This paper describes the morphology and response characteristics of two types of paired descending neurons (DNs) (classified as DNVII1 and DNIV1) and two lobula neurons (HR1 and HP1) in the honeybee, Apis mellifera.
1.  The terminal arborizations of the lobula neurons are in juxtaposition with the dendritic branches of the DNs (Figs. 2, 3b, 5). Both of the DNs descend into the ipsilateral side of the thoracic ganglia via the dorsal intermediate tract (Fig. 6) and send out many blebbed terminal branches into the surrounding motor neuropil (Figs. 3c, 7).
2.  Both the lobula and descending neurons respond in a directionally selective manner to the motion of widefield, periodic square-wave gratings.
3.  The neurons have broad directional tuning curves (Figs. 10, 11). HR1 is maximally sensitive to regressive (back-to-front) motion and HP1 is maximally sensitive to progressive (front-to-back) motion over the ipsilateral eye (Fig. 11). DNVII1 is maximally sensitive when there is simultaneous regressive motion over the ipsilateral eye and progressive motion over the contralateral eye (Fig. 12a). Conversely, DNIV1 is optimally stimulated when there is simultaneous progressive motion over the ipsilateral eye and regressive motion over the contralateral eye (Fig. 12b).
4.  The response of DNIV1 is shown to depend on the contrast frequency (CF) rather than the angular velocity of the periodic gratings used as stimuli. The peak responses of both regressive and progressive sensitive DNs are shown to occur at CFs of 8–10 Hz (Figs. 13, 14).

Pinocytic activity was quantified for rat yolk sacs incubated in a medium that was either serum-free or contained 10% (v/v) of calf serum. Absence of serum from the medium caused a small increase in the rate of pinosome formation, as determined by the rates of capture of both 125I-labelled poly(vinylpyrrolidone) and [14C]sucrose. In contrast, the rates of uptake of substrates ingested by adsorptive pinocytosis were greatly enhanced when serum proteins, which compete for the same binding sites on the plasma membrane as used by adsorbing substrates, were absent. Elimination of such competition greatly simplifies the quantitative analysis of the binding process, and permitted a detailed study of the binding to the plasma membrane of formaldehyde-denatured bovine serum albumin, a protein that is rapidly digested within the lysosomal system after its pinocytic capture. Binding obeyed Michaelis-Menten kinetics and showed a dissociation constant of approx. 1 micron, indicating the high affinity of this protein for binding sites on the surface of actively pinocytosing yolk-sac cells.  相似文献

Chronic lymphocytic leukemia (CLL) exhibits a highly variable clinical course. Altered DNA methylation of genes has shown promise as a source of novel prognostic makers in a number of cancers. Here we have studied the potential utility of a panel of methylation markers (CD38, HOXA4 and BTG4) in 118 CLL patients. Each of the three loci assessed exhibited frequent methylation, as determined by COBRA analysis and individually correlated with either good (CD38, BTG4 methylation) or poor (HOXA4 methylation) prognosis. Using a combined approach to produce an overall methylation score, we found that methylation score was significantly associated with time to first treatment in CLL patients. Multivariate Cox regression analysis revealed that methylation score was the strongest predictor of time to first treatment and was independent of IGHV gene mutational status and CD38 expression. This study provides proof of principle that a panel of methylation markers can be used for additional risk stratification of CLL patients.Key words: DNA methylation, prognostic marker, leukemia, CD38, HOXA4, BTG4Chronic lymphocytic leukemia (CLL) is the most commonly diagnosed form of leukemia in the UK and exhibits a highly variable clinical course.1 While some patients rapidly succumb to the disease, others can survive for well over ten years, often without the need for treatment. This extreme variability in clinical course among patients presenting with early CLL emphasizes the need for good prognostic markers to help direct patient treatment.1,2 A number of molecular markers are already in frequent use, most notably IGHV mutational status.1 In addition, other markers, such as protein expression of ZAP70 and CD38 and genomic abnormalities are also commonly used.2 However, the group of patients defined as having a good prognosis through the use of current markers is still large (about 60% of CLL patients3) and these patients still exhibit a wide variety in outcome. Thus, additional markers are needed to aid clinical management in these cases.Altered DNA methylation, especially at promoter associated CpG islands, is one of the hallmarks of cancer.4 The high frequency of DNA methylation changes, associated with the relative ease and sensitivity with which DNA methylation can be detected, have led to increasing interest in the use of methylation based markers for diagnosis and prognosis.5 A number of methylated genes have already been shown to correlate with clinical features of CLL. For example, TWIST2 has been reported to exhibit frequent hypermethylation in CLL with mutated IGHV genes, which have a good prognosis. It is, however, rarely methylated in unmutated CLL cases, which have a poorer prognosis.6 Methylation of ZAP70 was strongly correlated with lack of ZAP70 expression and, similarly to TWIST2 methylation, was also strongly correlated with mutation of IGHV.7 In contrast, we recently showed that the HOXA4 gene was a frequent target for hypermethylation in CLL but, in this case, methylation of the gene was associated with the poor prognosis IGHV unmutated cases.8 While in many cases the functional role of the altered methylation still remains to be clearly demonstrated, the association between differential methylation and specific subsets of CLL patients raises the possibility that altered gene methylation could be used to identify patient populations with differing clinical outcome.In this report we have more closely examined the relationship between methylation of specific candidate marker genes and patient outcome, using a panel of genes we have identified as frequently methylated in CLL. In addition to the previously described methylation of the HOXA48 gene, we have also found frequent methylation, as determined by COBRA analysis, of two further genes (CD38 and BTG4). All three genes independently exhibit a correlation with patient outcome. However, a combined approach using all three markers produced the strongest correlation with patient outcome. This combined marker was independently prognostic in multivariate analysis and identified a subset of patients with IGHV mutated genes (i.e., those regarded as good prognosis) who had a significantly greater likelihood of disease progression.  相似文献

Colonisation of freshwater habitats by the European eel Anguilla anguilla   总被引:1,自引:0,他引:1  
1. The spatial distribution of European eels in 18 U.K. rivers was related to distance from tidal limit using a negative exponential model. This function accounted for between 19 and 90% of the variation in eel density where quantitative data was available. For semiquantitative data the negative exponential function was a significant predictor of eel densities in only six out of 10 cases, although all rivers showed a consistent decline in abundance with distance upstream from the tidal limit. 2. The spatial distribution of different age groups of European eel in River Severn showed an initial rapid dispersion into freshwater followed by a much slower dispersion rate. Movement of the population upstream by a wave‐form migration process does not occur in this system. Instead colonisation of freshwaters can be seen as a two‐phase dispersion. Phase‐1 is a rapid dispersion upstream driven by density at the point source. Phase‐2 commences once the eels become yellow eels and is equivalent to random diffusion of particles. 3. These processes have important implications for the penetration of freshwaters with reduced numbers of eel larvae arriving on the coast of Europe and North America. Eel abundance will decrease more in freshwaters in an upstream direction whilst it may remain stable or decrease to a lesser extent in estuaries. They are also able to explain the demography of eels migrating upstream over weirs and the observations of varying sex ratios within catchments. We conclude that a dispersion model dependent on age, temperature, difficulty of migration, habitat quality and density of eels should be an important part of freshwater eel management.  相似文献

Recent behavioural studies have demonstrated that honeybees use visual feedback to stabilize their gaze. However, little is known about the neural circuits that perform the visual motor computations that underlie this ability. We investigated the motor neurons that innervate two neck muscles (m44 and m51), which produce stabilizing yaw movements of the head. Intracellular recordings were made from five (out of eight) identified neuron types in the first cervical nerve (IK1) of honeybees. Two motor neurons that innervate muscle 51 were found to be direction-selective, with a preference for horizontal image motion from the contralateral to the ipsilateral side of the head. Three neurons that innervate muscle 44 were tuned to detect motion in the opposite direction (from ipsilateral to contralateral). These cells were binocularly sensitive and responded optimally to frontal stimulation. By combining the directional tuning of the motor neurons in an opponent manner, the neck motor system would be able to mediate reflexive optomotor head turns in the direction of image motion, thus stabilising the retinal image. When the dorsal ocelli were covered, the spontaneous activity of neck motor neurons increased and visual responses were modified, suggesting an ocellar input in addition to that from the compound eyes.  相似文献
