The axenic strain BOT-22 (race B) of Botryococcus braunii was investigated for the influence of iron and glucose on its morphology to reveal the relationship with growth and oil production. The average size of the iron-rich cells was larger than that of iron-limited cells. The shapes of iron-rich cells were elliptical and that of iron-limited cells were conical. Several cells were tightly connected to form large colonies in the iron-rich culture; however, only a few cells were sparsely arranged to form the small colonies in the iron-limited culture. Glucose addition to the iron-rich culture further increased the size of the cells and colonies. The growth increased in the following order: iron-limited, iron-rich, and iron-rich with glucose cultures. The same was observed for the volume of hydrocarbons in iron-rich cultures. It was also speculated that the shapes of cells and the amount of extracellular oil are most likely related to colony size and form.  相似文献

The lateral water movement in the intact stem of a transpiring soybean plant was analyzed quantitatively by a real-time measurement system utilizing labeled water, H(2)(15)O and gamma ray detectors. A large volume of water escaping from xylem vessels during its transport was detected. The escape of water was not influenced by evaporation from the stem surface or mass flow in the sieve tubes. It was assumed that the total amount of water transported through xylem vessels was kept almost completely constant along the internode. As a result, most of the escaped water was found to re-enter the xylem vessels, i.e. water exchange occurred. The analysis of radiographs of tritiated water suggested that the self-diffusion effect of water was strong for lateral water movement, although another driving force besides thermal motion was included in the process, and that the process was also affected by the water permeability of the plasma membrane. An analysis based on a mathematical model showed that the net volume of water which escaped from xylem vessels was not dependent on the transpiration rate of the plant.  相似文献

The occurrence of radiocesium in food has raised sharp health concerns after nuclear accidents. Despite being present at low concentrations in contaminated soils (below μm ), cesium (Cs+) can be taken up by crops and transported to their edible parts. This plant capacity to take up Cs+ from low concentrations has notably affected the production of rice (Oryza sativa L.) in Japan after the nuclear accident at Fukushima in 2011. Several strategies have been put into practice to reduce Cs+ content in this crop species such as contaminated soil removal or adaptation of agricultural practices, including dedicated fertilizer management, with limited impact or pernicious side‐effects. Conversely, the development of biotechnological approaches aimed at reducing Cs+ accumulation in rice remain challenging. Here, we show that inactivation of the Cs+‐permeable K+ transporter OsHAK1 with the CRISPR‐Cas system dramatically reduced Cs+ uptake by rice plants. Cs+ uptake in rice roots and in transformed yeast cells that expressed OsHAK1 displayed very similar kinetics parameters. In rice, Cs+ uptake is dependent on two functional properties of OsHAK1: (i) a poor capacity of this system to discriminate between Cs+ and K+; and (ii) a high capacity to transport Cs+ from very low external concentrations that is likely to involve an active transport mechanism. In an experiment with a Fukushima soil highly contaminated with 137Cs+, plants lacking OsHAK1 function displayed strikingly reduced levels of 137Cs+ in roots and shoots. These results open stimulating perspectives to smartly produce safe food in regions contaminated by nuclear accidents.  相似文献

The circumnutation of a rice root under dark conditions was observed using a highly sensitive camera, a new super-HARP camera. A rice root showed regular rhythmic movement with fixed angle. When treated with Al (5 microM AlCl3), the rotation angle of the root tip was drastically decreased and then the movement was resumed again, whereas the root elongation rate was constant. With the increase of Al concentration, the cycle-fading period became shorter. This is the first report to show that an Al treatment ceased the rotation movement of the root but not elongation.  相似文献

Ionic nutrition is essential for plant development. Many techniques have been developed to image and (or) measure ionic movement in plants. Nevertheless, most of them are destructive and limit the analysis. Here, we present the development of radioisotope imaging techniques that overcome such restrictions and allow for real-time imaging of ionic movement. The first system, called macroimaging, was developed to visualize and measure ion uptake and translocation between organs at a whole-plant scale. Such a device is fully compatible with illumination of the sample. We also modified fluorescent microscopes to set up various solutions for ion uptake analysis at the microscopic level. Both systems allow numerical analysis of images and possess a wide dynamic range of detection because they are based on radioactivity.  相似文献

The green colonial alga Botryococcus braunii is characterized by the ability to produce and accumulate large amounts of hydrocarbons. We isolated and established an axenic clonal strain of B. braunii B70 and investigated the effects of organic carbon sources, including glucose, mannose, fructose, galactose, or acetate, on growth under light and dark conditions. This algal strain had the capacity to grow photo-, mixo-, or heterotrophically. Growth was promoted substantially following exposure of the algae to glucose or mannose under light exposure. Cells could grow under continuous darkness with glucose or mannose. In the presence of glucose under light or dark conditions, cell and colony size, and the intracellular granules containing oil, were markedly larger than those cultured without glucose.  相似文献

After the accident of the Fukushima 1 Nuclear Power Plant in March 2011, radioactive cesium was released and paddy fields in a wide area including Fukushima Prefecture were contaminated. To estimate the levels of radioactive Cs accumulation in rice produced in Fukushima, it is crucial to obtain the actual data of Cs accumulation levels in rice plants grown in the actual paddy field in Fukushima City. We herein conducted a two-year survey in 2011 and 2012 of radioactive and non-radioactive Cs accumulation in rice using a number of rice cultivars grown in the paddy field in Fukushima City. Our study demonstrated a substantial variation in Cs accumulation levels among the cultivars of rice.  相似文献
