The incidence of polyovular types in the growing follicle population was estimated using quantitative cytology. Of 15 species studied, polyovular follicles were recorded in the following species and in ascending order of abundance: rabbits, rhesus monkeys, humans, cats, dogs. The incidence in bitches was 14% in animals aged 1-2 years but only 5% at 7-11 years old. The frequency of the various types of polyovular preantral follicle varied inversely with the numbers of oocytes per follicle and the probability of finding a follicle with more than 5 oocytes was remote. In young ovaries the frequency was constant in the early stages of growth but decreased in the largest preantral stage. The pattern in ageing ovaries was, by contrast, one of declining frequency such that few if any polyovular types completed development. The ovary of the ageing bitch was also characterized by a higher incidence of degenerating follicles and a much smaller pool of primordial stages. Polyovular follicles were larger than uniovular types at comparable stages which were defined by the number of granulosa cell layers. Their oocytes were smaller but the overall ooplasmic mass was increased with a corresponding increase in the mass of granulosa cells.  相似文献

Nitrogenases from Anabaena cylindrica and Chloropseudomonas ethylicum were partially purified into two components. A. cylindrica fraction I protein complemented fraction II protein from C. ethylicum. The oocytes of saturniid moths take up proteins selectively from the blood. The distribution of blood proteins in the ovary during protein uptake was investigated by staining 2 µ sections of freeze-dried ovaries with fluorescein-labeled antibodies. The results indicate that blood proteins occur primarily in the intercellular spaces of the follicle cell layer, in association with a brush border at the surface of the oocyte, and within the oocyte in the yolk spheres. That proteins derived from the blood are associated with the yolk spheres was confirmed by isolating these bodies and showing that lysis, which can be induced by any of a number of mechanical means, causes them to release immunologically defined proteins known to be derived from the blood. That the level of blood proteins in the cytoplasm is low relatively to that in the yolk spheres was confirmed by the observation that the yellow pigments associated with several blood proteins, although conspicuous in the yolk spheres, are not visible in the translucent layer of centrifuged oocytes. From these and previous physiological observations, it is proposed that blood proteins reach the surface of the oocyte by an intercellular route, that they combine with some component of the brush border, and that they are transformed into yolk spheres by a process akin to pinocytosis.  相似文献

Communication between the oocyte and its somatic cells has been shown to be important in oocyte development. Here we examined how the oocyte may be involved in bovine cumulus cell expansion. Intact bovine cumulus oocyte complexes (COC) were obtained by puncturing antral follicles. From the intact COC, oocytectomised complexes (OOX) were produced by micro surgical removal of the oocyte. Clumps of cumulus cells (CC) were obtained by micro-dissection. Intact or OOX complexes or CC were matured in the presence of fetal calf serum and hFSH (6 mlU/ml) for 24 hr and the degree of expansion measured. The presence of the oocyte is not essential to allow bovine cumulus expansion to occur as expansion occurred in all groups. Murine OOX complexes from eCG primed 35–40-day-old C57BL6/CBA F1 hybrids (known to require the presence of an oocyte secreted factor for cumulus expansion) were cultured with or without denuded bovine oocytes (1 oocyte/μl). Murine OOX complexes expanded only in the presence of denuded bovine oocytes. Thus some factor produced by bovine oocytes enabled expansion of murine OOX complexes. To determine whether the factor is secreted by bovine oocytes, murine OOX were cultured with or without media conditioned by bovine oocytes (1 oocyte/μl for 4 hr). Significant expansion of murine OOX occurred in media conditioned by bovine oocytes. This shows that the cumulus expansion enabling effect of bovine oocytes is released into the surrounding media. Media conditioned by bovine oocytes and then frozen for up to 1 month showed that the activity by the factor can withstand freezing. © 1995 wiley-Liss, Inc.  相似文献

The toxicity of ochratoxin A (OTA), a mycotoxin produced by fungi ofAspergillus orPenicillium genera is now well documented. Its nephrotoxicity, immunosuppression, teratogenicity, and carcinogenicity have been widely studied. Physical and biochemical methods have been studied to prevent these toxinogenicAspergillus andPenicillium from producing OTA, and/or to destroy the mycotoxin when already produced in a liquid or a solid medium. Repeated freezing at ? 20?C and thawing at + 26?C aleatory reduce OTA production in a liquid medium. Exposure to UV B for different periods of time is efficient in preventing OTA production in a liquid medium. Gamma-irradiation from 2 to 5 kGy gives good results in preventing the production of OTA or destroying it when already produced. Carboxypeptidase is very efficient at 5 units/50 ml in a liquid medium for cleaving the OTA already produced.  相似文献

Activation of cAMP-dependent protein kinase (PKA) by cell-permeable analogs of cAMP causes early and mid-vitellogenic follicles of Hyalophora cecropia to terminate vitellogenin uptake [[Wang and Telfer, 1996], Insect Biochem. Mol. Biol. 26, 85-94 (1996)]. The response is shown here to entail the formation of an epithelial diffusion barrier. Follicle cells that have been loosely organized to provide intercellular pathways for the movement of vitellogenin to the oocyte surface transform into a tight epithelium within 1-2h of exposure to PKA activators. The follicle cells can now prevent the escape of Lucifer yellow CH that has been iontophoresed into the space surrounding the oocyte, and the entry of labeled vitellogenin from the medium. As they form this functional equivalent of a tight junction, the follicle cells further reduce the intercellular spaces by enlarging and pressing against each other, and by slowing the secretion of the sulfated glycosaminoglycan matrix that separates them during vitellogenesis. The activation of PKA in early and mid-vitellogenic follicles thus appears to trigger prematurely a set of changes that do not normally occur until the follicle has grown to a length of about 2.0mm.  相似文献

The extent of zoonotic infections in rural Sierra Leone, where both feral and pet sooty mangabeys harbor divergent members of the human immunodeficiency virus type 2 (HIV-2)-sooty mangabey simian immunodeficiency virus (SIVsm) family, was tested in blood samples collected from 9,309 human subjects in 1993. Using HIV-1- and HIV-2-specific enzyme immunoassays and confirmatory Western blot analysis to test for antibodies to SIVsm-related lentiviruses, we found only nine subjects (0.096%) who tested positive for HIV: seven tested positive for HIV-1 and two tested positive for HIV-2. Compared with other rural West African communities, Sierra Leone displayed the lowest seroprevalence (0.021%) of HIV-2 infection yet reported, much lower than the previously reported seroprevalence in SIVsm-infected feral and household pet sooty mangabeys. Heteroduplex analysis demonstrated that two of the newly found HIV-1 strains belonged to subtype A, the most common HIV-1 subtype in Africa, but this is the first report of subtype A in Sierra Leone. The two HIV-2-infected individuals harbored two distinct HIV-2 strains, designated 93SL1 and 93SL2. Phylogenetic analysis indicated that HIV-2 93SL1 is a member of HIV-2 subtype A, the first strain of this HIV-2 subtype found in Sierra Leone. In contrast, HIV-2 93SL2 belongs to none of the five previously characterized HIV-2 subtypes (A to E) but is a new subtype, herein designated F, having the most divergent transmembrane sequences yet reported for HIV-2. The fact that both of the two most divergent HIV-2 subtypes known, E and F, are rare and found as single occurrences in persons from Sierra Leone may be related to the fact that this small region of West Africa also contains free-living and household pet sooty mangabeys with highly divergent variants of SIVsm. This finding provides support for the hypotheses that new HIV-2 subtypes result from independent cross-species transmission of SIVsm to the human population and that these single-occurrence transmission events had not spread widely into the population by 1993.  相似文献
