This paper reports the effects of low O2 concentration (001,0055, and 0.115mol m3) in nutrient solutions onK+/Na+ selectivity of growing and mature root tissues of 6-to 8-d-old, intact, wheat (Triticum aestivum cv. Gamenya) seedlings. Increases in anaerobic catabolism and decreases in O2 uptake,K+ uptake and K+/Na+ selectivity were all more pronounced and/oroccurred at higher external O2 concentrations in the apex (02mm) than in the expanding tissues (24 mm); these growingtissues were, in turn, more affected than the expanded tissuesof the roots (412 mm). Selectivity for K+ over Na+ in roots and shoots was particularlysensitive to O2 deficiency. For example, in apical tissues (02mm) K + /Na+ selectivity was already reduced at 0.115 mol m3O2, yet at this O2 concentration there was no effect on eithergrowth or (K+/Na+) uptake. Upon transfer from 0.01 to 0.26 mol m3 O2, a detailedstudy of the 12 mm root tips showed that 70% of these tips regainedhigh (K+ + Na+) concentrations and K+/Na+ ratios. In contrast,there was no recovery in the remaining 30% of the 12 mm roottips. Net K+ transport to the shoots during the period afterre-aeration was negative for the population as a whole. Theseverity of these effects supports the view that the root tipsand the stele were more susceptible to O2 deficiency than wasthe cortex of the fully-developed root tissues. Efficiency of Selenite Cystine and TT Enrichment Broths for the Detection of Salmonella 总被引:1,自引:1,他引:0
Comparisons of selenite cystine (SC) and TT enrichment broths for detecting salmonellas were made with pure culture suspensions, with samples of naturally or artificially contaminated foods and with poultry feed. Selenite cystine recovered higher numbers of salmonellas from pure cultures and ground beef, while TT broth recovered higher numbers from pork sausage and poultry feed. Differences in the recovery of salmonellas from other food products appeared to be insignificant. The use of both SC and TT is thus recommended for maximal recovery of these organisms. 相似文献

Lanthanum fed to the base of excised leaves of Sesamum indicum L. and Helianthus annuus L. was used as a tracer to investigate by electron microscopy the path of water in the apoplast of leaves. The generally random distribution of lanthanum in cell walls provided no support for the hypothesis that cuticular transpiration may be greater for guard cells than for adjacent epidermal cells. Occasionally, accumulations of lanthanum were observed in anticlinal walls of epidermal cells and at the outer surface of the plasma membrane but lanthanum was not observed in the symplast. The influx of 86Rb to excised roots of sesame and sunflower was inhibited during incubation with 0.5 mM lanthanum or calcium for 15 or for 180 min. Stomata of sunflower partially closed when 2.5 mM lanthanum was supplied to the base of excised shoots in a potometer, whereas this treatment had little effect on stomatal conductance of sesame shoots maintained in a constant environment. Supplying 2.5 mM lanthanum to the base of sesame shoots strongly inhibited stomatal opening response to increase in ambient humidity but had little effect on stomatal opening response to light. It was concluded that stomatal opening response to increased humidity may be dependent upon some process, such as ion influx, that is inhibited by lanthanum, and that opening response to humidity may differ in mechanism from stomatal opening response to increased irradiance. 相似文献

The imposition of the stresses of climate change (higher temperatures and in many regions lower rainfall) on existing stressors, such as habitat loss and degradation, will increase pressures on native fauna already experiencing declines. We focused on assessing how the ‘Big Dry’ (severe drought, 1997–2010) in south‐eastern Australia affected populations of a small marsupial carnivore, the yellow‐footed antechinus (Antechinus flavipes), in box‐ironbark forests, which suffer a range of anthropogenic disturbances. Trapping of the mammal was conducted on 136 (0.25 ha) sites in two box‐ironbark forests in 2004, 2005 and 2011 (46 or 64 sites per year). Capture rates of all distinct individuals, males and second‐year females with suckled teats, and the number of suckled teats were positively associated with rainfall in the previous September (time of lactation and deposition of young in nests). Despite differences between forests in capture rates of all individuals, the positive effect of rainfall was evident in both forests. Populations in one forest, Chiltern, were substantially larger than other locations surveyed in 2004 and 2005, yet crashed to small numbers in 2011. This crash was most likely due to low rainfall in the preceding years including the lowest recorded annual rainfall (2006), below‐average annual rainfall (2007, 2008 and 2009) and well‐below‐average rainfall in September (2006, 2007 and 2008). The predicted drying and warming climate in south‐eastern Australia and habitat loss and degradation pose a threat to the viability of the yellow‐footed antechinus in box‐ironbark forests. An integrated approach to small‐mammal management is necessary given that the region may be facing additional losses, especially during droughts, to those already experienced since the early 1800s. There is an ongoing debate on how to correct leaf gas exchange measurements for the unavoidable diffusion leakage that occurs when measurements are done in non‐ambient CO2 concentrations. In this study, we present a theory on how the CO2 diffusion gradient over the gasket is affected by leaf‐mediated pores (LMP) and how LMP reduce diffusive exchange across the gaskets. Recent discussions have so far neglected the processes in the quasi‐laminar boundary layer around the gasket. Counter intuitively, LMP reduce the leakage through gaskets, which can be explained by assuming that the boundary layer at the exterior of the cuvette is enriched with air from the inside of the cuvette. The effect can thus be reduced by reducing the boundary layer thickness. The theory clarifies conflicting results from earlier studies. We developed leaf adaptor frames that eliminate LMP during measurements on delicate plant material such as grass leaves with circular cross section, and the effectiveness is shown with respiration measurements on a harp of Deschampsia flexuosa leaves. We conclude that the best solution for measurements with portable photosynthesis systems is to avoid LMP rather than trying to correct for the effects. 相似文献
