Action potential propagation along myelinated nerve fibers requires high-density protein complexes that include voltage-gated Na(+) channels at the nodes of Ranvier. Several complementary mechanisms may be involved in node assembly including: (1) interaction of nodal cell adhesion molecules with the extracellular matrix; (2) restriction of membrane protein mobility by paranodal junctions; and (3) stabilization of ion channel clusters by axonal cytoskeletal scaffolds. In the peripheral nervous system, a secreted glial protein at the nodal extracellular matrix interacts with axonal cell adhesion molecules to initiate node formation. In the central nervous system, both glial soluble factors and paranodal axoglial junctions may function in a complementary manner to contribute to node formation.  相似文献

In myelinated nerve fibers, action potential initiation and propagation requires that voltage-gated ion channels be clustered at high density in the axon initial segments and nodes of Ranvier. The molecular organization of these subdomains depends on specialized cytoskeletal and scaffolding proteins such as spectrins, ankyrins, and 4.1 proteins. These cytoskeletal proteins are considered to be important for 1) formation, localization, and maintenance of specific integral membrane protein complexes, 2) a barrier restricting the diffusion of both cytoplasmic and membrane proteins to distinct regions or compartments of the cell, and 3) stabilization of axonal membrane integrity. Increased insights into the role of the cytoskeleton could provide important clues about the pathophysiology of various neurological disorders.  相似文献

Transthyretin (TTR) amyloid fibril formation, which is triggered by the dissociation of tetrameric TTR, appears to be the causative factor in familial amyloidotic polyneuropathy and senile systemic amyloidosis. Binding of thyroxine (T(4)), a native ligand of TTR, stabilizes the tetramer, but the bioavailability of T(4) for TTR binding is limited due to the preferential binding of T(4) to globulin, the major T(4) carrier in plasma. Here, we show that Cr(3+) increased the T(4)-binding capacity of wild-type (WT) and amyloidogenic V30M-TTR. Moreover, we demonstrate that Cr(3+) and T(4) cooperatively suppressed in vitro fibril formation due to the stabilization of WT-TTR and V30M-TTR.  相似文献

A case of unilateral athelia with a subcutaneous dermoid cyst in the area in question is reported. The histologic features of the cyst wall suggest that it is derived from a base in the nipple-areola complex. Dozens of cases of congenital absence of the breast have been reported, some of them associated with congenital anhidrotic ectodermal dysplasia, others with additional musculoskeletal abnormalities, and still others normal in other respects. But to our knowledge, the combination of athelia and corresponding subcutaneous dermoid cyst has not been reported.  相似文献

The secretion of transthyretin (TTR) variants contributes to the pathogenesis of amyloidosis because they form aggregates in the extracellular environment. However, the mechanism of how TTR variants pass the quality control system in the endoplasmic reticulum (ER) has not yet been elucidated. We investigated here the mechanism of how TTR passes ER monitoring. Monomeric mutation introduced in TTRs (M-TTRs) resulted in the ER retention of amyloidogenic M-TTRs but not non-amyloidogenic M-TTRs. Retention of amyloidogenic M-TTRs induced the unfolded protein response and upregulated the expression of ER chaperones BiP and glucose-regulated protein (GRP) 94. Additionally, we showed that the ER-retained amyloidogenic M-TTRs are subject to ER-associated degradation. On the other hand, the amyloidogenic TTR variants and non-amyloidogenic M-TTRs were secreted normally. These findings suggest that unlike for wild-type TTR, the ER quality control system may differentially regulate the fate of the TTR variants and their monomeric counterparts.  相似文献

Fibronectin contains the active sequence Arg-Gly-Asp (RGD), along with its synergic site Pro-His-Ser-Arg-Asn (PHSRN). However, the PHSRN peptide does not show synergic activity when it is mixed with the RGD peptide, indicating that a spatial array between RGD and PHSRN in fibronectin may be necessary for synergic activity. Here, we have used an amino acid type poly(ethylene glycol) derivative (aaPEG) to design a bivalent PEG hybrid of fibronectin active peptides. We prepared the aaPEG hybrid peptides PHSRN-aaPEG, aaPEG-RGD, and PHSRN-aaPEG-RGD, and tested their biological activity. Whereas aaPEG-RGD promoted cell spreading activity, PHSRN-aaPEG had no activity. The PHSRN-aaPEG-RGD hybrid strongly promoted cell spreading compared with aaPEG-RGD. These results suggest that the PHSRN sequence in the PHSRN-aaPEG-RGD molecule synergistically enhances the cell spreading activity of the RGD sequence, and that the bivalent aaPEG hybrid method may be useful for conjugating functionally active peptides.  相似文献

Thymic stromal lymphopoietin (TSLP) exerts a marked influence on the polarization of dendritic cells to drive T helper (Th) 2 cytokine production, and has been linked to allergic airway diseases. Although TSLP is produced by airway epithelium, TSLP production in laryngeal arytenoid fibroblasts remains largely unexplored. We examined the effect of Toll-like receptor (TLR) ligands and the cross-talk that occurs among different TLR ligands on TSLP production in arytenoid fibroblasts. Since mRNA of TLR 2, 3, 4, and 9 has been found to be expressed in arytenoid fibroblasts, we examined the effect on its production of TLR ligands. TSLP production by arytenoid fibroblasts was strongly induced in the presence of polyinosinic-polycytidylic acid (poly(I:C)), a ligand of TLR3. Its production was synergistically induced in the presence of IL-4, to a level more than 100 times higher than that observed in the absence of poly(I:C) or IL-4. We also revealed that B type DNA containing CpG motifs (CpG-DNA) coding for a TLR9 ligand markedly suppressed both poly(I:C)-induced and poly(I:C)-plus-IL-4-induced TSLP production. B type CpG-DNA decreased the poly(I:C)-induced phosphorylation of c-Jun N-terminal kinase (JNK), and pre-incubation with SP600125 (inhibitor of JNK) reduced the poly(I:C)-induced TSLP-production. These results indicate that human arytenoid fibroblasts strongly induce TSLP production with stimulation by double-stranded RNA (dsRNA), which can be inhibited by CpG-DNA and participate in immune allergic responses.  相似文献

Productivity, nutrient input, nutrient uptake, and release rates were determined for a coral-dominated reef flat at La Réunion, France, to assess the influence of groundwater nitrogen on carbon and nutrient budgets. Water samples were collected offshore in the ocean, at the reef crest and back reef for nutrients, picoplankton, pH, and total alkalinity. Volume transport of ocean water across the reef flat was measured using both current meters and drogues. Groundwater advected onto the reef flat and mixed with incoming ocean water. Metabolic rates for the reef community were determined to be: gross primary production = 1,000 mmol C m−2 d−1, community respiration = 960 mmol C m−2 d−1, and community calcification = 210 mmol C m−2 d−1. Across the reef flat, silicate behaved conservatively, there was net uptake of phosphate (0.06 mmol P m−2 d−1) and net release of nitrate, ammonia, dissolved and particulate organic nitrogen (total 7.0 mmol N m−2 d−1). Groundwater nitrate contributed 37% of the increase in nitrate plus ammonia. The first-order mass transfer coefficient of phosphate was 3.3 m d−1, and for nitrate plus ammonia, 5.9 m d−1. Gross N and P uptake from estimates of mass transfer and uptake of particles were 0.37 mmol P m−2 d−1 and 7.2 mmol N m−2 d−1, respectively giving an N:P uptake ratio of 20:1. Thus, the elevation of nitrogen across the reef flat maintains a high N:P flux, enhancing algal growth downstream of the transect. We conclude that net community production (40 mmol C m−2 d−1) was sustained by net uptake of phosphate from the ocean and net uptake of new nitrogen from groundwater.
