A series of 26 new quinoline derivatives carrying active pharmacophores has been synthesized and evaluated for their in vitro antituberculosis activity against Mycobacterium tuberculosis H37Rv (MTB), Mycobacterium smegmatis (MC2), and Mycobacterium fortuitum following the broth micro dilution assay method. Compounds 13e, 13i, 13k, 14a, 14c, 14i, and 14k exhibited significant minimum inhibition concentrations, when compared with first line drugs isoniazid (INH) and rifampicin (RIF) and could be ideally suited for further modifications to obtain more efficacious compounds in the fight against multi-drug resistant tuberculosis.  相似文献

NK T cells are a unique lymphocyte population that have developmental requirements distinct from conventional T cells. Mice lacking the tyrosine kinase Fyn have 5- to 10-fold fewer mature NK T cells. This study shows that Fyn-deficient mice have decreased numbers of NK1.1(-) NK T cell progenitors as well. 5-Bromo-2'-deoxyuridine-labeling studies indicate that the NK T cells remaining in fyn(-/-) mice exhibit a similar turnover rate as wild-type cells. The fyn(-/-) NK T cells respond to alpha-galactosylceramide, a ligand recognized by NK T cells, and produce cytokines, but have depressed proliferative capacity. Transgenic expression of the NK T cell-specific TCR alpha-chain Valpha14Jalpha18 leads to a complete restoration of NK T cell numbers in fyn(-/-) mice. Together, these results suggest that Fyn may have a role before alpha-chain rearrangement rather than for positive selection or the peripheral upkeep of cell number. NK T cells can activate other lymphoid lineages via cytokine secretion. These secondary responses are impaired in Fyn-deficient mice, but occur normally in fyn mutants expressing the Valpha14Jalpha18 transgene. Because this transgene restores NK T cell numbers, the lack of secondary lymphocyte activation in the fyn-mutant mice is due to the decreased numbers of NK T cells present in the mutant, rather than an intrinsic defect in the ability of the other fyn(-/-) lymphoid populations to respond.  相似文献

The repellency of the essential oil of the shrubCleome monophylla (Family: Capparidaceae) and identified constituents of the oil were evaluated against the livestock tick,Rhipicephalus appendiculatus and the maize weevil,Sitophillus zeamais. In a tick climbing repellency bioassay, the oil ofC. monophylla exhibited repellency which, at the highest dose, was comparable to that of the commercial arthropod repellent N,N-diethyl toluamide (DEET). In a Y-tube olfactometer bioassay,C. monophylla oil showed higher or comparable repellency againstS. zeamais relative to DEET at all the doses tested. 14 Compounds were identified in theC. monophylla oil by GC, GC-MS and coinjection with authentic samples. Terpenolene was found to occur in largest quantity (14%) followed by 1-α-terpeneol (10%), pentacosane (9%), (α+β)-humulene (8%), phytol (5%) and 2-dodecanone (4%). The most repellent components againstR. appendiculatus andS. zeamais were 1-α-terpeneol and 2-dodecanone. The overall pattern of repellency activity of theC. monophylla constituents with respect to the two arthropods was, however, different. The potential ofC. monophylla in tick and maize weevil control is discussed.  相似文献

Solanum aculeastrum Dunal was investigated for the presence of molluscicidal compounds. This led to the isolation of solaculine A, from the root bark in addition to known steroidal alkaloids; solamargine and beta-solamarine from the berries. The structures were elucidated by spectroscopic techniques. Molluscicidal activity of the aqueous extracts of the berries and root bark, and the isolated compounds were investigated.  相似文献

Wheat (Triticum aestivum L.) cvs. HD 2285 (relatively tolerant) and WH 542 (susceptible) were exposed to ambient and elevated temperature (3–4 °C higher) in open top chambers during post anthesis period. The grain yield components were determined at the time of maturity. In order to elucidate the basis of differential tolerance of these cultivars, the excised developing grains (20 d after anthesis) of ambient grown plants were incubated at 15, 25, 35 and 45 °C for 2 h and then analysed for the activities of soluble starch synthase (SSS), granule bound starch synthase (GBSS), kinetic parameters of SSS and content of heat shock protein (HSP 100). The elevated temperature during grain development significantly decreased grain growth in WH 542 whereas no such decrease was observed in HD 2285. High temperature tolerance of HD 2285 was found to be associated with higher catalytic efficiency (Vmax/Km) of SSS at elevated temperature and higher content of HSP 100.  相似文献

Sixteen more Amaryllidaceae alkaloids have been isolated from bulbs of Crinum kirkii Baker of which noraugustamine and 4a,N-dedihydronoraugustamine are hitherto unknown. Their structures and those of earlier known alkaloids have been established by physical and spectroscopic analysis. Application of 2D NMR techniques was used for complete characterization of the alkaloids as well as of 3-O-acetylsanguinine. 1,2-Diacetyllycorine and 3-O-acetylsanguinine showed activity against Trypanosoma brucci rhodesiense, the parasite associated with sleeping sickness. 3-O-acetylsanguinine also showed some activity against Trypanosoma cruzi.  相似文献

IntmiR is a manually curated database of published intronic miRNAs of Human and Mouse genome. Each entry in the database, aims at providing a complete resource of intronic miRNA with their target gene and deregulation in various diseases with related tissues and pathways. The current release contains 426 intronic miRNA loci from human and 76 from mouse, expressing distinct target mRNA sequences. Database gives information on an intronic miRNA-disease relationship, including miRNA ID, pathaway connected and related tissues. All entries can be retrieved by miRNA ID or target gene. Using selected bio‐based feedstocks as alternative to fossil resources for producing biochemicals and derived materials is increasingly considered an important goal of a viable bioeconomy worldwide. However, to ensure that using bio‐based feedstocks is aligned with the global sustainability agenda, impacts along the entire life cycle of biochemical production systems need to be evaluated. This will help to identify those processes and technologies, which should be targeted for optimizing overall environmental sustainability performance. To address this need, we quantify environmental impacts of biochemical production using distinct bio‐based feedstocks, and discuss the potential for reducing impact hotspots via process optimization. Lactic acid (LA) was used as an example biochemical derived from corn, corn stover, and macroalgae (Laminaria sp.) as feedstocks of different technological maturity. We used environmental life cycle assessment (LCA), a standardized methodology, considering the full life cycle of the analyzed biochemical production systems and a broad range of environmental impact indicators. Across production systems, feedstock production and biorefinery processes dominate life cycle impact profiles, with choice in energy mix and biomass processing as main influencing aspects. Results show that uncertainty decreases with increasing technological maturity. When using Laminaria sp. (least mature among selected feedstocks), impacts are mainly driven by energy utilities (up to 86%) due to biomass drying. This suggests to focus on optimizing or avoiding this process for significantly increasing environmental sustainability of Laminaria sp.‐based LA production. Our results demonstrate that applying LCA is useful for identifying environmental impact hotspots at an earlier stage of technological development across biochemical production systems. With that, our approach contributes to improving the environmental sustainability of future biochemical production as part of moving toward a viable bioeconomy worldwide.  相似文献
