Characteristic of carbohydrate components of chickens and human's fibronectins

The paper presents new information about the carbohydrate structures of 39-days chicken's fibronectin. It is found out that chicken fibronectin contains mainly biantennary N-glycans with a core fucose and fucosylated O-glycans. It is shown that N-glycans of chicken fibronectin are poorly sialated, since this protein exhibits affinity for the PNA and weak binding to sialospecific SNA. A comparative analysis of lectin-binding activity of chicken and human fibronectins has shown that both glycoproteins differ in glycan composition.     

Factors interacting with promoter region of immunoglobulin genes. Determination of the binding site boundaries

The nuclear extracts of plasmacytomas producing antibodies were found to contain factors which formed complexes with the promoter fragment of the gene for immunoglobulin kappa-chains. The corresponding complexes found in the extracts of nonlymphoid cells had a different mobility. Two approaches were proposed for determining the boundaries of the region necessary for protein factors to be bound to DNA using nuclease Ba131. A 5'-ATTTGCAT-3' octanucleotide sequence was shown to be necessary for interaction with the protein nuclear factor in the studied plasmacytoma lines. The protein completely lost its affinity if at least one nucleotide was removed or substituted at the 5'- or 3'-end of this sequence. The procedures proposed for determining the precise boundaries of the sequence necessary for protein binding to DNA do not require a preliminary protein purification. The principles on which the procedures are based, set no limitations to their application to other systems used for studying the interaction of proteins with DNA.

Molecular forms of cathepsin D bound with subcellular structures were studied in the grey matter of the large hemispheres. Free and bound forms of the enzymes exposed to solubilization with detergent triton X-100 were fractionated by passage through a Sephadex G-100 column. Gel chromatographic analysis demonstrated three peaks of acid proteinase activity. Different areas of solubilization curves of acid proteinases corresponded to different molecular forms of cathepsins. The initial S-shape areas of solubilization curve corresponded to the first high molecular weight peak of the enzyme activity in the grey matter, whereas the subsequent linear ones -- to the second peak; the activity of free forms of the enzyme corresponded to the third peak.
