litter traps deployed in forests across a latitudinal gradient of 10°. Leaf litter analyses demonstrated that chewing herbivory increased with increasing latitude of collection site. However, the densities of miners and gallers decreased with latitude. To test whether latitudinal variation in herbivore damage occurs in the absence of geographically differentiated environmental cueing (e.g., physical stresses or herbivore damage), we measured both genetically determined constitutive leaf traits and herbivore damage in a common-garden experiment. In this experiment, miner density decreased with latitude, but chewing herbivory did not vary latitudinally. Galler density was higher on trees from native provenances than on trees from unrelated provenances likely because of local adaptations. Leaf mass per unit area (LMA), tannin, and phenolics all decreased with latitude of provenance. The latitudinal variation in one constitutive leaf trait (LMA) best explained latitudinal variation in chewing herbivory. Thus, different mechanisms account for feeding type-specific patterns of latitudinal variation in herbivore damage among different herbivore feeding types.  相似文献

The increased atmospheric deposition of nitrogen (N) may indirectly affect herbivorous insects by deposition-induced changes in host quantity and quality. To avoid the “lamp effect” that can occur in small-scale N fertilizations, large-scale N fertilization (ca. 9 ha, 100 kg N ha?1 year?1) experiments were performed in a deciduous, broad-leaved, cool temperate forest. The initial responses of mature oak canopy trees (Quercus crispula) and their herbivorous insects to N fertilization were evaluated by measuring the growth and leaf qualities of the trees. The feeding guilds and community structures of the herbivorous insects at control and fertilized sites before (2012) and after (2013–2014) N fertilization were then determined. In 2014, N fertilization enhanced plant growth. In 2013 but not 2014, N fertilization increased N content and decreased the carbon/nitrogen (C/N) ratio in leaves. Despite these changes in plant traits in 2013, N fertilization had no effect on either feeding guilds (chewing herbivory, galler density, and miner density) or community structures (species richness, diversity index, and relative abundance) of herbivorous insects in the same year. However, in 2014, the diversity index decreased significantly, whereas species richness and abundance were unchanged. This suggests that species-specific responses to changes in leaf qualities following N fertilization, in the form of altered insect fecundity, impact the diversity index of herbivorous insects, albeit with a 1-year lag time. Thus, our large-scale N fertilization experiment show the time-delayed bottom-up effects of N fertilization on insect community structure.  相似文献
