Next‐generation sequencing data can be mined for highly informative single nucleotide polymorphisms (SNPs) to develop high‐throughput genomic assays for nonmodel organisms. However, choosing a set of SNPs to address a variety of objectives can be difficult because SNPs are often not equally informative. We developed an optimal combination of 96 high‐throughput SNP assays from a total of 4439 SNPs identified in a previous study of Pacific lamprey (Entosphenus tridentatus) and used them to address four disparate objectives: parentage analysis, species identification and characterization of neutral and adaptive variation. Nine of these SNPs are FST outliers, and five of these outliers are localized within genes and significantly associated with geography, run‐timing and dwarf life history. Two of the 96 SNPs were diagnostic for two other lamprey species that were morphologically indistinguishable at early larval stages and were sympatric in the Pacific Northwest. The majority (85) of SNPs in the panel were highly informative for parentage analysis, that is, putatively neutral with high minor allele frequency across the species’ range. Results from three case studies are presented to demonstrate the broad utility of this panel of SNP markers in this species. As Pacific lamprey populations are undergoing rapid decline, these SNPs provide an important resource to address critical uncertainties associated with the conservation and recovery of this imperiled species.  相似文献

There has been much investigation into the use of music as an ergogenic aid to facilitate physical performance. However, previous studies have primarily focused on predetermined music and aerobic exercise. The purpose of this study was to investigate the effects of self-selected music (SSM) vs. those of no music (NM) on the mood and performance of the athletes performing bench press and squat jump. Twenty resistance trained collegiate men completed 2 experimental conditions, one while listening to SSM and the other with NM. The subjects reported their profile of mood states (POMS) and rating of perceived exertion (RPE) before and after performing 3 sets to failure of the bench press at 75% 1 repetition maximum (1RM) and 3 reps of the squat jump at 30% 1RM. Statistical analyses revealed no differences in squat jump height or relative ground reaction force, but the takeoff velocity (SSM-2.06 ± 0.17 m·s(-1); NM-1.99 ± 0.18 m·s(-1)), rate of velocity development (SSM-5.92 ± 1.46 m·s(-2); NM-5.63 ± 1.70 m·s(-2)), and rate of force development (SSM-3175.61 ± 1792.37 N·s(-1); NM-2519.12 ± 1470.32 N·s(-1)) were greater with SSM, whereas RPE (SSM-5.71 ± 1.37; NM-6.36 ± 1.61) was greater with NM. Bench press reps to failure and RPE were not different between conditions. The POMS scores of vigor (SSM-20.15 ± 5.58; NM-17.45 ± 5.84), tension (SSM-8.40 ± 3.99; NM-6.07 ± 3.26), and fatigue (SSM-8.65 ± 4.49; NM-7.40 ± 4.38) were greater with SSM. This study demonstrated increased performance during an explosive exercise and an altered mood state when listening to SSM. Therefore, listening to SSM might be beneficial for acute power performance.  相似文献

The relationship between isometric force-time curve characteristics and club head speed in recreational golfers. J Strength Cond Res 26(10): 2685-2697, 2012-The primary purpose of the present investigation was to examine the relationships between club head speed, isometric midthigh pull performance, and vertical jump performance in a cohort of recreational golfers. Twelve recreational golfers (age, 20.4 ± 1.0 years; weight, 77.0 ± 9.8 kg; height, 177.8 ± 6.3 cm; body fat, 17.1 ± 7.6%; handicap, 14.5 ± 7.3; experience, 8.9 ± 3.6 years) completed 3 testing sessions: (a) familiarization session and body composition measurements; (b) measurement of force-time curves in the isometric midthigh pull, countermovement, and static vertical jump (SJ); and (c) measurement of club head speed. During sessions 1 and 2, subjects performed 5 countermovement jumps, 5 SJ, and 2 isometric midthigh pulls. Isometric peak force was measured at 30, 50, 90, 100, 200, and 250 milliseconds. Rate of force development was measured among 0-30, 0-50, 0-90, 0-100, 0-200, and 0-250 milliseconds. Peak rate of force development was determined as the highest value in a 10-millisecond sampling windows. During session 3, subjects performed 10 maximal golf swings with a driver to measure club head speed; peak and average club head speed were analyzed across the 10 swings. Golf handicap was moderately correlated with average (r = -0.52, p = 0.04) and maximal club head speed (r = -0.45, p = 0.07). Force at 150 milliseconds during the isomeric midthigh pull test was moderately correlated with average (r = 0.46, p = 0.07) and maximal club head speed (r = 0.47, p = 0.06). Moderate correlations were also found between the rate of force development from 0 to 150 milliseconds and average (r = 0.38, p = 0.11) and maximal club head speed (r = 0.36, p = 0.12). The present findings suggest that the ability to exhibit high ground reaction forces in time frames <200 milliseconds are related to high club head speeds.  相似文献

Abstract Races 1 and 400 of Melampsosau bni the causal agent of flax [Lmum usitatissimum]), were crossed and the resulting F2 progeny evaluated on near-isogcnic fiax lines. The infection type of the parental cultures, the F12 and the segregating f2 progeny were used to evaluate the inheritance of pathogenicity. Races 1 and 400 as well as the F2 and all the F2 progeny were avirulent on lines N and P4 genes. This indicated both races to be homozygous avirulent. The F2 and all the F2 progeny were virulent on lines containing L9, M1, M4, and P. This indicated the F to be homuzygous for virulence at these loci. Segregation ratios of the F2 cultures fit monogenic ratios on lines with host genes K. L2, L5, L6, L7, L11, M, M2, M3, N1, P1, P2, and P3. Race 1 was avirulent, race 400 virulent and tht F, avirulent on these lines. Several linkage groups were indicated including close linkage at Ap1, Ap2 and Ap3. Segregation on lines L, L3, M5, M6 and cultivars Flor, Dufferin, Linott, and Wishek fit a digenic model. The infection tvpes of the parents, the F1 and the F2, segregations could usually be explained by classical gene-for-gene interactions. Possible exceptions were segregations on L4, L8 and N2 which did not fit a 3: 1 ratio but did fit a 9: 7 ratio. Segregation on L 10 fit a 7: 9 ratio but line L10 may be temperature sensitive. Segregation on L1 indicated a single dominant gene for pathogenicity.  相似文献

High power, nanosecond pulsed electric field (nsPEF) effects have been focused on bacterial decontamination, but the impact on mammalian cells is now being revealed. During nsPEF applications, electrical pulses of 10, 60 or 300 ns durations were applied to cells using electric field amplitudes as high as 300 kV/cm. Because of the ultra-short pulse durations, the energy transferred to cells is negligible, and only non-thermal effects are observed. We investigated the genotoxicity of nsPEF on adherent and non-adherent cell lines including 10 human lines and one mouse cell line with different origin and growth characteristics. We present data examining the effects of nsPEF exposure on cell survival assessed by clonogenic formation or live cell count; DNA damage determined by the comet assay and chromosome aberrations; and cell cycle parameters by measuring the mitotic indices of exposed cells. Using each of these indicators, we observed differential effects among cell types with non-adherent cells being more sensitive to the genotoxic effects of nsPEF exposures than adherent cells. Non-adherent cultures showed a rapid decrease in cell viability (90%), induction of DNA damage, and a decrease in the number of cells reaching mitosis after one 60 ns pulse with an electric field intensity of 60 kV/cm. These effects were not observed in cells grown as adherent cultures, with the exception of the mouse 3T3 cell line, which showed survival characteristics similar to non-adherent cultures. These data suggest that nsPEF genotoxicity may be cell type specific, and therefore have potential applications in the selective removal of one cell type from another, for example, in diseased states.  相似文献
