Changes in gonadal histology were used to determine the reproductivecycles of three trochid gastropods commonly found along theTranskei coast. They were dioecious, the sexes being evenlydistributed both in terms of numbers and size. Despite someasynchrony, Monodonta australis had a distinct reproductivecycle with a short inactive period during the winter, gonadaldevelopment coincided with increasing sea temperatures and spawningtook place from late summer to autumn. Both Oxystele tabularisand O. variegata exhibited asynchronous intermittent spawningthroughout the study period. However, intensified spawning activityby O. tabularis was noted between April and September. The reproductivecycles and mechanisms of these trochids are compared with thoseelsewhere. Molecular Characterization of a Cytoplasmic Dynein from Dictyostelium   总被引:1,自引:0,他引:1  
Cytoplasmic dynein is a high molecular weight, microtubule-based mechanochemical ATPase that is believed to provide motive force for a number of intracellular motilities, including transport of membrane-bound organelles. Cytoplasmic dynein also localizes to the mitotic spindles of some organisms and to the kinetochore regions of some condensed chromosomes, where it may play an active role in spindle assembly, spindle position, and/or chromosome movement during cell division. Despite active research efforts from a number of laboratories, little detail is yet available about dynein-based cellular activities. This paper describes our efforts to characterize cytoplasmic dynein from Dictyostelium and to use this protist as a molecular genetic factory to probe structure-function relationships of this molecule.  相似文献

The cytoskeletal events that assist restitution of the native intestinal epithelium are poorly understood. To enhance our understanding of repair mechanisms in the native intestinal epithelium we assessed the functional role of actin and the temporal and spatial alterations in actin and villin that occur in native enterocytes migrating in response to injury. Using a well-characterizedin vitroUssing chamber model of native intestine epithelial restitution, the actin inhibitor cytochalasin D (CD) was applied to determine the functional importance of actin to restitution as assessed by sensitive electrophysiological means and structural techniques. Additionally we used phalloidin and indirect immunohistochemistry to localize and semi-quantitate F-actin and villin in migrating cells during restitution. We report new data that shows that when cytoskeletal changes were impaired with CD, the epithelial monolayer was re-established in fewer than 20% of CD-treated villi, cells bordering the epithelial defect did not assume the characteristic phenotype associated with migrating cells, and transepithelial resistance did not return to pre-injury levels. F-actin and villin were present at the leading edge of the migrating cells, basolateral F-actin was decreased, and cytoplasmic villin was increased as determined by phalloidin and immunohistochemical methods. We conclude thatin vitrorepair of the native intestinal epithelium is functionally and structurally dependent on major changes in the cytoskeleton of cells involved in re-establishing the epithelial monolayer over a complex extracellular matrix.  相似文献

Viola pubescens is a perennial herb with a mixed mating system involving the production of both open, chasmogamous flowers and closed, cleistogamous flowers. Using a nonradioactive protocol, 12 microsatellite primers that amplified loci with perfect and imperfect repeats were developed in the species. As examined in five populations of V. pubescens using multiplex reactions, all loci exhibited polymorphism with an average of 5.4 alleles. Most primers also amplified in 12 additional Viola species and one cultivar, revealing the possibility of wide applicability across the genus. This is the first known report of microsatellite loci developed in Viola and the Violaceae.  相似文献

Several recent studies have found amphibian populations to be genetically highly structured over rather short geographical distances, and that the rate of genetically effective dispersal may differ between the sexes. However, apart from the common frog ( Rana temporaria ) little is known about the genetic structuring and sex-biased dispersal in northern European amphibians. We investigated the patterns of genetic diversity and differentiation within and among Scandinavian populations of the moor frog ( Rana arvalis ) using microsatellite markers. The genetic diversity within local R. arvalis populations was not a simple linear negative function of latitude but a convex one: genetic diversity peaked in mid-latitude populations, and declined thereafter dramatically towards the north. The average degree of genetic differentiation among populations ( F ST = 0.14) was lower than that observed for the common frog ( F ST = 0.21), though the pattern of isolation by distance was similar for both species. Contrary to common frogs, no evidence for female-biased dispersal was found. The results reinforce the view that amphibian populations are—in general—highly structured over relatively small geographical distances, even in comparatively recently colonized areas.  相似文献

1. Diapause is a dynamic process of low metabolic activity that allows insects to survive periods of harsh conditions. Notwithstanding the lowered metabolism, and because diapausing insects have no access to food, diapause has an energetic cost that may affect post‐diapause performance. 2. Previous studies on the solitary bee Osmia lignaria have shown that prolonged pre‐wintering periods (the time during which individuals already in diapause remain at warm temperatures) are associated with elevated lipid consumption, fat body depletion, and body weight loss. The present study investigated whether prolonged pre‐wintering also affects reproduction, i.e. whether the costs associated with diapause could have an effect on post‐diapause performance in this species. 3. Females were exposed to a range of pre‐wintering conditions, and ovary development and individual post‐wintering performance were monitored throughout their adult life span. 4. No evidence of an effect of pre‐wintering duration on post‐diapause reproductive success was found. Expected differences in the timing of establishment were not observed because ovary maturation was, surprisingly, not arrested during pre‐wintering. Prolonged pre‐wintering duration did not result in decreased life span, probably because emerging females could rapidly replenish their metabolic reserves through feeding. However, there was a very strong effect of the duration of the pre‐emergence period on the likelihood of nest establishment. 5. Longevity, the main factor determining fecundity in Osmia, is subjected to high levels of intrinsic variability, even among females of similar size exposed to identical conditions during development and nesting.  相似文献

Temporal and spatial variations in the density, size compostionand growth of exploited and non-exdploited populations of theintertidal limpet Cellana capensis from the Transkei coast ofsouthern Africa, between 1989 and 1991, are described. The timingand intensity of recruitment varied between shores and amongstsites within shores. Recruitment success was relatively poorand, in fact, at some of the protected sites no recruitmentwas recorded until July 1991. Juvenile and adult limpets wereboth significantly more abundant at the exploited than at theprotected sites. The gradual decline in density of limpets observedat the latter sites reflected poor recruitament as well as senescenceof older individuals. The extent of variations in density amongstshores was also found to vary from censius to census. Limpetsat the exploited sites tended to be much smaller than thosefound at the protected sites. Although there were initiallyno differences in the size of limpets amongst sites withingshores marked differences became apparent during the courseof the study. This finding, together with the observation ofmarked annual differences in population size compostion at severalof the study sites highlighted the temporal instability of thesepopulations. Such results have profound implications for comparativestudies of population size composition. The absence of significant differences in growth rate amongstshores suggests that differences in the intensity of competitiveinteractions may have been offset by the higher microalgal productionat the exploited shore. It is also suggested that upshore migrationof largter individuals may counter the intense competition experiencedat mid-tidal levels on the exploited shore. Differences in therate of recritment could also account for some of the observeddifferences in population characteristics. Both of these explanationsare viable alternatives to an earlier proposal which attributeddifferences solely to the effects of size-selective predationby man. Abstract.  Low temperature and desiccation stress are thought to be mechanistically similar in insects, and several studies indicate that there is a degree of cross-tolerance between them, such that increased cold tolerance results in greater desiccation tolerance and vice versa . This assertion is tested at an evolutionary scale by examining basal cold tolerance, rapid cold-hardening (RCH) and chill coma recovery in replicate populations of Drosophila melanogaster selected for desiccation resistance (with controls for both selection and concomitant starvation) for over 50 generations. All of the populations display a RCH response, and there is no effect of selection regime on RCH or basal cold tolerance, although there are differences in basal cold tolerance between sampling dates, apparently related to inter-individual variation in development time. Flies selected for desiccation tolerance recover from chill coma slightly, but significantly, faster than control and starvation-control flies. These findings provide little support for cross-tolerance between survival of near-lethal cold and desiccation stress in D. melanogaster .  相似文献
