Cytokinins in young wheat (Triticum aestivum cv. Chinese spring) spikes (2–15 mm) were isolated by immunoaffinity chromatography. High-performance liquid chromatography-radioimmunoassay and gas chromatography-mass spectrometry indicated that major cytokinins present weretrans-zeatin, ribosyl-trans-zeatin, ribosyldihydrozeatintrans-zeatin-9-glucoside, and the glucosides oftrans-zeatin, ribosyl-trans-zeatin, andtrans-zeatin-9-glucoside. Dihydrozeatin,iso-pentenyladenosine, andiso-pentenyladenine were also present but at lower concentrations.Mention of a trade name, proprietary product, or specific equipment does not constitute a guarantee or warranty by the U.S. Department of Agriculture and does not imply approval to the exclusion of other products that may also be suitable. Analyses of plumules from seedlings of a winter wheat variety (Elgin) and a spring wheat variety (Red Bobs) indicated that SH and SS contents based on protein levels were similar in the whiter and spring wheat tissues grown at 20 C. Boll) varieties, when grown nt 3 C, contained greater concentrations of SH and SS groups, based on protein levels, than those present in the tissues grown at 20 C. The spring wheat tissues at 3 C maintained relatively constant levels of SH and SS during the growth period. Sulfhydryl (SH) and SS groups based on fresh weights were present in greater quantities in the tissues of the spring wheat grown at 20 C than in the winter wheat tissues grown at 20 C. The high SH and SS concentrations present in the warm-grown spring wheat tissues were due to a high concentration of protein extracted from that tissue. The SH and SS contents per unit fresh weight were quite similar in the cold-grown tissues of both varieties during the growth period. Both varieties at both temperatures showed a similar decrease in SH and SS contents, based on fresh weights, as maturation progressed. The spring wheats grown at 3 C and 20 C and the winter wheat grown at 3 C all showed similar characteristics and differed considerably from the winter wheat grown at 20 C. These data were positively correlated with the flowering potential of the wheat varieties.  相似文献

Seven monoclonal anti-zeatin riboside antibodies were characterized by radioimmunoassay (RIA) and found to measure femtomole (10–15 M) quantities (20 pg) of this cytokinin. The antibodies had different measuring ranges defined by the linear portion of the logit/log plots; slopes and intercepts of the line varied considerably between the antibodies. Competitive binding trials againstcis-zeatin riboside (cZR), dihydrozeatin riboside (diHZR), zeatin (Z), and isopentenyl adenosine (iPA) showed differences among the seven antibodies in their cross-reactivities towards these structurally related cytokinins. It was possible to combine selected antibodies to provide a mixture with a predictable measuring range and cross-reactivity; the ability to prepare a highly specific reagent in this manner with well-defined reactivity was noted and differences between monoclonal antibody and polyclonal antiserum probes for measurement of cytokinins were discussed.The research was supported by the Agricultural Research Service, U.S. Department of Agriculture, Technical Paper No. 7373 of the Oregon Agricultural Experiment Station. Teliospores were aerated and agitated in a mineral salts medium and their free amino acid contents were analyzed at eight different times, from shortly after imbibition of water until just before germ tube emergence. In addition to the common amino acids, eight unidentified ninhydrin-positive components were detected. About 50 % or more of nearly each of the amino acids diffused out of the spores during the initial phase of germination. These released amino acids were actively taken up by the spores during the latter stages of germination. The free amino acids in largest amounts in the dormant spores of T. caries were arginine 15.0, glutamic acid 6.3, and alanine 3.7 μmoles per g dry spores. Together these three amino acids accounted for about 71 % of the total free amino acids in dormant spores of T. caries and T. controversa. The total amounts of free amino acids in spores of common bunt were much higher than in spores of dwarf bunt.  相似文献

Background  

Parkinson's disease (PD) is the second most common neurodegenerative disorder. As there is no definitive diagnostic test, its diagnosis is based on clinical criteria. Recently transcranial duplex scanning (TCD) of the substantia nigra in the brainstem has been proposed as an instrument to diagnose PD. We and others have found that TCD scanning of substantia nigra duplex is a relatively accurate diagnostic instrument in patients with parkinsonian symptoms. However, all studies on TCD so far have involved well-defined, later-stage PD patients, which will obviously lead to an overestimate of the diagnostic accuracy of TCD.  相似文献

On a wheat-based medium, the pathogenic phase of the common and dwarf bunt fungi grew slowly at 15–18 C and continued to produce massive quantities of teliospores in all subcultures for over 2 years. At warmer temperatures or on a chemically defined medium, the teliosporogenic colonies reverted to haploid mononucleate colonies. The hyphae of the teliosporogenic colonies were stained with a modified Giemsa technique and found to be thick, contorted, highly branched and short celled with usually one or two nuclei per cell. In contrast, the haploid mononucleate hyphae were thinner, straighter, and longer and never contained more than one nucleus per cell. The vegetative hyphae of T. caries and T. controversa were indistinguishable. Teliospores, formed at the terminal end of binucleate hyphae, were initially binucleate but became mononucleate before the mature cell wall formed.  相似文献

Abstract. Woodland colonization on wetlands is considered to have a detrimental effect on their ecological value, even though detailed analysis of this process is lacking. This paper provides an evaluation of the ecological changes resulting from succession of poor fen (base‐poor mire) to willow wet woodland on Goss Moor NNR in Cornwall, UK. Different ages of willow carr were associated with eight understorey communities. During willow colonization, in the ground flora, there was a progressive decrease in poor fen species and an associated increase in woodland species, which appeared to be related to an increase in canopy cover and therefore shade. The most diverse community was found to be the most recent willow and was dominated by poor fen species. The oldest willow was the second most diverse and was associated with a reduction in poor fen species and an increase in woodland species. Architectural features were used successfully to assess the general condition and structure of willow. Tree height and DBH were identified as useful parameters to accurately assess willow age in the field. The implications of active intervention to remove willow in order to conserve the full range of communities within the hydrosere are discussed.  相似文献
