Effect of alkylating agent dipin was studied on hematopoietic (CFU-S) and stromal (CFU-F) progenitor cells. Single administration of dipin (0.06 mg/g) to adult (CBA × C57Bl/6) F1 hybrid mice induced a long-term (2 years) oscillations in the numbers of day 7 CFU-S and day 11 CFU-S in the bone marrow and spleen. Dipin also damaged the hematopoietic stroma as indicated by decreased numbers of CFU-F which remained low for at least a year. The capacity of stromal cells to form ectopic hematopoietic foci was considerably decreased and also remained low for 10 months. The obtained data suggest high dipin sensitivity of the earliest hematopoietic and stromal cells. Antifouling paint booster biocides are a group of organic compounds added to antifouling paints to improve their efficacy. They have become prevalent since the requirement for alternative antifouling paints formulations for small boats (< 25 m). This need followed a ban on the use of triorganotin biocides in antifouling paints for small boats, in the late 1980's. Worldwide, around eighteen compounds are currently used as antifouling biocides, viz. benzmethylamide, chlorothalonil, copper pyrithione, dichlofluanid, diuron, fluorofolpet, Irgarol 1051, Sea‐Nine 211, Mancozeb, Polyphase, pyridine‐triphenyl‐borane, TCMS (2,3,5,6‐tetrachloro‐4‐methylsulfonyl) pyridine, TCMTB [2‐(thiocyanomethylthio)benzothia‐zole], Thiram, tolyfluanid, zinc pyrithione (ZPT), ziram and Zineb. Any booster biocide released into the environment is subjected to a complex set of processes. These processes include transport mechanisms, transformation, degradation, cross media partitioning, and bioaccumulation. This paper reviews the fate and behaviour data currently available in the public domain concerning antifouling paint booster biocides.  相似文献

X-ray analysis of enzyme–DNA interactions is very informative in revealing molecular contacts, but provides neither quantitative estimates of the relative importance of these contacts nor information on the relative contributions of specific and nonspecific interactions to the total affinity of enzymes for specific DNA. A stepwise increase in the ligand complexity approach is used to estimate the relative contributions of virtually every nucleotide unit of synthetic DNA containing abasic sites to its affinity for apurinic/apyrimidinic endonuclease (APE1) from human placenta. It was found that APE1 interacts with 9–10 nt units or base pairs of single-stranded and double-stranded ribooligonucleotides and deoxyribooligonucleotides of different lengths and sequences, mainly through weak additive contacts with internucleotide phosphate groups. Such nonspecific interactions of APE1 with nearly every nucleotide within its DNA-binding cleft provides up to seven orders of magnitude (ΔG° ~ −8.7 to −9.0 kcal/mol) of the enzyme affinity for any DNA substrate. In contrast, interactions with the abasic site together with other specific APE1–DNA interactions provide only one order of magnitude (ΔG° ~ −1.1 to −1.5 kcal/mol) of the total affinity of APE1 for specific DNA. We conclude that the enzyme's specificity for abasic sites in DNA is mostly due to a great increase (six to seven orders of magnitude) in the reaction rate with specific DNA, with formation of the Michaelis complex contributing to the substrate preference only marginally.  相似文献

Background

The function of proteins is a direct consequence of their three-dimensional structure. The structural classification of proteins describes the ways of folding patterns all proteins could adopt. Although, the protein folds were described in many ways the functional properties of individual folds were not studied.

Results

We have analyzed two β-barrel folds generally adopted by small proteins to be looking similar but have different topology. On the basis of the topology they could be divided into two different folds named SH3-fold and OB-fold. There was no sequence homology between any of the proteins considered. The sequence diversity and loop variability was found to be important for various binding functions.

Conclusions

The function of Oligonucleotide/oligosaccharide-binding (OB) fold proteins was restricted to either DNA/RNA binding or sugar binding whereas the Src homology 3 (SH3) domain like proteins bind to a variety of ligands through loop modulations. A question was raised whether the evolution of these two folds was through DNA shuffling.  相似文献

Results are presented from experimental studies on the unique beam-plasma generator of microwave radiation with a stochastically jumping phase (MWRSJP). To interpret the experimental results, a computer code was developed that allows one to simulate the process of gas ionization by electrons heated in the MWRSJP field and the behavior of plasma particles in such a field. The conditions for ignition and maintenance of a microwave discharge in air by MWRSJP are found both experimentally and theoretically, and the pressure range in which the power required for discharge ignition and maintenance is minimum are determined.  相似文献

A study was made of the interaction of 8-oxoguanine-DNA glycosylases of Escherichia coli (Fpg) and human (OGG1), as well as apurinic/apyrimidinic endonucleases of yeast (Apn1) and E. coli (Nfo), with oligodeoxyribonucleotides containing 8-oxoguaine (oxoG) and tetrahydrofuran (F, a stable analog of an apurinic site) separated by various numbers of nucleotides. Inhibitor analysis showed that the affinity of Fgp for single-stranded DNA ligands is virtually independent of the relative positions of oxoG and F. K M and k cat were determined for all the four enzymes and all double-stranded substrates studied. The effect of the second lesion strongly depended both on the relative position of the lesion and the enzyme of interest. The highest drop in the affinity of Fpg and OGG1 for the substrate (1.6-to 148-fold) and in the reaction rate (4.8-to 58-fold) was recorded for the oligonucleotides in which F was immediately 3′ or 5′ of oxoG. Introduction of the second lesion barely affected K M for nucleases Apn1 and Nfo. The reaction rate was five-to tenfold lower for the substrates containing two adjacent lesions. For all enzymes studied, an increase in the distance between two lesions in double-stranded DNA decreased their contribution to K M and k cat.  相似文献

The molecular basis of development and regeneration of skeletal muscles are reviewed. A model of parent-progeny relationships of mature animals?? skeletal muscles is proposed. Different cellular populations that contribute to myogenesis in vivo and in vitro are described. Both well-known typical cellular sources for muscle regeneration (satellite cells, muscle derived stem cells) and alternative cellular sources (CD133+ cells, pericytes, SP-cells from muscles and from bone marrow, mesangioblasts, embryonic stem cells and mesenchymal stromal cells) are presented. Moreover, some evidence for the existence of ectopic myogenic precursors in nonmuscle tissues is given.  相似文献

A one-dimensional model of an RF discharge in CO-containing gas mixtures is developed. The model takes into account the effect of the degree of vibrational excitation of CO molecules on the structure of the discharge and on its parameters. Experimental data are presented from measurements of the voltage-power characteristics of RF discharges in gas mixtures with different CO contents in the pressure range of 10–100 torr. The model developed is used to calculate the dependence of the root-mean-square discharge voltage on the specific power deposition in an RF discharge under our experimental conditions. The experimental data are compared to the results of numerical simulations. For working gas pressures of about 100 torr, which are typical of the operation of slab CO lasers, the calculated voltage-power characteristics of an RF discharge agree satisfactorily with those obtained experimentally. The theoretical model predicts that the vibrational excitation of CO molecules leads to a redistribution of the RF field in the discharge gap and to an increase in the laser efficiency.  相似文献

Quantum mechanics predicts the existence of power-law tails in the momentum distribution function of particles in dense media even under conditions of thermodynamic equilibrium. The generalized expressions allowing for the effect of the medium density show that quantum corrections lead to a sharp increase in the reaction rates of threshold exothermic processes (such as fusion and chemical reactions and vibrational-translational relaxation). The accompanying modification of the distribution function changes the wings of the emission and absorption lines. The profiles of the absorption lines in dense gaseous media are shown to be asymmetric with respect to the line center.  相似文献

New types of beam-plasma devices generating intense stochastic microwave radiation in the interaction of electron beams with hybrid plasma waveguides were developed and put into operation at the National Science Center Kharkov Institute of Physics and Technology (Ukraine). The objective of the paper is to discuss the results of theoretical and experimental studies and numerical simulations of the normal and oblique incidence of linearly polarized electromagnetic waves on an interface between a vacuum and an overcritical plasma. The main results of the reported investigations are as follows: (i) for the parameter values under analysis, the transmission coefficient for microwaves with a stochastically jumping phase is one order of magnitude greater than that for a broadband regular electromagnetic wave with the same spectral density; (ii) the electrons are heated most efficiently by obliquely incident waves with a stochastically jumping phase and, in addition, the electron distribution function has a high-energy tail; and (iii) necessary conditions for gas breakdown and for the initiation of a microwave discharge in stochastic fields in a light source are determined. The anomalously large transmission coefficient for microwaves, the anomalous character of the breakdown conditions, the anomalous behavior of microwave gas discharges, and the anomalous nature of collisionless electron heating, are attributed to stochastic jumps in the phase of microwave radiation.  相似文献
