Thirty-six percent of the wild potato (Solanum L. section Petota Dumort.) species are polyploid, and about half of the polyploids are tetraploid species (2n = 4x = 48). Determination of the type of polyploidy and development of the genome concept for members of section Petota traditionally has been based on the analysis of chromosome pairing in species and their hybrids and, most recently, DNA sequence phylogenetics. Based on these data, the genome designation AABB was proposed for Mexican tetraploid species of series Longipedicellata Buk. We investigated this hypothesis with genomic in situ hybridization (GISH) for both representatives of the series, S. stoloniferum Schltdl. and S. hjertingii Hawkes. GISH analysis supports an AABB genome constitution for these species, with S. verrucosum Schltdl. (or its progenitor) supported as the A genome donor and another North or Central American diploid species (S. cardiophyllum Lindl., S. ehrenbergii (Bitter) Rydb., or S. jamesii Torrey) as the B genome donor. GISH analysis of chromosome pairing of S. stoloniferum also confirms the strict allopolyploid nature of this species. In addition, fluorescence in situ hybridization data suggest that 45S rDNA regions of the two genomes of S. stoloniferum were changed during coevolution of A and B genomes of this allotetraploid species.  相似文献

Alzheimer's disease is the most prevalent form of dementia worldwide. Therapies are desperately needed to prevent and cure the disease. Mouse models of amyloid- deposition [APP and PSAPP transgenic (tg) mice] have been useful in determining the role of amyloid- (A) in both the pathogenesis and cognitive changes in AD. In addition, they have allowed scientists to investigate potential AD therapies in living animals. Active and passive A immunizations have been employed successfully in APP and PSAPP tg mice to lower cerebral A levels and improve cognition. Optimization of immunization protocols and characterization of immune responses in wildtype mice have been reported. Based on the promising results of A immunization studies in mice, a clinical trial was initiated for A vaccination in humans with AD. Although no adverse effects were reported in the Phase I safety trials, about 5% of AD patients in the phase II clinical trial developed meningoencephalitis, ending the trial prematurely in March 2002. Studies in AD mouse models and wildtype mice may help elucidate the mechanism for these unwanted side effects and will be useful for testing newer, safer vaccines for future use in human clinical trials.  相似文献

Polymerase chain reaction (PCR) primers designed to amplify the octapeptide repeat region of the bovine prion gene were used to test the association of genotypes with bovine spongiform encephalitis (BSE) in 56 BSE-affected and 177 unaffected animals. Three alleles (A, B, C) were detected as single-strand conformation polymorphisms (SSCPs) and two alleles (1,2 representing six or five copies of the octapeptide repeat respectively) were detected as amplified double-strand fragment length polymorphisms (AMFLPs). Observed genotypes of SSCPs and AMFLPs were analysed by x-square. The SSCP genotypes of nuclear family members of animals with BSE and BSE-affected animals were different (P < 0.001, P < 0.01) from unrelated animals of the same breed without BSE. No genotypic differences were found between the BSE-affected animals and their relatives (P > 0.469). No AMFLP genotypic differences were detected between BSE-affected animals, their relatives, unrelated animals of the same breed or animals of different breeds (P > 0.05). These data suggest that BSE-affected animals and their relatives are more likely to have the AA SSCP genotype than unrelated animals of the same breed or animals of different breeds.  相似文献

中间锦鸡儿(Caragana liouana)是中国毛乌素沙地的主要灌木建群种,在其主要分布区采集9个不同地理种源的种子,栽种至同质园,并测定不同器官(根、茎、叶)碳(C)、氮(N)、磷(P)含量,比较种源和器官间碳氮磷化学计量特征的差异及元素之间的相关性。结果显示:(1)不同种源中间锦鸡儿根、茎、叶的C含量差异显著,分别为361.12~426.30mg·g~(-1)、412.32~463.13mg·g~(-1)、419.21~478.94mg·g~(-1);N含量种源间差异显著,分别为20.52~33.67mg·g~(-1)、15.77~23.92mg·g~(-1)、27.60~36.44mg·g~(-1);P含量种源间差异显著,分别为1.52~3.73mg·g~(-1)、1.24~2.14mg·g~(-1)、1.44~2.38mg·g~(-1);不同器官的C/N、C/P、N/P也表现出种源间显著差异。(2)种源和器官对中间锦鸡儿碳氮磷化学计量特征的影响程度存在差异,种源对P、C/P、N/P影响较大,器官对C、N、C/N影响较大。(3)相关性分析表明,N、P分别对C/N和C/P的变异起主导作用,并共同影响N/P的变异。研究表明,中间锦鸡儿的碳氮磷化学计量特征在长期的适应进化过程中已产生遗传分化,并形成了自身的养分利用策略。  相似文献

从茎花葱臭木种子中分离得到5个化合物,经理化与波谱分析鉴定为β-谷甾醇(1)、没食子酸乙酯(2)、胡萝卜苷(3)、1-O-β-D-吡喃葡萄糖基-(2S,3S,4R,8Z)-2-N-(2 ′-羟基二十四烷酰氨基)十八二氧鞘氨-8-烯(4)和2,3,2″,3″-四氢穗花杉双黄酮(5).这5个化合物均首次从该植物中分离得到.其中化合物5进行细胞毒活性测试,没有显示抑制活性.  相似文献
