Leaves of the tea plant photosynthesizing in 14CO2 incorporatedmuch radioactivity into intermediates of the glycolate pathwayand little into C4 acids. Increased O2 in the atmosphere decreasedphotosynthesis, stimulated photorespiration, and increased theCO2 compensation point. In air the rate of photorespirationwas 19% of net photosynthesis. Abstract. The malaria transmission potential of wild, infective Anopheles from western Kenya was evaluated by determining the number of sporozoites transmitted in vitro by salivation when their mouthparts were inserted into capillary tubes containing either sucrose or blood. With sucrose, 86.6% of 102 infective Anopheles transmitted a geometric mean (GM) of 3.84 sporozoites (range 1–34). With blood, 23.1% of 104 infective Anopheles , tested on the day of collection, transmitted a GM of 2.30 sporozoites (range 1–117). For Anopheles held 5 days postcapture before testing with blood, 53.6% of 56 transmitted a GM of 6.04 sporozoites (range 1–420). Transmitting Anopheles contained significantly more salivary gland sporozoites than non-transmitters. No significant differences were detected between Anopheles gambiae Giles sensu lato and Anopheles funestus Giles in sporozoite transmission by individuals with sporozoites in their salivary glands.
Sporozoites were detected microscopically in the salivary duct from heads in 80.3% of 117 infective Anopheles (GM=11.2, range 1–71). Sporozoite detection in mosquito heads by ELISA was 25% less efficient than microscopic detection.
Over 98% of the infective Anopheles transmitted less than twenty-five sporozoites. Transmitted sporozoites represented only about 3% of the total sporozoites in the salivary glands suggesting that sporozoite transmission may be restricted to sporozoites in the salivary duct at the time of feeding. Results are discussed in relation to anti-sporozoite vaccine development.  相似文献

The durations of the photoperiod-sensitive and photoperiod-insensitivephases of development to panicle emergence were estimated infour contrasting indica cultivars of rice (Oryza sativa L.)in a reciprocal-transfer experiment. Plants were grown in potsin glasshouses maintained at warmer (32/26 C) or cooler (28/20C) day/night temperatures, and the durations from sowing topanicle emergence were determined for plants moved from relativelyshort (11 h) to relatively long (13.5 h) days and vice versaat various times after sowing. Panicle emergence was delayedby long days in all cultivars, but the traditional cvs Carreonand Peta were much more sensitive to photoperiod than the moderncvs IR8 and IR36 The durations of the photoperiod-insensitivepre-inductive phase (equivalent to some definitions of the basicvegetative phase) varied from 14.4 d in cv. Carreon at 32/26C to 42.0 d in cv. IR8 at 28/20 C. In all cultivars this initialphase was of a longer duration in the cool than in the warmregime. The duration of the photoperiod-insensitive post-inductivephase was also consistently greater, but usually only slightso, at cool than relatively warm temperatures; it varied from6.8 d in cv. IR8 at 32/26 C to 272 d in cv. Carreon at 28/20C. As expected, the length of the intervening photoperiod-sensitiveinductive phase was greater in long days, but the effect oftemperature on these durations was not consistent; for example,these durations were longer in warm than in cool temperaturesin cv. 1R8 but, if anything, they were slightly longer in coolthan in warm temperatures in cv. IR36. This difference is compatiblewith previous findings that cv. IR36 has a warmer optimum temperaturefor rate of progress towards panicle emergence than cv. IR8.A subsequent reciprocal-transfer experiment with cv. Peta providedestimates of the durations of the photoperiod-insensitive andphotoperiod-sensitive phases of pre-flowering development whichwere compatible with our earlier estimates. Furthermore, panicleinitiation was found to occur after about 80% of the photoperiod-sensitiveinductive phase had elapsed. We conclude that although the durationof the photoperiod-insensitive pre-inductive phase in rice isgreater than in many other annual crops, genotypic variationin this duration may well be less than was previously deduced.We also conclude that, despite common assumptions to the contrary,photoperiod-sensitivity during rice plant development does notend at panicle initiation. Storage experiments were carried out on barley seed (Hordeumdistichum L.) lasting from 1 min to 926 days, including 52 hermeticstorage environments covering a range of temperatures from 3to 90 °C and 5·5 per cent to 24·6 per centmoisture content (f. wt basis). Over the entire range of conditionssurvival curves conformed to negative cumulative normal distributionsand, for any given measure of longevity, e.g. half-viabilityperiod, longevity was roughly proportional to the negative exponentof both temperature and moisture content. Although previouslyreported viability equations were adequate to describe theserelationships over restricted ranges of environments, over theextended range of conditions tested here it was shown that therelationship between log seed viability and temperature is infact slightly convex, whilst that between log seed viabilityand moisture content is slightly concave. An improved viabilityequation was applied which takes into account those curvaturesand, at the same time takes into account the initial viabilityof a seed lot which reflects pre-storage deterioration. Thefit was excellent and thus it is now possible to predict percentageviability of any lot of barley seed after any storage periodunder a very wide range of conditions. A culture of rubus yellow net virus (RYNV) was obtained free from other detectable viruses by heat treatment of red raspberry (Rubus idaeus) cv. Mailing Jewel showing veinbanding mosaic symptoms. Graft inoculated black raspberry (JR. occidentalis) plants showed three kinds of ultrastructural abnormality: (1) cell wall outgrowths in many kinds of cells in the leaf blade and vascular bundles, (2) tubular structures c. 30 nm in diameter and up to 1100 nm long, in groups in the cytoplasm close to the nucleus and (3) small bacilliform virus-like particles c. 80–150 × 25 nm in size randomly distributed in the cytoplasm of many kinds of leaf cells, but especially in the phloem. The bacilliform particles, which in some cells were in large groups associated with lightly staining amorphous material, are considered to be those of RYNV.  相似文献

SYNOPSIS The development of 1st generation schizonts of Eimeria callospermophili was studied with cell cultures and with experimentally infected host animals, Spermophilus armatus. Sporozoite-shaped schizonts each had 5-10 nuclei and all of the organelles of the sporozoite; each nucleus had a nucleolus and an associated Golgi apparatus. In stages immediately preceding merozoite formation, an intranuclear spindle apparatus with conical polar areas were observed near the outer margin of each nucleus. Two centrioles, each having 9 single peripheral tubules and one central tubule, were observed near each pole in some specimens. Merozoite formation began internally, with anlagen of 2 merozoites developing near each nucleus. The inner membrane of the merozoites first appeared as 2 dense thickenings adjacent to the polar cones and centrioles; subpellicular microtubules appeared simultaneously. Two anterior annuli and the conoid formed between the 2 thickenings. Vesicles, possibly of Golgi origin, were located next to the forming inner membrane. As the forming merozoites underwent elongation, a rhoptries anlage, a Golgi apparatus, refractile bodies, and mitochondria were incorporated into each. Sporozoite-shaped schizonts with merozoite anlagen transformed into spheroid or ovoid schizonts; at this time the conoid, rhoptries, micronemes, and the inner membrane of the pellicle gradually disappeared; several small refractile bodies were formed from the larger one. When development was about 1/3 complete, the immature merozoites began to grow outward from the surface of the schizont. In this phase of development, the single surface membrane of the schizont became the outer membrane of the merozoite's pellicle, and additional organelles, including the nucleus, were incorporated. Finally, the merozoites became pinched off, leaving a residual body. Development in cell cultures and host tissues was similar. This type of schizogony, previously undescribed in Eimeria, is compared with corresponding stages of development in other species of Eimeria and Sporozoa.  相似文献
