Background

Glossina pallidipes is a haematophagous insect that serves as a cyclic transmitter of trypanosomes causing African Trypanosomiasis (AT). To fully assess the role of G. pallidipes in the epidemiology of AT, especially the human form of the disease (HAT), it is essential to know the microbial diversity inhabiting the gut of natural fly populations. This study aimed to examine the diversity of G. pallidipes fly gut bacteria by culture-dependent approaches.

Results

113 bacterial isolates were obtained from aerobic and anaerobic microorganisms originating from the gut of G. pallidipes. 16S rDNA of each isolate was PCR amplified and sequenced. The overall majority of identified bacteria belonged in descending order to the Firmicutes (86.6%), Actinobacteria (7.6%), Proteobacteria (5.5%)and Bacteroidetes (0.3%). Diversity of Firmicutes was found higher when enrichments and isolation were performed under anaerobic conditions than aerobic ones. Experiments conducted in the absence of oxygen (anaerobiosis) led to the isolation of bacteria pertaining to four phyla (83% Firmicutes, 15% Actinobacteria, 1% Proteobacteria and 0.5% Bacteroidetes, whereas those conducted in the presence of oxygen (aerobiosis) led to the isolation of bacteria affiliated to two phyla only (90% Firmicutes and 10% Proteobacteria). Phylogenetic analyses placed these isolates into 11 genera namely Bacillus, Acinetobacter, Mesorhizobium, Paracoccus, Microbacterium, Micrococcus, Arthrobacter, Corynobacterium, Curtobacterium, Vagococcus and Dietzia spp.which are known to be either facultative anaerobes, aerobes, or even microaerobes.

Conclusion

This study shows that G. pallidipes fly gut is an environmental reservoir for a vast number of bacterial species, which are likely to be important for ecological microbial well being of the fly and possibly on differing vectorial competence and refractoriness against AT epidemiology.

Replicate 2-D gels were stained with four visible or fluorescent dyes using published procedures, and 48 co-detected spots were selected for contrasting values in abundance, M(r) and pI. Success rate of identification and sequence coverage were affected in a dye-dependent manner by the three parameters. Frequency of missed cleavages and recovery of sulfur-containing peptides also depended on the dye. Finally, the dataset was used to predict the number of proteins identifiable when integrating the differential contribution of each parameter. Sypro Ruby appeared to combine several favorable features: no dependence of the identification rate upon the physicochemical properties of proteins, no impact on frequency of missed cleavages, and a higher predicted identification rate.  相似文献

Angiotensin-I-converting enzyme (ACE) inhibitory activity of a tryptic digest of bovine alpha(S2)-casein (alpha(S2)-CN) was extensively investigated. Forty-three peptide peaks were isolated and tested. Seven casokinins (i.e. CN-derived ACE inhibitory peptides) were identified and their IC50 values were determined. Four peptides exhibited an IC50 value lower than 20 microM. Peptides alpha(S2)-CN (f174-181) and alpha(S2)-CN (f174-179) had IC50 values of 4 microM. Surprisingly, deletion of the C-terminal dipeptide of two of these casokinins did not significantly alter their inhibitory activity.  相似文献

Tissue damage induces early recruitment of neutrophils through redox-regulated Src family kinase (SFK) signaling in neutrophils. Redox-SFK signaling in epithelium is also necessary for wound resolution and tissue regeneration. How neutrophil-mediated inflammation resolves remains unclear. In this paper, we studied the interactions between macrophages and neutrophils in response to tissue damage in zebrafish and found that macrophages contact neutrophils and induce resolution via neutrophil reverse migration. We found that redox-SFK signaling through p22phox and Yes-related kinase is necessary for macrophage wound attraction and the subsequent reverse migration of neutrophils. Importantly, macrophage-specific reconstitution of p22phox revealed that macrophage redox signaling is necessary for neutrophil reverse migration. Thus, redox-SFK signaling in adjacent tissues is essential for coordinated leukocyte wound attraction and repulsion through pathways that involve contact-mediated guidance.  相似文献

An improved genetic linkage map was constructed from a peach Ferjalou Jalousia® × Fantasia (J×F) F2 population. Ferjalou Jalousia® is a flat low-acidity clingstone peach, and Fantasia is a round, normally acidic freestone peach. This population is segregated for six Mendelian characters: pollen sterility, peach or nectarine fruit, flat or round fruit, clingstone, or freestone fruit. It also segregates for the D major gene controlling the fruit’s low acidity. A new character is reported here for the first time that segregates as a Mendelian character: trees bearing aborting fruits. These trees have flowers, but fruits start to fall 2 months after blooming. This recessive character has been named Af. We demonstrate that it is linked to the flat shape of the fruit. The previous map obtained from this cross was constructed using 63 individuals, whereas the present map was constructed using 207 individuals. Moreover, 82 simple-sequence repeat (SSR) markers, including 10 expressed sequence tag-SSRs, and 43 amplified fragment length polymorphism (AFLP) markers were added. Molecular markers linked to the six Mendelian characters were identified, and one of them has already been used for marker-assisted selection. This map will be used for detection of quantitative trait loci controlling organoleptic and nutritional fruit quality in peach.  相似文献

 Mono- and sesquiterpenes are good markers for distinguishing conifer species one from another, and provenances of the same species. In the genus Cedrus, the identification of C. libani requires C. libani to be distinguished from the main provenances of C. atlantica; two provenances of C. libani were recognized. Consequently it was decided to look for terpene markers among samples from six provenances of these two species. Longifolene appears as a specific marker of C. libani Pozanti; C. libani Barouk is less easy to identify by the presence of bornyl acetate, because one out of three analysed individuals does not contain it. Factorial Discriminant Analysis underlines the differences between C. atlantica and C. libani, according to one axis (β-myrcene/α- and β-pinene). The second axis allows Barouk and Pozanti provenances of C. libani to be distinguished by their limonene and β-pinene contents. In C. atlantica, the same procedure allows Algerian and Moroccan provenances to be separated. The Mann-Whitney test confirms that C. libani Pozanti is always specified by the lowest values of β-myrcene. Similarly, C. libani Barouk′s high β-pinene content differentiates it from various provenances of C. atlantica, except in the case of some individuals (Talassentane provenance). The filamentous fungus Penicillium funiculosum produces a range of glycoside hydrolases (GH). The XynD gene, encoding the sole P. funiculosum GH10 xylanase described so far, was cloned into the pPICZαA vector and expressed in methylotrophe yeast Pichia pastoris, in order to compare the results obtained with the P. funiculosum GH11 xylanases data.  相似文献
