Background

Ad35.CS.01 is a pre-erythrocytic malaria candidate vaccine. It is a codon optimized nucleotide sequence representing the P. falciparum circumsporozoite (CS) surface antigen inserted in a replication deficient Adenovirus 35 backbone. A Phase 1a trial has been conducted in the USA in naïve adults and showed that the vaccine was safe. The aim of this study is to assess the safety and immunogenicity of ascending dosages in sub Saharan Africa.

Methods

A double blind, randomized, controlled, dose escalation, phase Ib trial was conducted in a rural area of Balonghin, the Saponé health district (Burkina Faso). Forty-eight healthy adults aged 18-45 years were randomized into 4 cohorts of 12 to receive three vaccine doses (day 0, 28 and 84) of 109, 1010, 5X1010, 1011 vp of Ad35.CS.01 or normal saline by intra muscular injection. Subjects were monitored carefully during the 14 days following each vaccination for non serious adverse events. Severe and serious adverse events were collected throughout the participant study duration (12 months from the first vaccination). Humoral and cellular immune responses were measured on study days 0, 28, 56, 84, 112 and 140.

Results

Of the forty-eight subjects enrolled, forty-four (91.7%) received all three scheduled vaccine doses. Local reactions, all of mild severity, occurred in thirteen (27.1%) subjects. Severe (grade 3) laboratory abnormalities occurred in five (10.4%) subjects. One serious adverse event was reported and attributed to infection judged unrelated to vaccine. The vaccine induced both antibody titers and CD8 T cells producing IFNγ and TNFα with specificity to CS while eliciting modest neutralizing antibody responses against Ad35.

Conclusion

Study vaccine Ad35.CS.01 at four different dose levels was well-tolerated and modestly immunogenic in this population. These results suggest that Ad35.CS.01 should be further investigated for preliminary efficacy in human challenge models and as part of heterologous prime-boost vaccination strategies.

The hypothesis of the present study was that bacterial communities would differentiate under Eucalyptus camaldulensis and that an enhancement of arbuscular mycorrhizal (AM) density would minimize this exotic plant species effect. Treatments consisted of control plants, preplanting fertilizer application and AM inoculation. After 4 months of culture in autoclaved soil, E. camaldulensis seedlings were either harvested for growth measurement or transferred into containers filled with the same soil but not sterilized. Other containers were kept without E. camaldulensis seedlings. After 12 months, effects of fertilizer amendment and AM inoculation were measured on the growth of Eucalyptus seedlings and on soil microbial communities. The results clearly show that this plant species significantly modified the soil bacterial community. Both community structure (assessed by denaturing gradient gel electrophoresis profiles) and function (assessed by substrate-induced respiration responses including soil catabolic evenness) were significantly affected. Such changes in the bacterial structure and function were accompanied by disturbances in the composition of the herbaceous plant species layer. These results highlight the role of AM symbiosis in the processes involved in soil bio-functioning and plant coexistence and in afforestation programmes with exotic tree species that target preservation of native plant diversity.  相似文献

ABSTRACT: BACKGROUND: Genetic factors play a key role in determining resistance/susceptibility to infectious disease. Susceptibility of the human host to malaria infection has been reported to be influenced by genetic factors, which could be confounders if not taken into account in the assessment of the efficacy of interventions against malaria. This study aimed to assess the relationship between haemoglobin genotypes and malaria in children under five years in a site being characterized for future malaria vaccine trials. METHODS: The study population consisted of 452 children living in four rural villages. Hb genotype was determined at enrolment. Clinical malaria incidence was evaluated over a one-year period using combined active and passive surveillance. Prevalence of infection was evaluated via bi-annual cross-sectional surveys. At each follow-up visit, children received a brief clinical examination and thick and thin blood films were prepared for malaria diagnosis. A clinical malaria was defined as Plasmodium falciparum parasitaemia >2,500 parasites/ul and axillary temperature [greater than or equal to]37.5degreesC or reported fever over the previous 24 hours. RESULTS: Frequencies of Hb genotypes were 73.2% AA; 15.0% AC; 8.2% AS; 2.2% CC; 1.1% CS and 0.2% SS. Prevalence of infection at enrolment ranged from 61.9%-54.1% among AA, AC and AS children. After one year follow-up, clinical malaria incidence (95% CI) (episodes per person-year) was 1.9 (1.7-2.0) in AA, 1.6 (1.4-2.1) in AC, and 1.7 (1.4-2.0) in AS children. AC genotype was associated with lower incidence of clinical malaria relative to AA genotype among children aged 1-2 years [rate ratio (95% CI) 0.66 (0.42-1.05)] and 2-3 years [rate ratio (95% CI) 0.37 (0.18-0.75)]; an association of opposite direction was however apparent among children aged 3-4 years. AS genotype was associated with lower incidence of clinical malaria relative to AA genotype among children aged 2-3 years [rate ratio (95% CI) 0.63 (0.40-1.01)]. CONCLUSIONS: In this cohort of children, AC or AS genotype was associated with lower risk of clinical malaria relative to AA genotype only among children aged one to three years. It would be advisable for clinical studies of malaria in endemic regions to consider haemoglobin gene differences as a potentially important confounder, particularly among younger children.  相似文献

During development, the risk of developing mesial temporal lobe epilepsy (MTLE) increases when the developing brain is exposed to more than one insult in early life. Early life insults include abnormalities of cortical development, hypoxic-ischemic injury and prolonged febrile seizures. To study epileptogenesis, we have developed a two-hit model of MTLE characterized by two early-life insults: a freeze lesion-induced cortical malformation at post-natal day 1 (P1), and a prolonged hyperthermic seizure (HS) at P10. As early life stressors lead to sexual dimorphism in both acute response and long-term outcome, we hypothesized that our model could lead to gender-based differences in acute stress response and long-term risk of developing MTLE. Male and female pups underwent a freeze-lesion induced cortical microgyrus at P1 and were exposed to HS at P10. Animals were monitored by video-EEG from P90 to P120. Pre and post-procedure plasma corticosterone levels were used to measure stress response at P1 and P10. To confirm the role of sex steroids, androgenized female pups received daily testosterone injections to the mother pre-natally and post-natally for nine days while undergoing both insults. We demonstrated that after both insults females did not develop MTLE while all males did. This correlated with a rise in corticosterone levels at P1 following the lesion in males only. Interestingly, all androgenized females showed a similar rise in corticosterone at P1, and also developed MTLE. Moreover, we found that the cortical lesion significantly decreased the latency to generalized convulsion during hyperthermia at P10 in both genders. The cortical dysplasia volumes at adulthood were also similar between male and female individuals. Our data demonstrate sexual dimorphism in long-term vulnerability to develop epilepsy in the lesion + hyperthermia animal model of MTLE and suggest that the response to early-life stress at P1 contributes significantly to epileptogenesis in a gender-specific manner.  相似文献

Cowpea is one of the most important food legume crops in Burkina Faso. It is able to associate with arbuscular mycorrhizal fungi (AMF) and rhizobia. This dual symbiosis improves nitrogen and phosphorus nutrient uptake in cowpea. As the application of exotic inoculants frequently lacks positive responses in field experiments, this study set out to select well-adapted native symbiotic rhizobial and AMF strains. Soil samples were collected from six study sites in three different climatic zones of Burkina Faso to investigate their native symbiotic strains. Soil-extraction of native spores led to the identification of four AMF genera (Scutellospora, Gigaspora, Glomus and Entrophospora) by morpho-anatomical characterization. The two most effective cowpea fungal strains were selected after spore isolation from field-collected soils, multiplication on maize roots and inoculation on cowpea seedlings in a greenhouse experiment. Cowpea-nodulating rhizobial strains were trapped in the greenhouse by planting cowpea seeds in collected soil samples and the strains were characterized using molecular methods. This characterization led to the rhizobial isolates being classified in four clusters on the phylogenetic tree (using the Maximum-Likelihood Phylogenies method). All strains belonged to the Bradyrhizobium genus and most of them were included in the B. japonicum branch. Some groups were clearly distinct species already identified and may be new species. The two most effective strains for cowpea yield improvement in the field were selected after cowpea inoculation in a greenhouse experiment. The inoculation design in the field experiment consisted of four single inoculation treatments, either rhizobial or mycorrhizal, along with four dual inoculations, one treatment with chemical fertilizers, and one uninoculated control. The results showed that cowpea productivity was significatively improved by dual inoculation with native rhizobial and mycorrhizal strains, reaching the same level as the application of commonly used chemical fertilizers [Nitrogen, Phosphorus and Potassium fertilizers (NPK)]. In addition, dual inoculation resulted in the highest iron content in cowpea leaves.  相似文献

Historically, the Sahel of West Africa has been considered synonymous with desertification. In recent decades, however, satellite images reveal patterns of enhanced vegetation termed the “greening of the Sahel.” This greening is well-documented but its mechanisms remain poorly understood. The Sahel is also a region emerging from a 30 year period of reduced rainfall in which several severe droughts occurred. As a response to droughts and land degradation, farmers have rehabilitated thousands of hectares of degraded soils by constructing low barriers of rock through widespread soil and water conservation (SWC) development projects. Remote sensing analyses suggest that these extensive soil conservation projects may explain greening in northern Burkina Faso. This study combines ethnographic fieldwork with the analysis of Geographic Information System (GIS) and remote sensing (RS) data to test whether SWC investments contribute to greening. Ethnographic data reveal a tension between the perceptions of rural producers who feel that their SWC efforts contribute to greening and those of state officials who contend that SWC has only local impacts and that the regional landscape continues to degrade. Our analysis of GIS and RS data suggest that both perspectives are valid but contingent on the particular spatial and temporal scale used for analysis.  相似文献

IntroductionThe first and only Anger camera of Burkina Faso has been installed at the Yalgado Ouedraogo University hospital (CHU YO) in November 2011. The first clinical applications began in January 2012, 20 years after the training of the first nuclear physician of Burkina Faso. The department has opened with the cooperation of the International agency of atomic energy (IAEA) in the framework of a structured project. The objective of this study was to analyze the distribution of scintigraphic examinations performed in the nuclear medicine department, CHU YO, according to medical indications, during the first 6 months.MethodsThe examinations were performed from January 09, 2012 to 15 July 2012. All patients who underwent a scan in the nuclear medicine department were included in the study. The imaging device used was a Mediso? dual head camera installed in 2011. Tc 99m generator was delivered once per month by IBA? France. Cold kits had been delivering at the opening of the nuclear medicine department.ResultsSlightly more than half of the examinations have been performed in men (53.1%). June was the month that recorded the greatest number of examinations carried out (25.7%). Bone and thyroid scans represented 68.1% of nuclear medicine diagnostic procedures. Yalgado Ouedraogo University hospital represented more than half of the departments of origin of prescribers (60.2%). Most scans performed in the department of nuclear medicine were prescribed by internal medicine physicians (22.1%). Work-up for bone metastases in breast cancer was the first indication of bone scans (22.7%). Multinodular goiter (36.4%) and solitary nodules (36.4%) were the first indications of thyroid scintigraphy. Searching for stress-induced myocardial ischemia has been the reason for myocardial perfusion scintigraphy in more than half of the cases (53%). The study of separate renal function has been the commonest indication of renal scans (72.73%). Suspicion of pulmonary embolism was the only clinical indication for lung perfusion scans in the nuclear medicine department.ConclusionIn the future, this new department aims to develop by the extension of the supply of diagnostic examinations and the beginning of radionuclide therapy.  相似文献

Maruca vitrata is an economically significant insect pest of cowpea in sub-Saharan Africa. Understanding the seasonal population patterns of M. vitrata is essential for the establishment of effective pest management strategies. M. vitrata larval populations on cultivated cowpea and adult flying activities were monitored in addition to scouting for host plants and parasitoids during 2 consecutive years in 2010 and 2011 in southwestern Burkina Faso. Our data suggest that M. vitrata populations overlapped on cultivated cowpea and alternate host plants during the rainy season. During the cowpea off-season, M. vitrata maintained a permanent population on the wild host plants Mucuna poggei and Daniella oliveri. The parasitoid fauna include three species, Phanerotoma leucobasis Kri., Braunsia kriegeri End. and Bracon sp. Implications of these finding for pest management strategies are discussed.  相似文献

Caryedon serratus is found in different agro-ecological and agro-climatic zones, and its damage to groundnuts can vary up to an 80% quantitative loss of yield. This study seeks to demonstrate the effects of different agro-ecological refer to so as to very quickly process the content zones and the presence or absence of groundnut cultivation on the structure and genetic distribution of C. serratus in West Africa. Portions of the cytochrome b and 28S ribosomal genes of C. serratus were sequenced, using samples from four countries that represent different agro-ecological and agro-climatic sub-regions of West Africa. The results showed 37 haplotypes for the cytochrome b and 7 haplotypes for the 28S ribosomal gene. Although genetic diversity was different between agro-ecological zones tests show no significant differences in structuring according to agro-ecological zone. These tests, as well as the phylogenetic relationships that our results imply, indicate that there is a genetic differentiation between individuals from groundnut culturing areas compared to those from areas where the cultivation of groundnuts is absent or low.  相似文献
