The distribution of beta-galactosidase activity was studied in different reproductive organs, seminal plasma and spermatozoa of the bull. The highest specific activity of beta-galactosidase was found in testis and in different parts of the epididymis, where the activity seemed to be partly in secretory (cauda secretion) and partly in non-secretory, bound form (caput to cauda epididymidis). Gel filtration on Sepharose 6B at pH 7.0 revealed two beta-galactosidase forms (GF-1, Mr approximately 500,000-600,000 and GF-2, Mr approximately 190,000-220,000) in reproductive organs and seminal plasma. The pH-optimum of both beta-galactosidase forms was about 3.75-4.75. Hg2+ and p-chloromercuribenzoate inhibited strongly these activities. Further, form GF-2 seemed to be slightly more sensitive to the thermal inactivation at 50-70 degrees C than form GF-1. In chromatofocusing beta-galactosidase activities in bull seminal plasma coeluted with those of the cauda epididymidis (pI-values 7.5-6.4). On the contrary, prostate, Cowper's gland, testis, ampulla and seminal vesicles had enzyme activities eluting at lower pI-values (6.3-4.2). Thus, the seminal plasma activity is mainly an indicator for the function of the epididymal cauda.  相似文献

When monocytes isolated from human blood adhere to glass substratum, actin- and vinculin-containing punctate plaques rapidly appear at the ventral surface of the cells. We show here that highly purified human leukocyte interferon (IFN) can inhibit formation of these adhesion plaques in a dose-dependent manner. Complete inhibition was obtained when 300 IU/ml IFN were added into the cell-seeding medium. Plaques already formed in the absence of IFN were only partially affected by subsequent addition of IFN into the culture medium. Prevention by IFN of the formation of the adhesion plaques was associated with loosened attachment of the cells to the substratum. Effect of IFN on cellular morphology was complex. At higher doses, IFN added to the cultures within 24 h of seeding almost completely inhibited the differentiation of monocytes to macrophages and most of the cells remained rounded. At lower doses, however, an enhancement of the bipolar spreading was seen and the end result was a culture with predominantly elongated fibroblastoid cells. The latter cells, unlike the fibroblastoid cells in untreated monocyte-macrophage cultures, were completely devoid of the actin plaques, while the reorganization of vimentin-type intermediate filaments took place in a normal manner. These results further support the view that the actin- and vinculin-containing plaques have a role in mediating firm adherence of human monocytes to growth substratum.  相似文献

Summary The results reported in this paper substantiate Vilter's (1968) hypothesis that the absence of basophilia of acid mucosubstances after a so-called histochemical methylation with acidified methanol may be due to, among other factors, the lactonisation of carboxyl groups rather than, as is generally held, their esterification. Sections from several fixed tissues of Syrian hamster and mouse were treated at 60 ° or at room temperature with dry methanol, ethanol, butan-l-ol, benzene or cyclohexane containing 0.35% hydrogen chloride or 1% thionyl chloride. Others were treated with methanolic methyl iodide or etheral diazomethane. With most of these solutions, the azurophilia attributable to mucosubstance carboxyl and sulphate half-ester groups was eventually abolished; subsequent saponification restored only the azurophilia due to carboxyl groups. However, some variation was found for each methylating agent, and also between different mucosubstances.  相似文献

Summary Fractionation of the rat ovarial tissue homogenate was performed using gel filtration on Sephadex G 200 and by starch gel electrophoresis. The activities hydrolysing l-leucyl--naphthylamide (Leu--NA) and dl-alanyl--naphthylamide (Ala--NA) were determined and partially characterized. Leu--NA was hydrolysed by four separate enzyme activities separated by both methods. Two of them were thiol-activated, one metal-activated and inhibited by EDTA. One was affected by neither metal chelators nor by sulfhydryl reagents. Ala--NA was hydrolysed by the three first-mentioned activities, but not by the last one. In addition, Ala--NA was hydrolysed by two other activities which were totally inhibited by metal chelators. These were clearly separated only using starch gel electrophoresis. The possibilities for the histochemical demonstration of these activities are discussed.  相似文献

gamma-Glutamyl transpeptidase (gamma-GT), its substrate (GSH) and hydrolytic product (L-glutamic acid) were measured biochemically in mouse reproductive tissues. The epididymal caput and seminal vesicles showed the highest specific activities of gamma-GT, while GSH and L-glutamic acid were widely distributed in all tissues. Histochemically, gamma-GT displayed a strong apical and supranuclear reaction and a moderate basal activity in the ductuli efferents, a weak luminal reaction in the first, a moderate apical reaction in the second and a strong apical and supranuclear reaction in the third segment of the epididymal caput. In the epididymal corpus and cauda, the gamma-GT reaction was confined to the tubular lumina but an apical reaction was also present in the cauda. The daily administration of acivicin (12 mg/kg body weight), an irreversible inhibitor of gamma-GT, for 14 days resulted in a 60% suppression of the enzyme activity in the epididymal caput, while the gamma-GT inhibition in the kidney was greater than 95%. The treatment caused no change in the activity of alanyl aminopeptidase. Histochemically, the basal and supranuclear gamma-GT activities in the ductuli efferents and the third epididymal segment were suppressed, but the apical reactions were maintained. The in-vivo suppression of epididymal gamma-GT activity may have implications in the control of post-testicular sperm maturation.  相似文献

Changes in total activity, heart and ventilation rates were observed in 2-year-old brown trout, following handling stress, using non-contact bioelectronic monitoring equipment. Experiments were carried out in laboratory conditions at water temperatures below 4° C, Transfer between tanks as well as 5 min restraint stress increased the total activity of fish for 24 to 48 h, after which it declined to near the pre-stress level. The transfer and struggle both elevated the heart rate for 3 to 4 days. Ventilation rate was elevated to a maximum of about 30% above the nominal level and recovered within 3 to 4 days. Both heart and ventilation rates were higher in feeding fish relative to fasting fish after stress and rates remained higher throughout a 7 day period of recovery. A diel rhythm of lower rates during the night appeared in both heart and ventilation rates within 3 to 4 days after handling stress.  相似文献

The molds Botrytis cinerea, Cladosporium cladosporioides, and the yeast Aureobasidium pullulans, isolated from the leaves of three short-rotation Salix clones, were found to produce indole-3-acetic acid (IAA). Abscisic acid (ABA) production was detected in B. cinerea. The contents of IAA and ABA in the leaves of the Salix clones and the amounts of fungal propagules in these leaves were also measured, in order to evaluate whether the amounts of plant growth regulators produced by the fungi would make a significant contribution to the hormonal quantities of the leaves. The content of ABA, and to a lesser degree that of IAA, showed a positive correlation with the frequency of infection by the hormone-producing organisms. The amounts of hormone-producing fungi on leaves that bore visible colonies were, however, not sufficiently high to support the claim that either the fungal production of ABA or IAA would significantly contribute to the hormonal contents of the leaves of the Salix clones. The genomes of four Lactobacillus delbrueckii subsp. lactis bacteriophages were characterized by restriction endonuclease mapping, Southern hybridization, and heteroduplex analysis. The phages were isolated from different cheese processing plants in Finland between 1950 and 1972. All four phages had a small isometric head and a long noncontractile tail. Two different types of genome (double-stranded DNA) organization existed among the different phages, the pac type and the cos type, corresponding to alternative types of phage DNA packaging. Three phages belonged to the pac type, and a fourth was a cos-type phage. The pac-type phages were genetically closely related. In the genomes of the pac-type phages, three putative insertion/deletions (0.7 to 0.8 kb, 1.0 kb, and 1.5 kb) and one other region (0.9 kb) containing clustered base substitutions were discovered and localized. At the phenotype level, three main differences were observed among the pac-type phages. These concerned two minor structural proteins and the efficiency of phage DNA packaging. The genomes of the pac-type phages showed only weak homology with that of the cos-type phage. Phage-related DNA, probably a defective prophage, was located in the chromosome of the host strain sensitive to the cos-type phage. Thymoquinone (TQ), obtained from black cumin (Nigella sativa), is a natural product with anti-oxidant, anti-inflammatory, and hepatoprotective effects but unfortunately with poor bioavailability. Aiming to improve its poor oral bioavailability, TQ-loaded nanostructured lipid carriers (NLCs) were prepared by high-speed homogenization followed by ultrasonication and evaluated in vitro. Bioavailability and pharmacodynamic studies were also performed. The resultant NLCs showed poor physical homogeneity in Compritol 888 ATO Pluronic F127 system which consequently produced larger particle size and polydispersity index, smaller zeta potential values, and lower short-term (30 days) physical stability than other systems. Encapsulation efficiency percentage (EE%) lied between 84.6?±?5% and 96.2?±?1.6%. TQ AUC0–t values were higher in animals treated with NLCs, with a relative bioavailability of 2.03- and 3.97-fold (for F9 and F12, respectively) higher than TQ suspension, indicating bioavailability enhancement by NLC formulation. Hepatoprotective effects of F12 showed significant (P?<?0.05) decrease in both serum alanine amino transferase and aspartate amino transferase to reach 305.0?±?24.88 and 304.7?±?23.55 U/ml, respectively, when compared with untreated toxic group. Anti-oxidant efficacy of F12 showed significant (P?<?0.05) decline of malondialdehyde and elevation of reduced glutatione. This improvement was also confirmed histopathologically.  相似文献
