Quantitative trait loci (QTL) analysis of aluminium (Al) tolerance was performed using Ler/Cvi recombinant inbred (RI) lines of Arabidopsis thaliana. Relative root length (RRL) (root length with 4 µm Al/root length with no Al at pH 5.0) on day 5 was used as the Al tolerance index for QTL analysis. Al tolerance judged by RRL was well correlated to tolerance judged by other indexes, including accumulation of callose, reactive oxygen species in the root apex and growth performance on acid soil containing a large amount of exchangeable Al. Using data sets with an hb2 of 0.91, two QTLs were detected at the top of chromosome 1 and bottom of chromosome 3. These QTLs explained 40 and 16% of the phenotypic variation of Al tolerance, respectively, and the positive effect of the Cvi allele. The QTL on chromosome 1 overlapped with a major QTL in another recombinant inbred population, and is possibly related to malate excretion. A complete pair-wise search revealed 11 sets of epistatic interacting loci pairs, which accounted for the transgressive segregation among the RI population. Several epistatic interactions shared the same chromosomal region, indicating the possible involvement of regulatory proteins in Al tolerance in Arabidopsis.  相似文献

From leaves of barley and spinach, cellular components wereisolated and brought together under various conditions to investigatethe fate of ascorbic acid as affected by the components in thelight and dark. 2. A new colorimetric method for assaying ascorbic acid andsome other reducing substances was devised, measuring the colorof molybdenum-blue developed by the substances in the presenceof excess amounts of phosphomolybdate and inorganic acid. 3. The photooxidation of ascorbic acid by green and yellow filtrates,prepared from green and etiolated leaves of barley, was studiedby the ordinary as well as the new colorimetric method. In thepresence of oxygen, the oxidation of ascorbic acid was foundto be accelerated by light in the green filtrate, but not inthe yellow filtrate. 4. The oxidation of the endogenous reducing substance containedin the supernatant fraction of spinach leaf extracts was studiedin the presence of washed chloroplasts (spinach). In the presenceof oxygen, the rate of oxidation in the light was markedly higherthan in the dark. From the changes in absorption spectrum accompanyingthe reaction, the endogenous reducing substance in questionwas identified as ascorbic acid. 5. The occurrence of an endogenous precursor of ascorbic acidin spinach leaf extracts was disclosed. The photoreduction ofthis precursor into ascorbic acid was studied in the precenceof spinach chloroplasts. A specific inhibition of this reactionby phosphoglycerate and glycerophosphate was discovered. 6. Daily activity rhythms are widespread characteristics among many organisms and are usually related to one or more physical environmental variables, such as light and temperature. Even organisms that are not obviously exposed to wide fluctuations in environmental variables, such as troglodytic and endogeic species, exhibit some daily activity patterns. Termites inhabit underground areas or tree trunks where little or no fluctuation in light occurs. In the present study, the activity rhythms of field colonies of the subterranean termite Reticulitermes speratus Kolbe (Blattodea: Termitidae) are examined, during all four seasons, by monitoring their acoustic emissions using a newly‐modified method. Nest temperature affects termite activity significantly during all four seasons. By contrast, no significant effect of time is observed during the different seasons, with the exception of autumn. Activity and nest temperature are negatively correlated in summer and autumn, although thay are are positively correlated during winter and spring. The greatest activity occurs at 23–25 °C. The experimental observations suggest that the activity of R. speratus termite colonies is affected mainly by ambient temperature, in a manner that is based on optimum temperature.  相似文献
