Chromatographic separation of the liquid culture filtrate of the basidiomycete fungus Physisporinus sanguinolentus has yielded three new compounds viz., 2-methyl-4-pyrone, 2-methyl-5,6-dihydro-4-pyrone and the pyridone form of 4-hydroxy-2-methylpyridine, together with the known triacetic acid lactone, the sesquiterpene dialdehyde merulidial and a derivative of merulidial. Their structures were elucidated by spectroscopic analysis and by comparison to literature data and a synthetic sample. One of the compounds, merulidial, was shown to inhibit the germination of spores and the hyphal growth of the wood-rotting basidiomycete Heterobasidion annosum and the saprophytic mould Cladosporium cucumerinum.  相似文献

Nine polymorphic microsatellite markers were developed for the wood-decay basidiomycete Phlebiopsis gigantea, which is used commercially as a biocontrol agent for annosum root disease on conifers. Microsatellite sequences were isolated from repeat-enriched genomic libraries. Primers flanking these sequences were screened on P. gigantea isolates from Europe and North America. The number of alleles per locus ranged from 5 to 15, and gene diversity ranged from 0.72 to 0.90. These markers should be useful for studies of P. gigantea natural population structure and for making predictions about the impact of P. gigantea application in conifer forests.  相似文献

Summary Resinicium bicolor (Alb. & Schw. ex Fr.) Parm. [=Odontia bicolor (Alb. & Schw. ex Fr.) Bres.] is an outcrossing resupinate basidiomycete associated with root and butt rots of trees, but is itself only very weakly pathogenic. The distribution of genets among every spruce stump in a 70-year-old 1250 m2 spruce stand was analysed using somatic incompatibility testing. R. bicolor was present on 40% of 8-to 10-year-old stumps. Nineteen genets were found occupying 32 stumps; yielding probabilities of colonisation following establishment by basidiospores of 0.20–0.24 and by mycelial extension or dispersal of 0.16–0.20. The probability of colonisation decreased with increasing distance from a point of establishment. R. bicolor responded to both enrichment and destructive disturbances by the formation of an extensive cord system which enabled it to colonise discontinuously distributed resources and to overgrow fungi adjacent to it in a single resource unit, including Heterobasidion annosum.  相似文献

Picea abies logs were inoculated with Resinicium bicolor, Fomitopsis pinicola or left un-inoculated and placed in an old-growth boreal forest. Mass loss and fungal community data were collected after 6 yr to test whether simplification of the fungal community via inoculation affects mass loss and fungal community development. Three techniques were used to survey communities: (1) observation of fruiting structures; (2) culturing on media; and (3) cloning and sequencing of ITS rDNA. Fruit body surveys detected the smallest number of species (18, 3.8 per log), DNA-based methods detected the most species (72, 31.7 per log), and culturing detected an intermediate number (23, 7.2 per log). Initial colonizer affected community development and inoculation with F. pinicola led to significantly greater mass loss. Relationships among fungal community composition, community richness and mass loss are complex and further work is needed to determine whether simplification of fungal communities affects carbon sequestration in forests.  相似文献

Mutualism between microbes and insects is common and alignment of the reproductive interests of microbial symbionts with this lifestyle typically involves clonal reproduction and vertical transmission by insect partners. Here the Amylostereum fungus–Sirex woodwasp mutualism was used to consider whether their prolonged association and predominance of asexuality have affected the mating system of the fungal partner. Nucleotide information for the pheromone receptor gene rab1, as well as the translation elongation factor 1α gene and ribosomal RNA internal transcribed spacer region were utilized. The identification of rab1 alleles in Amylostereum chailletii and Amylostereum areolatum populations revealed that this gene is more polymorphic than the other two regions, although the diversity of all three regions was lower than what has been observed in free-living Agaricomycetes. Our data suggest that suppressed recombination might be implicated in the diversification of rab1, while no evidence of balancing selection was detected. We also detected positive selection at only two codons, suggesting that purifying selection is important for the evolution of rab1. The symbiotic relationship with their insect partners has therefore influenced the diversity of this gene and influenced the manner in which selection drives and maintains this diversity in A. areolatum and A. chailletii.  相似文献

Ash dieback is a recent widespread disease on ash (Fraxinus sp.) that is causing important economic and ecological losses throughout Europe. The disease is initiated by the ascomycetous fungus Hymenoscyphus pseudoalbidus (anamorph Chalara fraxinea). The main aim of this study was to investigate seasonal pattern of lesion development associated with ash dieback. We present data on the spread of 324 natural lesions in ash shoots, branches and stems surveyed over a 32 month period. Most lesions were active and showed the greatest rate of growth during the summer; however, lesions were active throughout the year. Tree mortality was high, with more than a third of the surveyed trees dying during the study. Although many lesions permanently ceased to develop, the rate at which new lesions emerged was greater than the rate at which lesions entered a resting phase. The most common cause for a lesion going into a permanent state of rest was that it had encountered a branch-base. Genotype analysis showed that multiple infections can occur in a single tree given that different genotypes were identified in different lesions as well as in single lesions. A weak positive correlation was noted between tree health and tree size and a weak negative correlation was noted between tree overall health and lesion activity. The lower limit for H. pseudoalbidus growth in culture was between 4.0°C and 0.5°C.  相似文献

The genetic structure of the wood-decay fungus Fomitopsis rosea (Alb. et Schw. Fr.) Karst is presented for populations sampled in Russia, Sweden and Finland. A total of 11 variable arbitrary primed (AP)–PCR markers were found to be segregated in a 1:1 ratio. The genotype for each fruiting body was inferred from the genotypes of the haploid single-spore isolates. Observed heterozygosity was lowest in southern Fennoscandia, the area with the longest history of forestry management. Overall, there was a deficit of heterozygotes compared with frequencies of heterozygotes predicted from Hardy–Weinberg expectations. When we partitioned the deficit of heterozygotes into the hierarchical genetic structure by means of F -statistics we found that the possible causes for it were both limited gene flow between regions and local nonrandom mating within all populations. Future management for the conservation of F. rosea populations and how the results relate to the life cycle of basidomycete fungi are discussed.  相似文献

Understanding the evolutionary histories of invasive species is critical to adopt appropriate management strategies, but this process can be exceedingly complex to unravel. As illustrated in this study of the worldwide invasion of the woodwasp Sirex noctilio, population genetic analyses using coalescent‐based scenario testing together with Bayesian clustering and historical records provide opportunities to address this problem. The pest spread from its native Eurasian range to the Southern Hemisphere in the 1900s and recently to Northern America, where it poses economic and potentially ecological threats to planted and native Pinus spp. To investigate the origins and pathways of invasion, samples from five continents were analysed using microsatellite and sequence data. The results of clustering analysis and scenario testing suggest that the invasion history is much more complex than previously believed, with most of the populations being admixtures resulting from independent introductions from Europe and subsequent spread among the invaded areas. Clustering analyses revealed two major source gene pools, one of which the scenario testing suggests is an as yet unsampled source. Results also shed light on the microevolutionary processes occurring during introductions, and showed that only few specimens gave rise to some of the populations. Analyses of microsatellites using clustering and scenario testing considered against historical data drastically altered our understanding of the invasion history of S. noctilio and will have important implications for the strategies employed to fight its spread. This study illustrates the value of combining clustering and ABC methods in a comprehensive framework to dissect the complex patterns of spread of global invaders.  相似文献
