Ocean acidification may have far‐reaching consequences for marine community and ecosystem dynamics, but its full impacts remain poorly understood due to the difficulty of manipulating pCO2 at the ecosystem level to mimic realistic fluctuations that occur on a number of different timescales. It is especially unclear how quickly communities at various stages of development respond to intermediate‐scale pCO2 change and, if high pCO2 is relieved mid‐succession, whether past acidification effects persist, are reversed by alleviation of pCO2 stress, or are worsened by departures from prior high pCO2 conditions to which organisms had acclimatized. Here, we used reciprocal transplant experiments along a shallow water volcanic pCO2 gradient to assess the importance of the timing and duration of high pCO2 exposure (i.e., discrete events at different stages of successional development vs. continuous exposure) on patterns of colonization and succession in a benthic fouling community. We show that succession at the acidified site was initially delayed (less community change by 8 weeks) but then caught up over the next 4 weeks. These changes in succession led to homogenization of communities maintained in or transplanted to acidified conditions, and altered community structure in ways that reflected both short‐ and longer‐term acidification history. These community shifts are likely a result of interspecific variability in response to increased pCO2 and changes in species interactions. High pCO2 altered biofilm development, allowing serpulids to do best at the acidified site by the end of the experiment, although early (pretransplant) negative effects of pCO2 on recruitment of these worms were still detectable. The ascidians Diplosoma sp. and Botryllus sp. settled later and were more tolerant to acidification. Overall, transient and persistent acidification‐driven changes in the biofouling community, via both past and more recent exposure, could have important implications for ecosystem function and food web dynamics.  相似文献

Aim Recreational boating is arguably the largest unregulated vector for the introduction and spread of marine invasive species. Hull fouling communities have been recognized to harbour non‐indigenous species (NIS), but presence should not be equated with transport. In this study, we characterize the presence of NIS in hull fouling communities, determine if host vessels transport these species and evaluate the importance of recreational boating as a vector for introduction and spread. Location Coastal British Columbia (BC), Canada. Methods Dive surveys in BC marinas were conducted to record the presence of NIS and to estimate their per cent cover. In addition, a boater questionnaire survey was used to determine common travel and maintenance practices. These results were combined to investigate the potential for recreational boats to transport NIS. Results Nine NIS, including the highly invasive ascidians Styela clava and Botrylloides violaceus, and the macroalga Sargassum muticum, were found in hull fouling communities on recreational boats. Overall, per cent cover was generally low; however, niche areas were commonly fouled, even on active and otherwise clean boats. Fouling of niche areas was not related to either antifouling paint age or travel frequency, and fouling levels were highly variable among individual boats both within marinas and across regions. Main conclusions Recreational boating is a major vector contributing to the spread of marine invasive species. Our results indicate that recreational boats represent a high‐risk vector both for primary introduction and secondary spread of marine NIS and should be subject to vector management regulations.  相似文献

We tested the links among biodiversity, habitat heterogeneity and physical stress in a system of artificial rock pools on the north coast of Jamaica that mimic natural aquatic invertebrate communities. The experimental design consisted of three tiers of small plastic pools arranged at increasing distances from the shore. As a result of community development over six months (January to June 1997), we observed considerable differentiation of physical conditions among replicate habitats at the benign end of the physical gradient, with a concurrent increase in biodiversity (species richness per habitat unit). The most probable explanation for this observed gradient is self-generated habitat heterogeneity that, in turn, promotes biodiversity, likely through species interactions. Using additional analyses, including randomization techniques, we excluded the effects of sample size and external factors as sources for the observed increase in biodiversity in the third tier (furthest from the sea). We interpret this result as evidence for the complex causal relationship among physical stress, habitat heterogeneity and biodiversity.  相似文献

Bone tissue has an exceptional quality to regenerate to native tissue in response to injury. However, the fracture repair process requires mechanical stability or a viable biological microenvironment or both to ensure successful healing to native tissue. An improved understanding of the molecular and cellular events that occur during bone repair and remodeling has led to the development of biologic agents that can augment the biological microenvironment and enhance bone repair. Orthobiologics, including stem cells, osteoinductive growth factors, osteoconductive matrices, and anabolic agents, are available clinically for accelerating fracture repair and treatment of compromised bone repair situations like delayed unions and nonunions. Preclinical and clinical studies using biologic agents like recombinant bone morphogenetic proteins have demonstrated an efficacy similar or better than that of autologous bone graft in acute fracture healing. A lack of standardized outcome measures for comparison of biologic agents in clinical fracture repair trials, frequent off-label use, and a limited understanding of the biological activity of these agents at the bone repair site have limited their efficacy in clinical applications.  相似文献

Improving our understanding of species responses to environmental changes is an important contribution ecologists can make to facilitate effective management decisions. Novel synthetic approaches to assessing biodiversity and ecosystem integrity are needed, ideally including all species living in a community and the dynamics defining their ecological relationships. Here, we present and apply an integrative approach that links high‐throughput, multicharacter taxonomy with community ecology. The overall purpose is to enable the coupling of biodiversity assessments with investigations into the nature of ecological interactions in a community‐level data set. We collected 1195 gastropods and crabs in British Columbia. First, the General mixed Yule‐coalescent (GMYC) and the Poisson Tree Processes (PTP) methods for proposing primary species‐hypotheses based on cox1 sequences were evaluated against an integrative taxonomic framework. We then used data on the geographic distribution of delineated species to test species co‐occurrence patterns for nonrandomness using community‐wide and pairwise approaches. Results showed that PTP generally outperformed GMYC and thus constitutes a more effective option for producing species‐hypotheses in community‐level data sets. Nonrandom species co‐occurrence patterns indicative of ecological relationships or habitat preferences were observed for grazer gastropods, whereas assemblages of carnivorous gastropods and crabs appeared influenced by random processes. Species‐pair associations were consistent with current ecological knowledge, thus suggesting that applying community assembly within a large taxonomical framework constitutes a valuable tool for assessing ecological interactions. Combining phylogenetic, morphological and co‐occurrence data enabled an integrated view of communities, providing both a conceptual and pragmatic framework for biodiversity assessments and investigations into community dynamics.  相似文献

A large (1200 1) seawater sample from the Gulf of Maine waskept under constant temperature and light conditions for a periodof 72 h. Aim The introduction of non‐indigenous species has resulted in wide‐ranging ecological and economic impacts. Predictive modelling of the introduction and establishment of non‐indigenous species is imperative to identify areas at high risk of invasion to effectively manage non‐indigenous species and conserve native populations. Smallmouth bass (Micropterus dolomieu), a warm water fish species native to central North America has negatively impacted native fish communities, including cyprinids and salmonid populations, as a result of intentional introductions. We predicted the introduction risk; species establishment based on habitat suitability; identified lakes at high risk of invasion; and finally assessed the consequential impacts on native salmon, trout and cyprinid populations. Location Ontario and British Columbia, Canada. Methods Classification tree and logistic regression models were developed and validated to predict the introduction and establishment of smallmouth bass for thousands of lakes. Results Densely human populated areas and larger lake surface areas successfully identify lakes associated with the introduction of smallmouth bass (introduction model) in British Columbia. Climate, lake morphology and water chemistry variables were the driving environmental parameters to define suitable smallmouth bass habitat (establishment model). A combination of the introduction and establishment model identified 138 lakes that are currently at risk in British Columbia to the introduction and establishment of smallmouth bass. Of these 138 high‐risk lakes, 95% of them contain at least one species of salmon, trout or cyprinid, thereby increasing the potential impact of an invasion by smallmouth bass. Main conclusions Our framework can be applied to other terrestrial and aquatic species to obtain a better understanding of the potential risk posed by a non‐indigenous species to an ecosystem. Furthermore, our methodology can be used to focus management efforts on areas at higher risk (e.g. number of potential releases, more favourable habitats) to control future introductions of non‐indigenous species, thereby conserving native populations.  相似文献

Breast cancer patients frequently develop metastases. This process requires the degradation of extracellular matrix proteins which act as a barrier to tumour cell passage. These proteins can be degraded by proteases, mainly the matrix metalloproteinases (MMPs). MMP-2 and -9 which are frequently detected in breast cancer tissues. ProMMPs are released from cancer cells, and their activation is considered to be a crucial step in metastases development. In breast cancer, estrogen metabolism is altered favouring the accumulation of 2- and 4-hydroxyestradiol (2- and 4-OHE(2)). These estradiol metabolites can generate free radicals. Since reactive species are known activators of proMMPs, this study was designed to determine if the free radicals generated by 2- and 4-OHE(2) can activate proMMP-2 and -9. Activation of MMPs by hydroxyestradiol was determined by monitoring the cleavage of a fluorogenic peptide and by zymography analysis. Both estradiol metabolites activated the MMP-2 and -9. 4-OHE(2) was a more potent activator than 2-OHE(2), which reflects its higher capacity to generate free radicals. ProMMPs activation was mainly mediated through O(2)*-, although the free radical HO* also activated the proMMPs but to a lesser extent. ProMMPs activation was not observed with estrogens that cannot generate free radicals, i.e. estradiol, estrone, 2- and 4-methoxyestradiol, and 16alpha-hydroxyestrone. These results demonstrate that 2- and 4-OHE(2) at a concentration as low as 10(-8)M can activate the proMMP-2 and -9 and might play an important role in the invasion of breast cancer cells.  相似文献

A method is described for the detection of glutathione (GSH) and glutathione disulfide (GSSG) based on a HPLC postcolumn reaction with ortho-phthalaldehyde (OPT) at pH 12 followed by fluorescence detection. Although similar methods have been reported, the high pH of the postcolumn reaction adds considerable selectivity and sensitivity to the measurement of GSH and glutathione disulfide. The limit of detection approaches 100 fmol, which is sufficient to detect whole-cell glutathione disulfide in 10,000 cells or mitochondrial glutathione disulfide in 20 million cells. Using this method, glutathione and glutathione disulfide were measured in human lymphocytes, granulocytes, and cultured Jurkat T cells, as well as in the corresponding samples of mitochondria. The percentage of glutathione disulfide to total glutathione in whole-cell extracts was approximately 1%. In contrast, the percentage was relatively high in mitochondria, with the mitochondria of granulocytes having the highest (25%) followed by those of lymphocytes (15%) and finally by cultured Jurkat T cells (9%). This method extends the analysis of glutathione and glutathione disulfide to mitochondria obtained from a relatively small number of cells.  相似文献
