Summary The response of tomato (Lycopersicon esculentum Mill) to inoculation with the vasicular arbuscular mycorrhizal (VAM) fungusGlomus fasiculatum andAzotobacter vinelandii singly and in combination was tested in the field. It was found thatG. fasiculatum as well asA. vinelandii significantly increased leaf area, shoot dry weight, nitrogen content phosphorus content and yield in respect to uninoculated
control. While, VAM fungal treatment alone could bring about substantial increase in growth, nitrogen content, phosphorus
content and yield, its combination withA. vinelandii produced additional effects on leaf area, shoot dry weight, phosphorus content and yield.

Summary Bean (Phaseolus vulgaris L. Cv. Burpees Stringless) seeds were subjected to two cycles of presowing soaking and drying treatments with sodium molybdate and cobalt nitrite at 1 and 5 ppm concentrations used separately and also in combination. Sodium molybdate 2 ppm and cobalt nitrite 1 ppm used singly proved better than the remaining treatments with respect to nodulation, dry matter, nitrogen and yield. Combined treatment with sodium molybdate and cobalt nitrite did not produce additive effect on any parameter studied compared to their usage alone. 相似文献

We present a fast protocol that can be used to obtain highly purified cultures of proliferating adult human and rat Schwann cells accessible for non-viral transfection methods. The use of enriched genetically modified adult Schwann cells is of interest in the context of autologous cell transplantation within nerve transplants for peripheral nerve repair. Cell preparation from pre-degenerated adult peripheral nerves is described, together with the use of melanocyte growth medium plus forskolin, fibroblast growth factor-2 (FGF-2), pituitary extract and heregulin as a selective, serum-free culture medium and a subsequent cell enrichment step (cold jet). Proliferating adult Schwann cells can be efficiently genetically modified using optimized, non-viral electroporation protocols. The protocol results in Schwann cell cultures that are more than 90-95% pure, and transfection efficiencies vary depending on the initial cell constitution from 20 to 40%. The procedure takes up to 21 d, depending on the length of the pre-degeneration period. 相似文献

β-arrestin2 is a key molecule involved in signaling and internalization of activated G protein-coupled receptors including µ-opioid receptors (MOR). Previously we have shown that decreased expression of β-arrestin2 upon chronic morphine is associated with the development of opioid tolerance in the gastrointestinal tract. However, the localization of β-arrestin2 within the gastrointestinal wall is not known. In this study we found that β-arrestin2 is localized in the soma of a select group of neurons in the myenteric ganglia but not in smooth muscle. The density of β-arestin2 was significantly higher in the ileum than the colon. We identified four variants of β-arrestin2 in the ileum, with ARRB-005 and ARRB-013 being the most abundant. Further, the current study utilized multiple-labeling immunofluorescence to characterize the chemical coding of neurons expressing β-arrestin2 in the murine myenteric plexus and the co-localization of MOR1 and β-arrestin2. β-arrestin2 co-localized with choline acetyltransferase and calretinin. In contrast, β-arrestin2 neither co-localized with substance P, nitric oxide synthase nor calbindin. Genetic deletion of β-arrestin2 did not affect cholinergic neuron activation by nicotine in the isolated ileum (-log M EC50: wild type = 5.8 vs. β-arrestin2 knockout = 5.9). Our findings suggest specificity in the localization of β-arrestin2 in the myenteric plexus within MOR1-expressing neurons and provide a relation for direct intracellular crosstalk between MOR1 receptor activation and β-arrestin2 signaling in the myenteric neurons. β-arrestin2 deletion does not directly alter basal enteric cholinergic neuronal function. 相似文献

Summary Growth inhibitors are washed down from the aerial vegetative parts ofParthenium hysterophorus L. during rain. These get adsorbed to soil particles thereby rendering the substratum inhibitory. The trichomes covering the vegetative parts contain inhibitors. The trichomes which get easily detached from dry parts when happen to settle on leaf surface of other plants in high quantity, reduce the chlorophyll and dry matter content in those species.Part I appeared in Plant and Soil53, 27 (1979). 相似文献

Effect of precolonization of banana cv Neeypovan roots with Pseudomonas fluorescens on infection with Fusarium oxysporum f.sp. cubense was studied. Under in vitro conditions Pseudomonas fluorescens clearly inhibited Fusarium oxysporum f.sp. cubense. Fluorescein isothiocyanate-tagged antibodies raised in a rabbit system for Pseudomonas fluorescens and Fusarium oxysporum f.sp. cubense separately were used to study the spread of both organisms in banana root. It was observed that precolonization with Pseudomonas fluorescens could reduce Fusarium oxysporum f.sp. cubense colonization by 72%, and also correlated with a number of structural changes in the cortical cells, mainly with densely stained
amorphous material and polymorphic wall thickenings as revealed by light and electron microscopic studies. Massive depositions
of unusual structures at sites of fungal entry was also noticed, which clearly indicated that bacterized root cells were signalled
to mobilize a number of defence structures for preventing the spread of pathogen in the tissue.

Nitrogen fixation (acetylene reduction) was found in intact tomato (Lycopersicon esculentum Mill ‘Pusa Ruby’) plants in the field, in pots and also in aseptic cultures. The unsterilized as well as sterilized rhizoplane
and phylloplane of the plant when assayed separately also responded to the test. From root bits of tomato sterilized upto
20 minutes with 0.1% mercuric chloride, growth of the bacterium from the interior of the root into the medium was observed
thereby indicating their presence within the endorhizosphere. Phase contrast and electron microscopic studies of the root
system of tomato revealed the presence of bacterial colonies in the epidermis, cortex and vascular bundles. Bacterial numbers
in the endosphere, of root and leaf were 30×104 and 12×104, respectively, per gram fresh weight of tissue. The bacteria were predominantly rod-shaped 1.4–4.8×0.9–1.95 μm in 24-h-old
cultures, pleomorphic, polar or bipolary flagellated having β-hydroxy butyrate granules. The bacterium has been identified

Summary Association withParthenium hysterophorus L. caused retarded growth and nodulation in bean (Phaseolus vulgaris var. Burpees Stringless) the inhibition decreasing with increasing distance from the weed. Leachate collected from Parthenium grown pots also caused similar inhibition in bean growth. The inhibitory nature of the root exudate was confirmed under sterile cultural conditions by its effect on wheat (Triticum aestivum var. UP301) seedling growth. At the rosette and flowering stage of the weed there was maximum exudation of inhibitors which remained active for about thirty days. 相似文献

An efficient protocol was developed using cell suspensions for somatic embryogenesis and plantlet regeneration in a most popular diploid AB banana (M.accuminata X M.bulbisiana hybrid) cv. Elakki Bale (syn Neypoovan) known for its taste and keeping quality in southern India. Floral primodia from position 8–16 of male inflorescence which were more responsive for embryogenesis were used as explants for the embryogenic callus production in MS media supplemented with different concentration of 2,4-D. A concentration of 18.1 μM 2, 4-D produced maximum embryogenic calli in 1 % of the explants inoculated. Embryogenic calli on repeated sub culturing on MA2 media produced good embryogenic cell suspensions (ECS). Microscopic examination of ECS showed globular, smaller with dense cytoplasm filled with starchy granules characteristic of embryogenic cells. Highest number of somatic embryos (189) was produced on modified MA3 media. A germination percentage of 31 % were observed in BAP 22.19 μM concentration. Regenerated plants with normal shoot and root were hardened in soilrite. Direct somatic embryogenesis and plant regeneration was also noticed in embryogenic calli which did not pass through the ECS stage. The protocol optimized for somatic embryogenesis through cell suspension and also direct embryogenesis leading to plantlet regeneration can be used for the micropropagation and genetic manipulation. 相似文献

Panama wilt of banana caused by Fusarium oxysporum f. sp. cubense (race 1) is a serious disease devastating the important cultivar Neypoovan (syn Elakki Bale AB) in southern India. Chemical control methods are not very effective in controlling the disease. The objective of this study was to evaluate biocontrol agents (BCAs) under controlled and field conditions for their efficacy against the pathogen and to detect and quantify the reduction in FOC population. Arbuscular mycorrhizal (AM) fungi, Trichoderma harzianum and Pseudomonas fluorescens were inoculated at the time of planting in single, dual and tripartite combinations allowing colonization up to 0, 45 and 90 days. Plants were challenged thereafter with 50 g of FOC inoculum multiplied on sorghum grains containing 1.5×106 cfu g?1. Uninoculated plants and those inoculated with pathogen only were controls. Plant growth parameters were measured and structural modifications in the roots were studied. FOC populations in the roots were determined by ELISA every month and final yield was recorded. At the end of 7 months, plants pre-inoculated with BCAs i.e., G. mosseae+T. harzianum and challenged with Fusarium under field conditions could sustain 61 and 70% improvement in plant height and girth, respectively, and 75% in bunch weight over plants not precolonised with BCAs but challenged with FOC which finally succumbed to the disease. ELISA study revealed Fusarium population was reduced to 0.58 OD in 7 months in G. mosseae and T. harzianum treatment compared to a level of 1.9 OD in Fusarium alone treated plants. Beneficial effect of BCAs may be due to the over all protection provided by them by causing physical modifications in the cell wall, growth promotion and through induction of disease resistance. 相似文献
