A series of excellent hydrogels were prepared from poly(vinyl alcohol) (PVA) and carboxymethylated chitosan (CM-chitosan) with electron beam irradiation (EB) at room temperature. Electron spectroscopy analysis of the blend hydrogels revealed that good miscibility was sustained between CM-chitosan and PVA. The properties of the prepared hydrogels, such as the mechanical properties, gel fraction and swelling behavior were investigated. The mechanical properties and equilibrium degree of swelling improved obviously after adding CM-chitosan into PVA hydrogels. The gel fraction determined gravimetrically showed that a part of CM-chitosan was immobilized onto PVA hydrogel. The further analyses of FTIR and DSC spectra of the prepared gels after extracting sol manifested that there was a grafting interaction between PVA and CM-chitosan molecules under irradiation. The antibacterial activity of the hydrogels against Escherichia coli was also measured via optical density method. The blend hydrogels exhibited satisfying antibacterial activity against E. coli, even when the CM-chitosan concentration was only 3 wt%.  相似文献

[11C]Methionine was supplied through barley roots and the 11C signal was followed for 90 min using a real-time imaging system (PETIS), with subsequent development of autoradiographic images of the whole plant. In all cases, [11C]methionine was first translocated to the 'discrimination center', the basal part of the shoot, and this part was most strongly labeled. Methionine absorbed by the roots of the plants was subsequently translocated to other parts of the plant. In Fe-deficient barley plants, a drastic reduction in [11C]methionine translocation from the roots to the shoot was observed, while a greater amount of 11C was found in the leaves of Fe-sufficient or methionine-pretreated Fe-deficient plants. Treatment of Fe-deficient plants with aminooxyacetic acid, an inhibitor of nicotianamine aminotransferase, increased the translocation of [11C]methionine to the shoot. The retention of exogenously supplied [11C]methionine in the roots of Fe-deficient barley indicates that the methionine is used in the biosynthesis of mugineic acid phytosiderophores in barley roots. This and the absence of methionine movement from shoots to the roots suggest that the mugineic acid precursor methionine originates in the roots of plants.  相似文献

An unknown polyacetylene was isolated from the subterranean stems of Solidago altissima L. in which the two poly acetylenes, dehydromatricaria ester (I) and methyl 10-[(Z)-2-methyl- 2-butenoyloxy]-(2Z,8Z)-2,8-decadiene-4,6-diynoate (II) had already been found, and its structure was identified as (4Z)-2,4-decadiene-6,8-diyn-4-olide (dehydromatricaria lactone) (III). The lactone (III), as well as I, strongly inhibited the growth of the seedlings of barnyard millet (Panicum crus-galli L. var. frumentaceum Trin.).

The seasonal variations in the polyacetylene contents of the subterranean stems were closely investigated, with the results that III occurred only in the fall of the year while I and II hardly varied throughout the year. II was a major polyacetylene component and the content was 0.9~1.0 µmol/g fresh wt.  相似文献

The irradiation effects on histamine in fish meals and fish solubles for animal feeds and in aqueous solution were investigated. The amount of histamine in fish meal (0.4mg/100g) was not changed by up to 5.0 Mrad (50kGy), while the histamine content in fish soluble (45.7 mg/100 g) decreased ca. 16% at 5.0 Mrad. There occurred no accumulation of histamine by irradiation or during storage.

The G values of histamine formation from histidine solution and histamine decomposition by irradiation in deoxygenated neutral solutions were 0.077 and 1.90, respectively. Oxygen inhibited the formation of histamine from histidine but it had little effect on histamine decomposition. Nitrous oxide accelerated the decomposition of histamine and inhibited the accumulation of histamine in histidine solution by irradiation. These results show that there is no problem of histamine accumulation in feedstuffs caused by irradiation since the decomposition of histamine is preferred to its formation.  相似文献

The upper part of a nodulated soybean root hydroponically cultured in a glass bottle was monitored using a computer microscope under controlled environmental conditions, and the diameter of individual nodules was measured from 10-24 d after planting. The diameter of a root nodule attached to the primary root increased from 1 mm to 6 mm for 2 weeks under nitrogen-free conditions. The increase in diameter of the nodules was almost completely stopped after 1 d of supplying 5 mM nitrate, and was due to the cessation of nodule cell expansion. However, nodule growth quickly returned to the normal growth rate following withdrawal of nitrate from the solution. The reversible depression of nodule growth by nitrate was similar to the restriction of photoassimilate supply by continuous dark treatment for 2 d followed by normal light/dark conditions. In addition, the inhibitory effect of nitrate was partially alleviated by the addition of 3% (w/v) sucrose to the medium. Plant leaves were exposed to (11)C or (14)C-labelled carbon dioxide to investigate the effects of 5 mM nitrate on the translocation and distribution of photosynthates to nodules and roots. Supplying 5 mM nitrate stimulated the translocation rate and the distribution of labelled C in nitrate-fed parts of the roots. However, the (14)C partitioning to nodules decreased from 9% to 4% of total (14)C under conditions of 5 mM nitrate supply. These results indicate that the decrease in photoassimilate supply to nodules may be involved in the quick and reversible nitrate inhibition of soybean nodule growth.  相似文献

The effect of radiation pasteurization of sugar cane bagasse and rice straw and fermentation using various strains of fungi were studied for upgrading of cellulosic wastes. The initial contamination by fungi and aerobic bacteria both in bagasse and straw was high. The doses of 30 kGy for sterilization and 8 kGy for elimination of fungi were required. Irradiation effect showed that rice straw contained comparatively radioresistant microorganisms. It was observed that all the fungi (Hericium erinacium, Pleurotus djamor, Ganoderma lucidum, Auricularia auricula, Lentinus sajor-caju, Coriolus versicolor, Polyporus arcularius, Coprinus cinereus) grow extending over the entire substrates during one month after inoculation in irradiated bagasse and rice straw with 3% rice bran and 65% moisture content incubated at 30°C. Initially, sugar cane bagasse and rice straw substrates contained 39.4% and 25.9% of cellulose, 22.9% and 26.9% of hemicellulose, and 19.6% and 13.9% of lignin + cutin, respectively. Neutral detergent fibre (NDF) values decreased significantly in sugar cane bagasse fermented byG. lucidum, A. auricula andP. arcularius, and in rice straw fermented by all the 8 strains of fungi. Acid detergent fibre (ADF) values also decreased in bagasse and rice straw fermented by all the fungi.P. arcularius, H. erinacium, G. lucidum andC. cinereus were found to be the most effective strains for delignification of sugar cane bagasse.  相似文献

Nitrogen movement through the xylem vessels and sieve tubes in rice plants was studied using xylem and phloem sap analysis in combination with stable and radioactive nitrogen isotope techniques.More than 90% of nitrogen was translocated in the sieve tubes of rice plants as amino acids. When 15N (99.6 atom%) was applied as a nitrate to the root, 15N first appeared in phloem sap of the leaf sheath within 10 minutes and increased to 37 atom% excess 5 hours after the experiment had started. In long-term experiments, 63% of nitrogen in the phloem sap of the leaf sheath and 15% in that of the uppermost internode came from nitrogen absorbed within the last 24 hours and 50 hours, respectively.To obtain information about the more rapid circulation of nitrogen in the plant, radioactive 13N was used as a tracer. A positron-emitting tracer imaging system was used to show that 13N was transferred to the leaf sheath within 8 minutes of its application to the roots. Analysis of the xylem sap of the leaf sheath showed that when the nitrate was applied to the roots, most of the nitrogen in the xylem was transported as a nitrate.These data showed that phloem and xylem sap analysis together with the stable and radioactive nitrogen techniques provide a good method for the detection of nitrogen cycles in plants.  相似文献

The lelvels of seven heavy metals and their toxicity towardGanoderma lucidum under various cultivation conditions were assessed. The contents of Mn, Cu, Zn, Cd, Hg, Pb and U in the fruitbodies of cultivatedG. lucidum, and sawdust substrates were determined to be at trace levels for U, 0.01–0.1 μg/g for Cd and Hg, and 1–5 μg/g for Pb, 10–120 μg/g for Mn, Cu and Zn. The effects of heavy metals, on the growth of mycelia ofG. lucidium in pure cultures were examined over a wide range of concentrations (10–3,000 μg/ml), and their toxicities were found to decrease in the order: Hg>Cd>Cu>U>Pb>Mn=Zn. The translocation and accumulation of Zn from contaminated substrates (at 10 μg/g) in fruitbodies were investigated by using65Zn tracer, andG. lucidum was found to take up Zn with an efficiency of >60%, leading to accumulation of >100 μ/g, in fruitbodies and >80 μ/g Zn in basidiospores.  相似文献

Responses of mycelia ofGanoderma lucidum to vanadium, selenium and germanium were examined over a wide range of concentrations (10–1, 120 μg/ml) in pure culture. Se and V were found to be highly toxic, but Ge was not toxic at the levels tested.Ganododerma lucidum cultivated on substrates of sawdust with V (30–80 μg/g) developed mature fruitbodies, but the bioaccumulation of V was quite low (2.5–7 μg/g in pileus, 12.5–21.5 μg/g in stipe and <1 μg/g in basidiospores). Se as Na2SeO4 labeled with75Se was effectively taken up from substrates and accumulated in fruitbodies (mainly in pileus), then depleted by discharge of basidiospores. Ge as GeCl4 labeled with77Ge was easily uptaken and translocated into fruitbodies.  相似文献
