Rabbit alveolar macrophages exhibit a chemiluminescent response which is associated with phagocytosis of zymosan and polystyrene-butadiene particles. The chemiluminescence reaches a peak in 15 to 25 minutes and then gradually diminishes over the next 1 to 3 hours. During the time of maximal light emission there appears to be no actual uptake of particles, but the response is dependent upon the particle concentration. The metabolic inhibitor, DNP (2,4-dinitrophenol), causes a rapid inhibition of the chemiluminescent response. The addition of ATP to the medium prior to exposure of the cells to particles causes the chemiluminescent response to be greatly diminished, i.e., 0.3mM ATP virtually abolishes the response. These experiments suggest that some metabolic response of the cell to phagocytosis is responsible for the chemiluminescence. 相似文献

Unstimulated alveolar macrophages, but not polymorphonuclear leukocytes, elicit chemiluminescence from lucigenin which cannot be entirely accounted for by the resting level of superoxide generation. This chemiluminescence was inhibited by both superoxide scavengers and inhibitors of mitochondrial respiration. Although 12-O-tetradecanoyl phorbol-13-acetate addition resulted in a significant increase in cellular superoxide generation, an unexpected decrease in lucigenin chemiluminescence was noted. These results suggest that mitochondria in alveolar macrophages may be a site of lucigenin accumulation and dioxygenation and that 12-O-tetradecanoyl phorbol-13-acetate may modulate this activity. 相似文献

To test the possibility that the theta-rhythm may spread passively from the hippocampus into the neocortex in rabbits and to analyze the properties of these rhythms recorded from the surface of the neocortex, an investigation was made of activity recorded simultaneously from the hippocampus and various points of the neocortex, including from a strip of the sensomotor cortex isolated completely with respect to its nervous connections. The use of special correlation methods showed that an appreciable contribution to the potentials recorded from the isolated strip can be made by the passive conduction of hippocampal activity (provided that it generates a regular high-amplitude theta-rhythm). At the same time, potentials in a theta-rhythm may be generated in the isolated strip itself. The theta-rhythm recorded in the intact rabbit cortex may thus be of combined genesis: generated in the neocortex itself and spreading passively from the hippocampus through the brain tissues as through a volume conductor. 相似文献

The interaction of bleomycin A2 with rat lung microsomes results in bleomycin-mediated DNA chain breakage due to the mixed-function oxidase catalyzed activation of bleomycin. This study demonstrates that the addition of exogenous Fe3+ significantly enhances the bleomycin-mediated cleavage of DNA deoxyribose, that the enhancing effect of Fe3+ is maximum when a 1:1 ratio of bleomycin to Fe3+ is achieved and that either NADPH or NADH can serve as pyridine cofactors for this reaction. Since the activation of bleomycin can be facilitated by iron in the Fe2+ form, these observations support the hypothesis that the mixed-function oxidase system may serve to maintain either adventitious or exogenous iron in the Fe2+ form. In the absence of DNA, the interaction of bleomycin with rat lung microsomes results in the self-inactivation of bleomycin, a reaction which is also enhanced by the addition of exogenous Fe3+. Thus, the microsomal mixed-function oxidase system represents an efficient biological system for the ‘activation-inactivation’ of bleomycin. 相似文献

Feeding male weanling rats on a vitamin A-deficient diet for 6 weeks resulted in significant increases (44-57%) in glutathione S-aryl-, S-aralkyl- S-alkyl- and S-epoxidetransferase activities in the liver cytosol. Only the S-aralkyl- (27%) and S-alkyltransferase (14%) activities were significantly increased in the kidney as a result of deficiency. There was no effect on any of the pulmonary glutathione S-transferase activities. The increases in hepatic transferase activities were due primarily to increases (25-96%) in the apparent Vmax. There were no changes in the apparant Km of any of the four drug substrates employed. With 3,4-dichloronitrobenzene as the second substrate, the apparent Km for glutathione was increased by over 2-fold in vitamin A-deficient livers as compared with controls. The relationship between these results and enhanced susceptibility to chemical carcinogens in vitamin A deficiency is briefly discussed, and comparison is made between the effects of this nutritional state and pretreatment with drug inducers on the glutathione S-transferases. 相似文献
