Reproduction of Japanese encephalitis virus in human glioma cells, 118MGC: inhibitory effect of host factor(s) 总被引:1,自引:0,他引:1
Japanese encephalitis viruses (JEV) were well propagated in human glioma cells, 118MGC until the first 24 hrs after virus infection. However, after 24 hrs, virus growth rate was quickly reduced. This unusual pattern of virus growth was different from the cases in others cells, e.g. IMR-32, Vero and C6/36 cells. The fact that actinomycin-D retained the high yields of JEV in 118MGC cells suggests that some suppressing factors against JEV replication are produced in MGC cells. Interestingly, culture fluids of 118MGC cells indicated inhibitory effect to JEV reproduction, but other culture fluids from several cell lines had no effect. This inhibitory effect of the MGC-culture fluids was lost by heat-treatment at 60 C. In addition, the infectivity of JEV was rapidly decreased by the incubation with MGC-culture fluids. These findings suggest that 118MGC cells produce and secret some inhibitory factors against JEV replication. 相似文献

The antibiotic cordycepin (3'-deoxyadenosine), a known specificinhibitor of nucleic acid synthesis and polyadenylation of RNA,remarkably retarded decrease in the chlorophyll and proteincontents of senescing tobacco leaf disks. The effectivenessof cordycepin, at its optimum concentration (ca. 4 x 105M), was 53% as effective as a cytokinin, benzyladenine at 106M. Adenosine and 2'-deoxyadenosine had no anti-senescence action. The rate and period of DNA synthesis during meiotic prophasewere examined using lily microsporocytes. Meiocytes at the earlyleptotene stage were cultured for discrete periods in the presenceof inhibitors of DNA synthesis, deoxyadenosine and nalidixicacid. Deoxyadenosine, which arrests meiotic development at theearly zygotene stage, markedly suppressed DNA synthesis to 35%of control at 2 mM. Nalidixic acid simply reduced the rate ofDNA synthesis, resulting in prolongation of the synthetic period.The relevance of DNA synthesis to meiotic development is discussed. In tobacco leaf disks floated on water in the dark, chloroplastrRNA (23S and 16S rRNA) decreased rapidly, and this decreasewas inhibited by 106M BA. The cytoplasmic rRNA (25S and18S rRNA) level changed little during a few days of dark incubationregardless of the presence or absence of BA. Cycloheximide at105 M completely removed the effect of BA. The high incidence of progressive multifocal leukoencephalopathy (PML) in AIDS patients compared with many other immunosuppressive diseases suggests that HIV-1 infection is strictly related to the activation of JC virus (JCV) propagation. In this report, propagation of PML-type JCV in COS-7-derived cell lines stably expressing HIV-1 Tat (COS-tat cells) has been examined. In COS-tat cells, production of viral particles and replication of genomic DNA were markedly increased compared to COS-7 cells, as judged by HA and real-time PCR analyses. These results demonstrate that COS-tat cells provide a useful model system for studying HIV-1 Tat-mediated propagation of PML-type JCV. 相似文献

The sea star Astropecten scoparius has feeding bipinnarian larvae, whereas its congener Astropecten latespinosus has nonfeeding barrel-shaped larvae. To investigate evolutionary changes in the development of asteroids, we performed reciprocal crosses between these two species with different larval forms. In the cross between A. scoparius eggs and A. latespinosus sperm, embryos developed into bipinnaria-like larvae. The larvae exhibited either a functional digestive system (a maternal feature) or a nonfunctional digestive system with the tip of the archenteron not connected to the stomodeum (a paternal characteristic). However, in the reciprocal cross between A. latespinosus eggs and A. scoparius sperm, barrel-shaped larvae resembling those of A. latespinosus were produced, in addition to bipinnaria-like larvae, some with functional digestive systems and some with nonfunctional ones. Juveniles were produced from all types of crosses. 18S rDNA was used as a gene marker in cycle sequencing analysis to investigate the genetic features of these juveniles. The sequences of juveniles from bipinnaria-like larvae showed double-peak nucleotide signals, indicating a biparental genome. On the other hand, juveniles from barrel-shaped larvae from A. latespinosus eggs and A. scoparius sperm showed the same sequence as A. latespinosus juveniles. This suggests that bipinnaria-like larvae of both crosses are always hybrids, whereas barrel-shaped larvae develop parthenogenetically. 相似文献
