Resident cardiac stem cells, known as “cardiogenic progenitor cells” (CPCs), are a heterogeneous population of immature cells residing in the myocardium and capable of self-renewal and differentiation into cardiomyocyte-like and vascular-like cells. CPCs are usually isolated by long enzymatic digestion of heart tissue and selection with stem cell markers. However, long exposure to enzymatic digestion and the small size of a myocardial sample significantly hinder acquiring a large number of viable cells. To avoid these problems, we developed a method based on CPC growth ex vivo and subsequent immunomagnetic selection.  相似文献

Influence of dsRNA isolated from killer yeast of S. cerevisiae on humoral and cellular immune responses in mice CBA/CaY and C57Bl/6Y with opposing reaction to the antigen was studied. It was shown that after administration of the yeast dsRNA preparation to the animals simultaneously with the antigen there was an increase in the number of the antibody forming cells in the spleen and the titer of hemolytic antibodies in blood serum of the animals with high and low reactions to the antigen. After sensitization with different doses of sheep red blood cells (10(7) and 10(8)) the preparation had immunomodulating action on development of DTH in mice CBA/CaY. The effect of the dsRNA preparation on the immunity system depended on the preparation dose, antigen loading and animal genotype and was the most marked in mice CBA/CaY with interferon levels in blood serum 2-3 times higher than those in mice C57Bl/6Y.  相似文献

We studied stomatal conductance (gs) in leaves of cucumber plants (Cucumis sativus L., cv. Zozulya) subjected at early developmental stages to either short-term daily cooling (2 h at the end of night periods) or continuous chilling (12°C throughout the day and night). Irrespective of the irradiance during measurements, continuous chilling either lowered gs or had no effect, as compared to gs of control plants (23°C). In plants subjected to periodic short-term cooling, the gs was found to increase at low temperatures both at moderate and high irradiance; it also increased at high temperature (33°C) but only at photosynthetically active irradiance of 800 ??mol/(m2 s). It is supposed that heat-loving plants, subjected to different types of low-temperature treatment, mobilize different mechanisms of stomatal regulation and employ different strategies of adaptation in response to low- and high-temperature treatments. The unusual behavior of stomata, manifested in stomatal opening at both low and high temperatures, extends the adaptive potential of plants subjected daily to short-term low-temperature treatments. This leads to a high level of photosynthesis, biomass accumulation, and supports high physiological activity in plants.  相似文献

Effect of short-term (2 h a day) and long-term (6 days) exposure to low temperature (5°C) on cold tolerance was investigated in two cultivars of potato (Solanum tuberosum L.): resistant (Sudarynya) and susceptible (Nevskii) to potato cyst nematode (Globodera rostochiensis Woll.). The extent of their infestation and changes in the expression of the genes of resistance to nematode (H1 and Gro1-4) were also analyzed. In both cultivars, exposure to low temperature enhanced cold resistance of potato plants. Enhancing cold resistance of cv. Sudarynya induced by a short-term exposure to chilling did not affect the extent of nematode infestation, whereas in susceptible cv. Nevskii, the extent of infestation decreased by almost three times. The level of expression of H1 gene in the leaves of the susceptible cultivar rose almost twofold both after short-term and long exposure to chilling, while in the resistant cultivar, gene expression increased only after a short-term effect of cold. The level of Gro1-4 gene expression increased after both temperature treatments only in the resistant cv. Sudarynya. Thus, the expression of genes for potato resistance to nematode infestation became more active in the susceptible cultivar as regards the gene H1 and in the resistant cultivar, regarding the gene Gro1-4. In the nematode-susceptible cv. Nevskii, the level of infestation decreased and cold resistance increased, apparently indicating cross adaptation to two factors of different nature.  相似文献

The development of plants depends on the photoperiod length, light intensity, temperature, and length of light day integral. The reaction of a plant to the day length or daily light integral can depend on both the range of studied light intensities and photoperiod. Based on the data concerning the effects of light and thermal integrals on the developmental rate of plants of different photoperiodic groups, a photothermal model of plant development was proposed. The model was used to calculate the lengths of optimal photoperiods and ranges of daily temperature gradients ensuring the highest developmental rate of some plants, such as soybean, wheat, cucumber, and barley.  相似文献

The thermomorphological responses of young cucumber plants to a temperature drop lasting 2, 4 and 6 hours applied at the beginning, in the middle and at the end of the night were examined. Plant height, leaf petiole length and plant dry weight were measured and, based on the experimental data, the regression dependences of growth variables on the temperature drop were fitted. A method to determine the optimal temperature drop (the duration and time of exposure) which results in shortened plant height and leaf petiole length without a decrease in plant dry weight is proposed.  相似文献

The maintenance of pluripotency of mesenchymal stromal cells (MSCs), their proliferation and initiation of differentiation may critically depend on functional expression of ion channels. Despite such a possibility, mechanisms of electrogenesis in MSCs remain poorly understood. In particular, little is known about a variety of ion channels active in resting MSCs or activated upon MSC stimulation. Here we aimed at uncovering ion channels operating in MSCs, including those being active at rest, using the patch clamp technique and inhibitory analysis. In trying to evaluate a contribution of anion channels in MSC resting potential, we employed a number of diverse inhibitors of anion channels and transporters, including niflumic acid (NFA). Basically, NFA caused hyperpolarization of MSCs that was accompanied by a marked increase in ion conductance of their plasma membranes. The blockage of Cl? channels could not underlie such a NFA effect, given that cells dialyzed with a CsCl solution were weakly or negligibly sensitive to this blocker. This and other findings indicated that NFA affected the MSC ion permeability not by targeting Cl? channels but by stimulating K+ channels. NFA-activated K+ current was TEA and diltiazem blockable, and K+ channels involved were potentiated from outside by solution acidification and Cu2+ ions. Taken together, the data obtained implicated two-pore domain K+ channels of the TREK-2 subtype in mediating stimulatory effects of NFA on MSCs. The notable inference from our work is that TREK-2 channels should be expressed and functional virtually in every MSC, given that all cells examined by us (n > 100) similarly responded to NFA by increasing their TREK-2-like K+ conductance.  相似文献

Based on the data obtained in the experiments with constant and fluctuating diurnal temperatures, the temperature dependence of developmental rate was calculated for different photoperiodic groups of plants using the square equations of regression. These equations made it possible to calculate the temperature areas of maximum developmental rates of plants under different photoperiodic conditions.  相似文献
