The constituents of the bark, timber and seeds of Mesua thwaitesii were examined by column chromatography and GC-MS. 1,5-Dihydroxyxanthone, 1,7-dihydroxyxanthone (euxanthone), 1,3-dimethoxy-5-hydroxyxanthone, 1,5,6-trihydroxyxanthone, mammeisin, (4-phenylcoumarin) and sitosterol have been characterized. Nine 4-phenylcoumarins including mammeigin, mesuagin, mammeisin, mesuol, or its isomers, have been identified in the seed extract. One coumarin has not been previously reported.  相似文献

A series of 11-oxa prostaglandin analogs was evaluated for FP receptor binding and activation. Several compounds having aryloxy-terminated lower chains were found to be potent agonists. Topical ocular dosing of AL-12182, the isopropyl ester prodrug of the potent agonist 13, lowered intraocular pressure in the monkey by 40% accompanied by minimal conjunctival hyperemia in the rabbit. AL-12182 was synthesized on multigram scale starting with D-sorbitol.  相似文献

Colorectal cancer (CRC) is the third most common cause of cancer death in the Western world. Calcineurin (CaN), a Ca2+/calmodulin (CaM)-dependent protein phosphatase, is important for Ca2+-mediated signal transduction. The main objective of this study is to examine the potential role of Ca2+/CaM-dependent protein phosphatase in both normal and in invasive tumor components of human samples. In this study, we carried out 45 cases of CaN activity, 13 cases of CaN protein expression by Western blot analysis, and 6 cases for immunohistochemical analysis in both normal and invasive tumor components of human samples. Immunohistochemical analysis revealed that strong cytoplasmic staining of varying intensity was observed in colon tumors of all patients compared to normal mucosa. In addition, Western blot analysis revealed a prominent overexpressed immunoreactive band with an apparent molecular mass of 60 kDa catalytic alpha subunit (CaN A) as well as CaN Aalpha and beta in colon tumor samples. Elevated CaN protein expression appears to be a possible link between Ca2+ signaling and oncogenic processes.  相似文献

Prior work has implicated viral protein R (Vpr) in the arrest of human immunodeficiency virus type 1 (HIV-1)-infected cells in the G2 phase of the cell cycle, associated with increased viral replication and host cell apoptosis. We and others have recently shown that virion infectivity factor (Vif ) also plays a role in the G2 arrest of HIV-1-infected cells. Here, we demonstrate that, paradoxically, at early time points postinfection, Vif expression blocks Vpr-mediated G2 arrest, while deletion of Vif from the HIV-1 genome leads to a marked increase in G2 arrest of infected CD4 T-cells. Consistent with this increased G2 arrest, T-cells infected with Vif-deleted HIV-1 express higher levels of Vpr protein than cells infected with wild-type virus. Further, expression of exogenous Vif inhibits the expression of Vpr, associated with a decrease in G2 arrest of both infected and transfected cells. Treatment with the proteasome inhibitor MG132 increases Vpr protein expression and G2 arrest in wild-type, but not Vif-deleted, NL4-3-infected cells, and in cells cotransfected with Vif and Vpr. In addition, Vpr coimmunoprecipitates with Vif in cotransfected cells in the presence of MG132. This suggests that inhibition of Vpr by Vif is mediated at least in part by proteasomal degradation, similar to Vif-induced degradation of APOBEC3G. Together, these data show that Vif mediates the degradation of Vpr and modulates Vpr-induced G2 arrest in HIV-1-infected T-cells.  相似文献

Following the oral administration of disulfiram to alcoholic patient volunteers, methyl diethyldithiocarbamate (MeDDC) was noted in blood withdrawn between one and two hours after dosing. This is consistent with previous reports of MeDDC in mice and dogs. It is suggested that MeDDC is an intermediate in the formation of urinary sulfate.  相似文献

The sterile insect technique has been used for more than 50 years to control a range of insects around the world. Sterile insect technique is rapidly becoming a major component of many area‐wide fruit fly management programmes. Irradiation of immature life stages induces sterility in adults, which are then distributed over large areas to mate with wild flies, resulting in no viable offspring. However, irradiation in normal air results in declining adult quality. To optimize the quality of sterile adult flies, several techniques are available to lower the levels of oxygen in fruit fly tissues prior to irradiation. The simplest method is to seal pupae in plastic bags and allow the oxygen consumption of pupae to minimize oxygen in both the air and pupal tissue. Some fruit fly species have rapid decreases in eclosion as a result of low oxygen atmospheres. We tested the tolerance of Queensland fruit fly, Bactrocera tryoni (Froggatt) (Diptera: Tephritidae), to low oxygen for the first time. In the first two experiments, unirradiated B. tryoni pupae were tested for different periods in sealed plastic bags at 17, 21, and 26 °C. Optimum eclosion occurred at 21 °C with the lowest eclosion at 26 °C. In general, mean full eclosion declined at ca. 0.1% eclosion per hour sealed in plastic bags during the first 96 h for all temperatures. In the third and fourth experiments at 17 °C, there was a decline in average eclosion for irradiated and unirradiated pupae of about 13.4% after they were sealed in plastic bags for 192 h. In general, B. tryoni eclosion declined at 0.1% per hour inside sealed plastic bags for periods up to 192 h at 17 °C. Queensland fruit flies can tolerate long periods of conditions found inside sealed plastic bags and current practices for sterile B. tryoni release programmes will result in minimum decrease in eclosion. The possible evolution of tolerance of these conditions is discussed.  相似文献

A series of prostaglandin DP agonists containing a 3-oxa-15-cyclohexyl motif was synthesized and evaluated in several in vitro and in vivo biological assays. The reference compound ZK 118.182 (9beta-chloro-15-cyclohexyl-3-oxa-omega-pentanor PGF(2alpha)) is a potent full agonist at the prostaglandin DP receptor. Saturation of the 13,14 olefin affords AL-6556, which is less potent but is still a full agonist. Replacement of the 9-chlorine with a hydrogen atom or inversion of the carbon 15 stereochemistry also reduces affinity. In in vivo studies ZK 118.182 lowers intraocular pressure (IOP) upon topical application in the ocular hypertensive monkey. Ester, 1-alcohol, and selected amide prodrugs of the carboxylic acid enhance in vivo potency, presumably by increasing bioavailability. The clinical candidate AL-6598, the isopropyl ester prodrug of AL-6556, produces a maximum 53% drop in monkey IOP with a 1 microg dose (0.003% w/w) using a twice-daily dosing regime. Synthetically, AL-6598 was accessed from known intermediate 1 using a novel key sequence to install the cis allyl ether in the alpha chain, involving a selective Swern oxidative desilylation of a primary silyl ether in the presence of a secondary silyl ether. In this manner, 136 g of AL-6598 was synthesized under GMP conditions for evaluation in phase I clinical trials.  相似文献

Tubers of terrestrial orchids are harvested and traded from the eastern Mediterranean to the Caspian Sea for the traditional product Salep. Overexploitation of wild populations and increased middle‐class prosperity have escalated prices for Salep, causing overharvesting, depletion of native populations and providing an incentive to expand harvesting to untapped areas in Iran. Limited morphological distinctiveness among traded Salep tubers renders species identification impossible, making it difficult to establish which species are targeted and affected the most. In this study, a reference database of 490 nrITS, trnL‐F spacer and matK sequences of 133 taxa was used to identify 150 individual tubers from 31 batches purchased in 12 cities in Iran to assess species diversity in commerce. The sequence reference database consisted of 211 nrITS, 158 trnL‐F and 121 matK sequences, including 238 new sequences from collections made for this study. The markers enabled unambiguous species identification with tree‐based methods for nrITS in 67% of the tested tubers, 58% for trnL‐F and 59% for matK. Species in the genera Orchis (34%), Anacamptis (27%) and Dactylorhiza (19%) were the most common in Salep. Our study shows that all tuberous orchid species in this area are threatened by this trade, and further stresses the urgency of controlling illegal harvesting and cross‐border trade of Salep tubers.  相似文献
