Background

Polyporus umbellatus sclerotia have been used as a diuretic agent in China for over two thousand years. A shortage of the natural P. umbellatus has prompted researchers to induce sclerotial formation in the laboratory.

Methodology/Principal Finding

P. umbellatus cultivation in a sawdust-based substrate was investigated to evaluate the effect of low temperature conditions on sclerotial formation. A phenol-sulfuric acid method was employed to determine the polysaccharide content of wild P. umbellatus sclerotia and mycelia and sclerotia grown in low-temperature treatments. In addition, reactive oxygen species (ROS) content, expressed as the fluorescence intensity of mycelia during sclerotial differentiation was determined. Analysis of ROS generation and sclerotial formation in mycelia after treatment with the antioxidants such as diphenyleneiodonium chloride (DPI), apocynin (Apo), or vitamin C were studied. Furthermore, macroscopic and microscopic characteristics of sclerotial differentiation were observed. Sclerotia were not induced by continuous cultivation at 25°C. The polysaccharide content of the artificial sclerotia is 78% of that of wild sclerotia. In the low-temperature treatment group, the fluorescent intensity of ROS was higher than that of the room temperature (25°C) group which did not induce sclerotial formation all through the cultivation. The antioxidants DPI and Apo reduced ROS levels and did not induce sclerotial formation. Although the concentration-dependent effects of vitamin C (5–15 mg mL−1) also reduced ROS generation and inhibited sclerotial formation, using a low concentration of vitamin C (1 mg mL−1) successfully induced sclerotial differentiation and increased ROS production.

Conclusions/Significance

Exposure to low temperatures induced P. umbellatus sclerotial morphogenesis during cultivation. Low temperature treatment enhanced ROS in mycelia, which may be important in triggering sclerotial differentiation in P. umbellatus. Moreover, the application of antioxidants impaired ROS generation and inhibited sclerotial formation. Our findings may help to provide new insights into the biological mechanisms underlying sclerotial morphogenesis in P. umbellatus.  相似文献

基于40号标本的形态研究,对中国小头蛇属Oligodon进行分类修订。四线小头蛇Oligodon taeniatus(Güther,1861)在中国没有分布,当初发表该纪录所依据的3号标本为紫棕小头蛇指名亚种Oligodon cinereus cinereus(Güther,1864);中国记录的管状小头蛇Oligodon cyclurus(Cantor,1839)应订正为束纹小头蛇Oligodon fascioatus(Güther,1864)。根据色斑、鳞被和半阴茎等综合特征,提出中国记录的喜山小头蛇Oligodon albocinctus包括2个物种,其中云南省陇川县标本为喜山小头蛇Oligodon albcinctus(Cantor,1839),而西藏墨脱县标本是尚未明确的小头蛇属未定种Oligodonsp.,其分类地位有待进一步研究。最后,提供中国分布的小头蛇属物种检索表。  相似文献

目的探讨α2b-IFN对PBMC内CD25的诱导及对HCV-RNA、抗-HCV转阴效果。方法用BSA法检测慢性丙肝患者治疗前后CD25表达水平。以α2b-IFN(3 MU/d)治疗3个月为一疗程,共2个疗程,于治疗前后分别检测患者PBMC内HCV-RNA和血清HCV-RNA、抗-HCV。结果慢性丙肝患者α2b-IFN治疗后静息期、诱导期CD25表达水平分别为(3.44±0.77)%、(33.62±3.95)%,PBMC、血清内HCV-RNA和抗-HCV转阴率分别为42.31%(11/26)、57.69%(15/26)和65.38%(17/26),与对照相比差异有显著性(P<0.01~P<0.05)。结论α2b-IFN可诱导CD25表达,对PBMC内HCV-RNA具有肯定的治疗作用,其疗效优于常规治疗。  相似文献

Nicotiana sect. Repandae is a group of four allotetraploid species originating from a single allopolyploidisation event approximately 5 million years ago. Previous phylogenetic analyses support the hypothesis of N. nudicaulis as sister to the other three species. This is concordant with changes in genome size, separating those with genome downsizing (N. nudicaulis) from those with genome upsizing (N. repanda, N. nesophila, N. stocktonii). However, a recent analysis reflecting genome dynamics of different transposable element families reconstructed greater similarity between N. nudicaulis and the Revillagigedo Island taxa (N. nesophila and N. stocktonii), thereby placing N. repanda as sister to the rest of the group. This could reflect a different phylogenetic hypothesis or the unique evolutionary history of these particular elements. Here we re-examine relationships in this group and investigate genome-wide patterns in repetitive DNA, utilising high-throughput sequencing and a genome skimming approach. Repetitive DNA clusters provide support for N. nudicaulis as sister to the rest of the section, with N. repanda sister to the two Revillagigedo Island species. Clade-specific patterns in the occurrence and abundance of particular repeats confirm the original (N. nudicaulis (N. repanda (N. nesophila + N. stocktonii))) hypothesis. Furthermore, overall repeat dynamics in the island species N. nesophila and N. stocktonii confirm their similarity to N. repanda and the distinctive patterns between these three species and N. nudicaulis. Together these results suggest that broad-scale repeat dynamics do in fact reflect evolutionary history and could be predicted based on phylogenetic distance.  相似文献

A plant-specific gene was cloned from melon fruit. This gene was named downward leaf curling (CmDLC) based on the phenotype of transgenic Arabidopsis plants overexpressing the gene. This expression level of this gene was especially upregulated during melon fruit enlargement. Overexpression of CmDLC in Arabidopsis resulted in dwarfism and narrow, epinastically curled leaves. These phenotypes were found to be caused by a reduction in cell number and cell size on the adaxial and abaxial sides of the epidermis, with a greater reduction on the abaxial side of the leaves. These phenotypic characteristics, combined with the more wavy morphology of epidermal cells in overexpression lines, indicate that CmDLC overexpression affects cell elongation and cell morphology. To investigate intracellular protein localization, a CmDLC-GFP fusion protein was made and expressed in onion epidermal cells. This protein was observed to be preferentially localized close to the cell membrane. Thus, we report here a new plant-specific gene that is localized to the cell membrane and that controls leaf cell number, size and morphology.  相似文献

The impact of agricultural land use on the composition and structure of aquatic insect assemblages (i.e., taxa of Ephemeroptera, Plecoptera, Trichoptera, and Coleoptera (EPTC)) was investigated in tributary streams of the Garonne river basin, southern France. The self-organizing map (SOM) method was applied to compare both instream environmental conditions and EPTC assemblages between forest and agricultural streams. According to the SOM model, the study sites were classified into three main clusters corresponding to distinct EPTC assemblages. The SOM cluster associated with most of the agricultural sites had lower EPTC species richness and diversity. This cluster was also characterized by high levels of total dissolved solids, nitrate (NO3), and chemical oxygen demand. Overall, our study shows that agricultural streams when compared with forest streams had lower biological integrity. In accordance with the European Water Framework Directive, our results indicate that the sites most impacted by agricultural land use should be restored and that the least-impacted forest sites could serve as reference conditions.  相似文献

Secreted proteins (secretomes) play crucial roles during bacterial pathogenesis in both plant and human hosts. The identification and characterization of secretomes in the two plant pathogens Burkholderia glumae BGR1 and B. gladioli BSR3, which cause diseases in rice such as seedling blight, panicle blight, and grain rot, are important steps to not only understand the disease-causing mechanisms but also find remedies for the diseases. Here, we identified two datasets of secretomes in B. glumae BGR1 and B. gladioli BSR3, which consist of 118 and 111 proteins, respectively, using mass spectrometry approach and literature curation. Next, we characterized the functional properties, potential secretion pathways and sequence information properties of secretomes of two plant pathogens in a comparative analysis by various computational approaches. The ratio of potential non-classically secreted proteins (NCSPs) to classically secreted proteins (CSPs) in B. glumae BGR1 was greater than that in B. gladioli BSR3. For CSPs, the putative hydrophobic regions (PHRs) which are essential for secretion process of CSPs were screened in detail at their N-terminal sequences using hidden Markov model (HMM)–based method. Total 31 pairs of homologous proteins in two bacterial secretomes were indicated based on the global alignment (identity ≥ 70%). Our results may facilitate the understanding of the species-specific features of secretomes in two plant pathogenic Burkholderia species.  相似文献
